A total of 200 field strains were isolated from spoiling meat, and 50 strains were isolated from freshwater or soil. In addition, 55 reference strains (including 23 type strains and 4 clinical strains) were obtained. The strains clustered into 25 clusters at the 75% level when the Jaccard similarity coefficient was used. The 10 clusters that were considered significant were assigned to the Pseudomonas fragi complex (131 strains), Pseudomonas lundensis (40 strains), Pseudomonasfluorescens biovar 1 (27 strains), P.fluorescens biovar 2 (5 strains), P.fluorescens biovar 3 (6 strains), P.fluorescens biovar 4 (16 strains), Pseudomonas aureofaciens-Pseudomonas chlororaphis (3 strains), Pseudomonas aeruginosa (4 strains), Pseudomonas glathei (2 strains), and Pseudomonas mephitica (2 strains). The P. fragi complex was further divided into subclusters; the major subcluster (comprising 93 strains, including the type strain) was regarded as P. fragi sensu stricto. P. fluorescens and allied bacteria closely matched the descriptions given by Stanier et al. (J. Gen. Microbiol. 43:159-271, 1966). The characteristics for the 10 significant clusters are given. Also given are criteria which differentiate the P. fragi subclusters. The phylogenetic relationships among the meat-associated taxa were calculated, P.fluorescens biovars 2 and 3 were clearly separated from the remaining taxa. -

Thus, the surveillance on MDR microorganisms has recently been strengthened, taking into account the control of antibiotic abuse as well as the mechanisms underlying the transfer of antibiotic genes (ARGs) among microbiota naturally occurring in the environment. Indeed, ARGs are not only confined to pathogenic bacteria, whose diffusion in the clinical field has aroused serious concerns, but are widespread in saprophytic bacterial communities such as those dominating the food industry. In particular, fresh dairy products can be considered a reservoir of Pseudomonas spp. resistome, potentially transmittable to consumers. Milk and fresh dairy cheeses products represent one of a few “hubs” where commensal or opportunistic pseudomonads frequently cohabit together with food microbiota and hazard pathogens even across their manufacturing processes. Pseudomonas spp., widely studied for food spoilage effects, are instead underestimated for their possible impact on human health. Recent evidences have highlighted that non-pathogenic pseudomonads strains (P. fluorescens, P. putida) are associated with some human diseases, but are still poorly considered in comparison to the pathogen P. aeruginosa. -

However subsequent molecular tests did not always confirm the results obtained, thus highlighting the limits of API 20NE. Instead RFLP ITS1 and the sequencing of 16S rRNA gene grouped the isolates into 6 clusters: Pseudomonas fluorescens (cluster I), Pseudomonas fragi (cluster II and V) Pseudomonas migulae (cluster III), Pseudomonas aeruginosa (cluster IV) and Pseudomonas chicorii (cluster VI). The pectinolytic activity was typical of species isolated from vegetable products, especially Pseudomonas fluorescens. Instead Pseudomonas fragi, predominantly isolated from meat was characterised by proteolytic and lipolytic activities. Key words: Pseudomonas fluorescens, enzymatic activity, ITS1. ABSTRACT CRAIG, KELLY. Examination of Pseudomonas sp. Response to Industrial Processing Stresses (Under the direction of Dr. Amy Grunden). Members of the genus Pseudomonas have drawn interest for their biotechnological and agricultural potential along with their medical importance as plant and animal pathogens. Several species of Pseudomonas have been targeted for their ability to repress microbial plant pathogens, insects, and nematodes. To succeed as viable candidates for agricultural applications, biological control strains need to survive the formulation process, prolonged periods of storage, and challenging environmental conditions. During the formulation process, beneficial bacteria can be dried to halt metabolism and improve shelf-life stability, transportability, and ease of application in the field. A high throughput screening strategy was developed to identify soil-associated microbes capable of surviving drying methods. The microbial diversity of soil bacterial communities was analyzed after exposure to spray drying and oven tray drying. In addition, a Gram-negative bacteria targeted isolation method was established to study the survival capabilities of asporulous Gram-negative bacteria. Bacillus, a Gram-positive spore forming bacterium, has an advantage in surviving and quickly recovering from harsh drying methods such as spray drying, whereas Pseudomonas sp. and other Gram-negative bacteria are capable of surviving milder formulation strategies such as oven tray drying. A diverse set of Pseudomonas species was subjected to heat shock conditions to determine which species are capable of surviving heat shock. A second study assessed the ability of a panel of protectants to improve stress recovery of the Pseudomonas strains. P. thermotolerans and P. aeruginosa were the top two species capable of surviving a 60°C five- minute heat shock. -

González3, Silvia Restrepo3 and Sandra Baena1* Abstract Here we present the physiological features of Pseudomonas extremaustralis strain USBA-GBX-515 (CMPUJU 515), isolated from soils in Superparamo ecosystems, > 4000 m.a.s.l, in the northern Andes of South America, as well as the thorough analysis of the draft genome. Strain USBA-GBX-515 is a Gram-negative rod shaped bacterium of 1.0–3. 0 μm × 0.5–1 μm, motile and unable to form spores, it grows aerobically and cells show one single flagellum. Several genetic indices, the phylogenetic analysis of the 16S rRNA gene sequence and the phenotypic characterization confirmed that USBA-GBX-515 is a member of Pseudomonas genus and, the similarity of the 16S rDNA sequence was 100% with P. extremaustralis strain CT14–3T. The draft genome of P. extremaustralis strain USBA-GBX-515 consisted of 6,143,638 Mb with a G + C content of 60.9 mol%. A total of 5665 genes were predicted and of those, 5544 were protein coding genes and 121 were RNA genes. The distribution of genes into COG functional categories showed that most genes were classified in the category of amino acid transport and metabolism (10.5%) followed by transcription (8.4%) and signal transduction mechanisms (7.3%). -
