In contrast to NAI, SUR displayed a post-inflammatory phenotype in the intestine and head kidney, characteristic of inflammation resolution (low il1β, high il10 and low hsp90α ex- pression). 1. Abstract: Supplemental breeding is an intensive population management strategy wherein adults are cap- tured from nature and spawned in controlled settings, and the resulting offspring are later released into the wild. To be effective, supplemental breeding programs require crossing strategies that maximize offspring production while maintaining genetic diversity within each supplemental year class. We used computer simula- tions to assess the efficacy of different mating designs to jointly maximize offspring production and maintain high levels of genetic diversity (as measured by the effective population size) under a variety of biological conditions particularly relevant to species with high fecundity and external fertilization, such as many fishes. We investigated four basic supplemental breeding designs involving either monogamous pairings or complete factorial designs (in which every female is mated to every male and vice versa), each with or without the added stipulation that all breeders contribute equally to the total reproductive output. In general, complete factorial designs that did not equalize parental contributions came closest to the goal of maximizing offspring production while still maintaining relatively large effective population sizes. Next, we estimated the effective population size of 10 different supplemental year classes within the breeding program of the robust redhorse (Moxostoma robustum). -

Halanych 1* Ctenophora, comprising approximately 200 described species, is an important lineage for understanding metazoan evolution and is of great ecological and economic importance. Ctenophore diversity includes species with unique colloblasts used for prey capture, smooth and striated muscles, benthic and pelagic lifestyles, and locomotion with ciliated paddles or muscular propul- sion. However, the ancestral states of traits are debated and relationships among many lineages are unresolved. Here, using 27 newly sequenced ctenophore transcriptomes, publicly available data and methods to control systematic error, we establish the placement of Ctenophora as the sister group to all other animals and refine the phylogenetic relationships within ctenophores. Molecular clock analyses suggest modern ctenophore diversity originated approximately 350 million years ago ± 88 million years, conflicting with previous hypotheses, which suggest it originated approximately 65 million years ago. We recover Euplokamis dunlapae—a species with striated muscles—as the sister lineage to other sampled ctenophores. Ancestral state reconstruction shows that the most recent common ancestor of extant ctenophores was pelagic, possessed tentacles, was bio- luminescent and did not have separate sexes. Our results imply at least two transitions from a pelagic to benthic lifestyle within Ctenophora, suggesting that such transitions were more common in animal diversification than previously thought. tenophores, or comb jellies, have successfully colonized from species across most of the known phylogenetic diversity of nearly every marine environment and can be key species in Ctenophora. -

The credibility of rDNA as a suitable marker for Myxozoa is uncertain and needs to be proved. Furthermore, we need to know the history of myxospore evolution to understand the great diversity of modern species. Results: Phylogenetic analysis of elongation factor 2 supports the ribosomal DNA-based reconstruction of myxozoan evolution. We propose that SSU rDNA is a reliable marker for inferring myxozoan relationships, even though SSU rDNA analysis markedly disagrees with the current taxonomy. The analyses of character evolution of 15 morphological and 5 bionomical characters show the evolution of individual characters and uncover the main evolutionary changes in the myxosporean spore morphology and bionomy. Most bionomical and several morphological characters were found to be congruent with the phylogeny. The summary of character analyses leads to the simulation of myxozoan ancestral morphotypes and their evolution to the current species. As such, the ancestor of all myxozoans appears to have infected the renal tubules of freshwater fish, was sphaerosporid in shape, and had a spore with polar capsules that discharged slightly sideways. After the separation of Malacosporea, the spore of the common myxosporean ancestor then changed to the typical sphaerosporid morphotype. -

To overcome problems related to the detection and enumeration of mixotrophic and hetero- trophic cells containing food vacuoles, we combined flow cytometry and an acidotropic blue probe as an alternative method. Flow cytometry allows the analysis of thousands of cells per minute with high sensitivity to the autofluorescence of different groups of cells and to probe fluorescence. The method was first tested in a grazing experiment where the heterotrophic dinoflagellate Oxyrrhis marina fed on Rhodomonas salina. The maximum ingestion rate of O. marina was 1.7 prey ind.–1 h–1, and the fre- quency of cells with R. salina in the food vacuoles increased from 0 to 2.4 ± 0.5 × 103 cells ml–1 within 6 h. The blue probe stained 100% of O. marina cells that had R. salina in the food vacuoles. The acidotropic blue probe was also effective in staining food vacuoles in the mixotrophic dinoflagellate Dinophysis norvegica. We observed that 75% of the D. norvegica population in the aphotic zone pos- sessed food vacuoles. Overall, in cells without food vacuoles, blue fluorescence was as low as in cells that were kept probe free. Blue fluorescence in O. marina cells with food vacuoles was 6-fold higher than in those without food vacuoles (20 ± 4 and 3 ± 0 relative blue fluorescence cell–1, respectively), while in D. -

Avian β-defensins (AvBDs) are the member of antimicrobial peptides, and Toll-like receptors (TLRs) are the primary receptors that recognize pathogen-associated molecular patterns (PAMPs) of microbes. The aim of this study was to characterize the innate immune system with the focus on the expression of AvBDs in the gastrointestinal tract and reproductive organs for the strategy to enhance the disease resistance of chickens. The proventriculus and cecum of broiler chicks expressed TLRs and AvBDs. It is suggested that a variety of PAMPs of microbes are recognized by different TLRs, probably leading to regulate the synthesis of innate immune factors including AvBDs. In laying hens, TLRs and AvBDs were expressed in the theca and granulosa layers of ovarian follicles and in the oviduct. In vivo LPS challenge increased the expression of several AvBDs in the theca tissue. In contrast, in the cultured theca tissue, LPS upregulated the expression of IL1β and IL6, but did not affect the AvBDs expression; whereas IL1β upregulated the expression of the AvBD12 gene and protein. -

Fifteen fish from a non-infected farm were included as a control. Clinical signs appeared only in extensively and severely infected fish. We observed Enteromyxum leei almost exclusively in the intestinal tract, and very rarely in the intrahepatic biliary ducts or gall bladder. We observed heavily infected intestinal villi adjacent to par- asite-free villi. Histological changes indicated a parasite infection gradually extending from villus to villus, originating from an initial limited infected area probably located in the rectum. The parasite forms were exclusively pansporoblasts located along the epithelial basement membrane. Periodic acid-Schiff (PAS)–Alcian blue was the most useful histological stain for identifying the parasite and characterising the degree of intestinal infection. We observed severe enteritis in infected fish, with inflammatory cell infiltration and sclerosis of the lamina propria. -

This myxozoan has low host specificity and is the causative agent of myxozoan emaciation disease, known for heavy mor- talities and significant financial losses within Mediterranean, Red Sea, and Asian aquaculture industries. The disease has rarely been documented within public aquaria and, to our knowledge, has never been confirmed within the USA. This case report describes an outbreak of E. leei in a population of mixed-species east African/Indo-Pacific marine fish undergoing quarantine at a public aquarium within the USA. Four of 16 different species of fish in the population, each of a different taxonomic family, were confirmed infected by the myxozoan through cloacal flush or intestinal wet mount cytology at necropsy. -
