DNA-DNA relative reassociation experiments in which the S1 nuclease method was used showed that T. hyodysenteriae B78T and B204 had 93% sequence homology with each other and approximately 40% sequence homology with T. innocens B256T and 4/71. Both T. hyodysenteriae B7ST and T. innocens B256T exhibited negligible levels of DNA homology (55%)with T. succinifaciens 6091T. The results of comparisons of protein electrophoretic profiles corroborated the DNA-DNA reassociation results. We found high levels of similarity (296%) in electrophoretic profiles among T. hyodysenteriae strains, moderate levels of similarity (43 to 49%) between T. hyodysenteriae and T. innocens, and no detectable similarity between the profiles of either T. -

Fall 08 Molecular Studies of Treponema pallidum Craig Tipple Imperial College London Department of Medicine Section of Infectious Diseases Thesis submitted in fulfillment of the requirements for the degree of Doctor of Philosophy of Imperial College London 2013 1 Abstract Syphilis, caused by Treponema pallidum (T. pallidum), has re-emerged in the UK and globally. There are 11 million new cases annually. The WHO stated the urgent need for single-dose oral treatments for syphilis to replace penicillin injections. Azithromycin showed initial promise, but macrolide resistance-associated mutations are emerging. Response to treatment is monitored by serological assays that can take months to indicate treatment success, thus a new test for identifying treatment failure rapidly in future clinical trials is required. Molecular studies are key in syphilis research, as T. pallidum cannot be sustained in culture. The work presented in this thesis aimed to design and validate both a qPCR and a RT- qPCR to quantify T. pallidum in clinical samples and use these assays to characterise treatment responses to standard therapy by determining the rate of T. pallidum clearance from blood and ulcer exudates. Finally, using samples from three cross-sectional studies, it aimed to establish the prevalence of T. pallidum strains, including those with macrolide resistance in London and Colombo, Sri Lanka. The sensitivity of T. pallidum detection in ulcers was significantly higher than in blood samples, the likely result of higher bacterial loads in ulcers. RNA detection during primary and latent disease was more sensitive than DNA and higher RNA quantities were detected at all stages. Bacteraemic patients most often had secondary disease and HIV-1 infected patients had higher bacterial loads in primary chancres. -

Many of the bacterial species detected in metagenomic analyses of DD lesions are difficult to culture, and their antimicrobial resistance status is largely unknown. Recently, a novel proximity ligation-guided metagenomic approach (Hi-C ProxiMeta) has been used to identify bacterial reservoirs of antimicrobial resistance genes (ARGs) directly from microbial communities, without the need to culture individual bacteria. The objective of this study was to track tetracycline resistance determinants in bacteria involved in DD pathogenesis using Hi-C. Taxonomy of the Lyme Disease Spirochetes

Department of Microbiology, University of Minnesota Medical School, Minneapolis, Minnesota Received January 23, 1984 Morphology, physiology, and DNA nucleotide composition of Lyme disease spirochetes, Borrelia, Treponema, and Leptospira were compared. Morphologically, Lyme disease spirochetes resemble Borrelia. They lack cytoplasmic tubules present in Treponema, and have more than one periplasmic flagellum per cell end and lack the tight coiling which are characteristic of Leptospira. Lyme disease spirochetes are also similar to Borrelia in being microaerophilic, catalase-negative bacteria. They utilize carbohydrates such as glucose as their major carbon and energy sources and produce lactic acid. Long-chain fatty acids are not degraded but are incorporated unaltered into cellular lipids. The diamino amino acid present in the peptidoglycan is ornithine. The mole % guanine plus cytosine values for Lyme disease spirochete DNA were 27.3-30.5 percent. These values are similar to the 28.0-30.5 percent for the Borrelia but differed from the values of 35.3-53 percent for Treponema and Leptospira. DNA reannealing studies demonstrated that Lyme disease spirochetes represent a new species of Borrelia, exhibiting a 31-59 percent DNA homology with the three species of North American borreliae. In addition, these studies showed that the three Lyme disease spirochetes comprise a single species with DNA homologies ranging from 76-100 percent. The three North American borreliae also constitute a single species, displaying DNA homologies of 75-95 per- cent. -

Introduction Intestinal spirochaetes have become recognized over the last 25 years as an important group of enteric pathogens. These bacteria cause disease in a variety of animal species, especially pigs, poultry, dogs and human beings (Hampson and Stanton, 1996). In pigs, the bacteria cause two well-recognized conditions, swine dysentery (SD), and intestinal spirochaetosis (IS) (Taylor et al., 1980; Hampson, 1991). A third condition, referred to here as spirochaetal colitis (SC), is less clearly defined, but is associated with certain weakly beta-haemolytic spirochaetes other than those causing IS. Swine dysentery is one of the most significant production-limiting diseases of pigs and is a common problem throughout the world. The significance of IS and SC in reducing production is less clear; certainly, clinical manifestations of the conditions are much less severe than with SD. The prevalence of the diseases is not known, but the authors' observations suggest that IS occurs commonly in pigs in Australia and North America, whilst cases of IS and SC also have been reported in Europe (Taylor, 1992). General description of spirochaetes Spirochaetes are chemoheterotrophic bacteria, characterized by a unique cellular anatomy and a distinctive morphology. They are spiral-shaped, with their main structural component being a coiled protoplasmic cylinder consisting of cytoplasmic and nuclear regions, surrounded by a cytoplasmic membrane-cell wall complex. Periplasmic flagellae, that are wound around the helical protoplasmic cylinder, run from each end of the cell, and overlap near its middle.
