Sargis4 Disrupting Chemicals and Diabetes Risk in Vulnerable Populations https://doi.org/10.2337/dc16-2765 Burgeoning epidemiological, animal, and cellular data link environmental endocrine- disrupting chemicals (EDCs) to metabolic dysfunction. Disproportionate exposure to diabetes-associated EDCs may be an underappreciated contributor to disparities in metabolic disease risk. The burden of diabetes is not uniformly borne by American society; rather, this disease disproportionately affects certain populations, including African Americans, Latinos, and low-income individuals. The purpose of this study was to review the evidence linking unequal exposures to EDCs with racial, ethnic, and socioeconomic diabetes disparities in the U.S.; discuss social forces promoting these disparities; and explore potential interventions. Articles examining the links between chemical exposures and metabolic disease were extracted from the U.S. National REVIEW Library of Medicine for the period of 1966 to 3 December 2016. EDCs associated with diabetes in the literature were then searched for evidence of racial, ethnic, and socioeconomic exposure disparities. Among Latinos, African Americans, and low- income individuals, numerous studies have reported significantly higher exposures to diabetogenic EDCs, including polychlorinated biphenyls, organochlorine pesti- cides, multiple chemical constituents of air pollution,

    PCBs 443 4. CHEMICAL AND PHYSICAL INFORMATION 4.1 CHEMICAL IDENTITY PCBs are a class of chemical compounds in which 2–10 chlorine atoms are attached to the biphenyl molecule. Monochlorinated biphenyls (i.e., one chlorine atom attached to the biphenyl molecule) are often included when describing PCBs. The general chemical structure of chlorinated biphenyls is shown below. 23 23 2' 3' Clx Cly 1 1' 4 4' 5 6 5'6' It can be seen from the structure that a large number of chlorinated compounds are possible. The 209 possible compounds are called congeners. PCBs can also be categorized by degree of chlorination. The term “homolog” is used to refer to all PCBs with the same number of chlorines (e.g., trichloro­ biphenyls). Homologs with different substitution patterns are referred to as isomers. For example, the dichlorophenyl homolog contains 12 isomers. The numbering system for the PCBs is also shown above. Positions 2, 2', 6, and 6' are called ortho positions, positions 3, 3', 5, and 5' are called meta positions, and positions 4 and 4' are called para positions. The benzene rings can rotate around the bond connecting them; the two extreme configurations are planar (the two benzene rings in the same plane) and the nonplanar in which the benzene rings are at a 90E angle to each other. The degree of planarity is largely determined by the number of substitutions in the ortho positions. The replacement of hydrogen atoms in the ortho positions with larger chlorine atoms forces the benzene rings to rotate out of the planar configuration.

Safe Department of Veterinary Physiology and Pharmacology Texas A&M University College Station, Texas 77843-4466 ABSTRACT Commercial polychlorinated biphenyls (PCBs) and environmental extracts contain complex mixtures of isomers and congeners which can be unequivocally identified and quantitated. PCBs elicit a spectrum of biochemical and toxic responses in humans and laboratory animals and many of these effects resemble those caused by 2.3,7,8- tetrachlorodibenzo-p-dioxin (TCDD) and related halogenated aromatic hydrocarbons which act through the aryl hydrocarbon (Ah) receptor signal transduction pathway. Structure- activity relationships developed for PCB congeners and metabolites have demonstrated that several structural classes of compounds exhibit diverse biochemical and toxic responses. Structure-toxicity studies suggest that the coplanar PCBs, namely 3.3 ',4,4'- tetrachlorobiphenyl (tetraCB), 3,3',4,4'-pentaCB and 3,3',4,4',5,5'-hexaCB and their monoortho analogs are Ah receptor agonists and contribute significantly to the toxicity of the PCB mixtures. Previous studies with TCDD and structurally related compounds have utilized a toxic equivalency factor (TEF) approach for the hazard and risk assessment of polychlorinated dibenzo-p-dioxin (PCDD) and polychlorinated dibenzofuran (PCDF) congeners in which the TCDD or toxic equivalents (TEQ) of a mixture is related to the TEQ = £ ( [ PCDF, x TEF, ]„ ) + £ ( [ PCDD, x TEFt ]„ ) TEFs and concentrations of the individual (i) congeners (note: n = the number of congeners). Based on the results of quantitative structure-activity data, the following TEFs are recommended for the coplanar and monoortho coplanar PCBs: 3,3',4,4',5-pentaCB, 0.1; 3,3',4,4',5,5'-hexaCB, 0.05; 3,3',4,4'-tetraCB, 0.02; 2,3,3', 4,4 '-pentaCB, 0.001; 2,3',4,4',5- pentaCB, 0.0001; 2,3,3',4,4',5-hexaCB, 0.0003; 2,3,3',4,4',5'-hexaCB, 0.0003; 2',3,4,4',5- pentaCB.
