Prisinzano Date approved: July 18, 2012 ii ABSTRACT Kappa opioid (KOP) receptors have been suggested as an alternative target to the mu opioid (MOP) receptor for the treatment of pain because KOP activation is associated with fewer negative side-effects (respiratory depression, constipation, tolerance, and dependence). The KOP receptor has also been implicated in several abuse-related effects in the central nervous system (CNS). KOP ligands have been investigated as pharmacotherapies for drug abuse; KOP agonists have been shown to modulate dopamine concentrations in the CNS as well as attenuate the self-administration of cocaine in a variety of species, and KOP antagonists have potential in the treatment of relapse. One drawback of current opioid ligand investigation is that many compounds are based on the morphine scaffold and thus have similar properties, both positive and negative, to the parent molecule. Thus there is increasing need to discover new chemical scaffolds with opioid receptor activity. -

Superoxide Generation A number of free radical generating systems were used to investigate the antioxidant properties and structure-activity relationships of a series of monohydroxylated and dihydrox ylated flavones. Ortho-dihydroxylated flavones showed the highest inhibitory activity on en zymic and non-enzymic microsomal lipid peroxidation as well as on peroxyl radical scaveng ing. Most flavones were weak scavengers of hydroxyl radical, while ortho-dihydroxylated flavones interacted with superoxide anion generated by an enzymic system or by human neutrophils. This series of compounds did not exert cytotoxic effects on these cells. Scaveng ing of superoxide and peroxyl radicals may determine the antioxidant properties of these active flavones. In fact, many studies have shown that polyphenol-rich diets have protective effects against most chronic diseases. However, these health benefits are strongly related to both polyphenol content and bioavailability, which in turn depend on their origin, food matrix, processing, digestion, and cellular metabolism. Although most fruits and vegetables are valuable sources of polyphenols, they are not usually con- sumed raw. Instead, they go through some processing steps, either industrially or domestically (e.g., cooling, heating, drying, fermentation, etc.), that affect their content, bioaccessibility, and bioavail- ability. This review summarizes the status of knowledge on the possible (positive or negative) effects of commonly used food-processing techniques on phenolic compound content and bioavailability in fruits and vegetables. These effects depend on the plant type and applied processing parameters (type, duration, media, and intensity). This review attempts to shed light on the importance of more comprehensive dietary guidelines that consider the recommendations of processing parameters to take full advantage of phenolic compounds toward healthier foods. The most abundant phenolics in soybeans are isoflavones, mainly daidzein, 20 glycitein, and genistein and several derivatives of them. Other phenolic compounds are also 21 present but they have been minimally studied. In this work, the soybean seeds were fermented by 22 the microbiota present on the seeds or by inoculation with Aspergillus oryzae, Rhizopus oryzae or 23 Bacillus subtilis . The effect of microbial fermentation on the phenolic composition of soybeans 24 was studied. All the assayed microorganisms brought out variations in phenolics. An increase in 25 the concentration of phenolic acids was observed. Flavonoids were differently metabolized 26 depending on the microorganism used. Interest in phenolic compounds has dramatically increased during the last decade due to their biological effects and promising therapeutic applications. In this review, we discuss the importance of phenolic compounds’ bioavailability to accomplish their physiological functions, and highlight main factors affecting such parameter throughout metabolism of phenolics, from absorption to excretion. Besides, we give an updated overview of the health benefits of phenolic compounds, which are mainly linked to both their direct (e.g., free-radical scavenging ability) and indirect (e.g., by stimulating activity of antioxidant enzymes) antioxidant properties. Such antioxidant actions reportedly help them to prevent chronic and oxidative stress-related disorders such as cancer, cardiovascular and neurodegenerative diseases, among others. Last, we comment on development of cutting-edge delivery systems intended to improve bioavailability and enhance stability of phenolic compounds in the human body. Kalikar Department of Pharmacy,Government Polytechnic, Gadgenagar, VMV Road, Amravati(MS), India 444603 Received on:14-07-2012; Revised on: 19-08-2012; Accepted on:23-09-2012 ABSTRACT Flavonoids belong to a group of polyphenolic compounds, which are classified as flavonols, flavonones, flavones, flavan-3-ols and isoflavones, anthocynidins according to the positions of the substitutes present on the parent molecule. Flavonoids occur as aglycones, glycosides and methylated derivatives in plants. Flavonoids occur naturally in fruit, vegetables, and beverages such as tea and wine and over 4000 structurally unique flavonoids have been identified in plant sources. These are primarily recognized as the pigments responsible for the many shades of yellow, orange, and red in flowers, fruits, and leaves. The major actions of flavonoids are those against cardiovascular diseases, ulcers, viruses, inflammation, osteoporosis, diarrhea and arthritis. Flavonoids have also been known to possess biochemical effects, which inhibit a number of enzymes such as aldosereductase, xanthine oxidase, phosphodiesterase, Ca+2-ATPase, lipoxygenase, cycloxygenase, etc. They also have a regulatory role on different hormones like estrogens, androgens and thyroid hormones. Flavonoids like quercetin are shown to produce anti-inflammatory effect. The flavonoids in tea are found to be responsible for the prevention of osteoporosis. Quercetin is now found to show inhibiting effects against herpes simplex virus. Antidiarrheal effect is found to be shown by the flavonoids present in cocoa. -
