However, the mechanistic insight into the development of gallbladder cancer (GBC) and cancer-associated thrombosis remains unclear. This study aimed to investigate the mechanistic role of Sciellin (SCEL) in GBC cell proliferation and the development of venous thromboembolism. The expression level of SCEL was determined by immunohistochemical staining. Roles of SCEL in gallbladder cancer cell were determined by molecular and cell biology methods. SCEL was markedly upregulated in GBC and associated with advanced TNM stages and a poor prognosis. Furthermore, SCEL interacted with EGFR and stabilized EGFR expression that activates downstream PI3K and Akt pathway, leading to cell proliferation. In addition, SCEL induces tumor cell IL-8 production that stimulates the formation of neutrophil extracellular traps (NETs), accelerating thromboembolism. In xenografts, SCEL-expressing GBCs developed larger tumors and thrombosis compared with control cells. Methods: We conducted a prospective study involving 91 patients undergoing cardiac surgery. AKI was defined according to the AKIN definition. The renal outcomes within 7 days after operation were evaluated. Perioperative levels of urine Klotho and urine neutrophil gelatinase-associated lipocalin (NGAL) were measured by using ELISA. Results: Of 91 participants, 33 patients (36.26%) developed AKI. Of these AKI patients, 21 (63.64%), 8 (24.24%), and 4 (12.12%) were staged 1, 2, and 3, respectively. Serum creatinine in AKI patients began to slightly increase at first postoperative time and reached the AKI diagnostic value 1 day after operation. Postoperative urine Klotho peaked at the first postoperative time (0 h after admission to the intensive care unit (ICU)) in patients with AKI, and was higher than that in non-AKI patients up to day 3. The AUC of detecting AKI for urine Klotho was higher than urine NGAL at the first postoperative time and 4 h after admission to the ICU.

    Hypertension and Chronic Kidney Disease in China Nan Chen MD, Department of Nephrology, Ruijin Hospital, Shanghai Jiao Tong University School of Medicine The global rise in the number of patients with chronic kidney disease (CKD) and consequent end-stage renal disease (ESRD) requiring costly renal replacement therapy is putting a substantial burden on worldwide health care resources. Hypertension could be both an initial and progressive factor in CKD. Furthermore, it is a major risk factor for cardiovascular disease, stroke and other comorbidities in CKD patients. Given this background, further investigation of hypertension in CKD patients could be beneficial in improving both renal and cardiovascular outcomes. China is now the world’s largest developing country with a large population of CKD patients. According to the China National Nutrition and Health Survey 2002, one in six Chinese adults is hypertensive and few have their hypertension effectively under control [1]. Moreover, the prevalence of hypertension increases significantly thereafter--according to recently published data from the World Health Organization, the estimated prevalence INTERNATIONAL ADVISOR of raised blood pressure in China reached 38.2% in 2008 [2]. The changing epidemiology of hypertension Nan Chen nowadays shows that the number of hypertensive subjects has increased rapidly during the past decade, and the situation is even more serious in CKD patients. These data therefore underscore the necessity of further studies focusing on hypertension and CKD in Chinese patients. In our previous newsletter, we introduced recent hotspots for CKD research as well as epidemiological data on CKD in China [3] and received much positive EDITOR IN CHIEF feedback.
