IMMUNOGEN Recombinant Merlin isoform I

SPECIFICITY This antibody is specific for the ~69 kDa Merlin protein, also known as type 2 (NF2) or schwannomin. Cross reactivity with related was not observed.

REACTIVITY Reactivity has been confirmed with human U251 glioma cell lysates, primary cultured rat Schwann cells, and Merlin- overexpressed NIH-3T3 cell lysates in both Western blot and immunoprecipitation assays.

USAGE Working concentrations for specific applications should be determined by the investigator. The following concentration ranges are recommended starting points for this product.

BACKGROUND The neurofibromatosis type 2 (NF2) encodes a protein product, merlin, or schwannomin, that is related to the -- (ERM) family of proteins. The two predominant merlin isoforms, I and II, differ only in the carboxy terminal 16 residues, and only isoform I is anti-proliferative.1 Merlin is involved with numerous pathways that regulate cell growth and embryonic development. of merlin is associated with development of schwannoma tumors, and patients of NF2 are predisposed to other nervous system tumors such as ependymomas, meningiomas, and mesotheliomas.

Recent studies have shown that merlin increased stability by inhibiting the -mediated degradation of p53,2 and also directly inhibits the -activated -1 (PAK1), which is essential for malignant growth of NF2-deficient cells.3
