To dissect the genetic basis of these desirable traits, we sequenced and assembled the 408 Mb genome of Corymbia citriodora, anchored into eleven chromosomes. Comparative analysis with Eucalyptus grandis reveals high synteny, although the two diverged approximately 60 million years ago and have different genome sizes (408 vs 641 Mb), with few large intra-chromosomal rearrangements.

BCorresponding author. Email: [email protected] Abstract. Quambalaria spp. cause leaf and shoot dieback diseases on young Eucalyptus trees in Australia, Thailand, South America and South Africa. The disease was first recorded in South Africa in the early 1990s but has been restricted to nurseries in the subtropical north-east coastal area of the country, without resulting in great effect. Recent disease surveys in the Mpumalanga Province of South Africa have revealed extensive shoot and leaf dieback, as well as stem cankers on 1-year-old E. nitens trees. Some symptoms of the disease resembled Quambalaria leaf and shoot blight. However, this was the first time it had occurred on the stems of larger trees, on E. nitens or in the cold temperate region of the country. The aim of this study was to identify the causal agent of the disease and to test different Eucalyptus spp. and clones of relevance to the South African forestry industry for their susceptibility to the pathogen. Comparisons of DNA sequence data for the ITS and 5.8S regions were used to identify the fungus. Results showed that the pathogen represented Q. eucalypti. Inoculation trials showed that all the material tested was susceptible to infection by Q.

The phylogenetic relationship of Quambalaria spp., previously classified in genera such as Sporothrix and Ramularia, has never been addressed. It has, however, been suggested that they belong to the basidiomycete orders Exobasidiales or Ustilaginales. The aim of this study was thus to consider the ordinal relationships of Q. eucalypti and Q. pitereka using ribosomal LSU sequences. Sequence data from the ITS nrDNA were used to determine the phylogenetic relationship of the two Quambalaria species together with Fugomyces (= Cerinosterus) cyanescens. In addition to sequence data, the ultrastructure of the septal pores of the species in question was compared. From the LSU sequence data it was concluded that Quambalaria spp.

The main biological constraint to further expansion of spotted gum plantations is Quambalaria shoot blight caused by the fungus Quambalaria pitereka. Surveys conducted to evaluate the impact of Quambalaria shoot blight have shown that the disease is present in all spotted gum plantations and on a range of Corymbia species and hybrids in subtropical and tropical regions surveyed in eastern Australia. More recently, Q. eucalypti has also been identified from a range of Eucalyptus species in these regions. Both pathogens have also been found associated with foliage blight and die-back of amenity trees and Q. pitereka in native stands of Corymbia species, which is the probable initial infection source for plantations.
