Wiley, for being not only a very helpful mentor, but also a friend to all of his students. Working in a group that felt more like my family let me concentrate my energy on moving forward with my research rather than losing it in a tense unfriendly atmosphere. He has always been there for all of us by keeping the door of his office open and lending us a pair of ears to listen to all of our concerns and questions and to sincerely offer us his best. I would like to also thank Harry Rees, the technical engineer of the chemistry department, who helps everyone whenever there is something wrong with anything electrical. Without him many research plans would halt due to a puzzling electrical glitch. Making a life from scratch in a new country is quite difficult, and having to experience that along with a Ph.D. career makes it twice as hard. I would like to thank all of my dear friends that I had to leave behind in my country who never gave up on me, especially Anahita, Shima, iv Soheyla, Maryam, Mahsa, Arezoo, Raha, and Mehrnaz. I’d like to send my thanks to my family that I’ve never stopped missing: my aunts, uncles, and cousins. Being who we are at the moment is also partly because of those who hurt us the most in our lives, I would like to acknowledge the ones I had to erase from my life, but having them crossing my path made me a much stronger person. I am foremost thankful to my God who never let me down in the face of all those bad moments, and always had me protected under his mighty arms. He had me survive the worst moments with the least possible harm and the most treasured lessons, becoming the strongest version of myself. I am blessed to have a loving and supportive family who made it easier to achieve more in my career: my parents and brothers, as well as my husband and his wonderful family that I love no less than my own. I also truly wish that my sweet grandmother that I lost about ten years ago would hear my appreciation for her as I never forget her loving hands. I would like to specifically thank my mom, husband, and brother, Hesam. My mom is where I can define unconditional love, and Hesam was the reason I started enjoying science and learned how good I am at math and chemistry. Foremost, my husband, Kourosh is the reason that every afternoon I can find comfort, love and peace at home, regardless of the teething troubles at work. He might think that he is lucky to have me, but I believe that I am the lucky one to have someone this supportive and loving as my spouse. There are many mediocre things in my life, but the love I found through him and our son is not definitely one of them.     March 24, 2015 Mr. Miles Reitner, CEO Driscoll’s 345 Westridge Drive Watsonville, CA 95076-4169 Dear Mr. Reitner: I am enclosing a letter signed by nearly 10,000 consumers asKing Driscoll’s to suspend buying berries from SaKuma Brother’s Berry Farm in Burlington, Washington until farm owners and farmworKers negotiate a fair contact. As you are undoubtedly aware, farmworKers have been asKing for a fair contract covering wages and conditions for several years now. Because of SaKuma Brothers’ owners unwillingness to negotiate a fair contact, worKers have called for a boycott of these berries. Driscoll’s is one of SaKuma Brothers’ largest buyers and you have the ability to insist on a fair contract for farmworKers before you continue to purchase berries from this farm. Given your commitment to community and sustainability, this should be standard practice. The consumers who have signed the attached letter are not only expressing concern and solidarity for farmworKers, they are pledging to boycott all berries sold under the Driscoll’s brand until this is resolved. If you act now, a fair contract can be in place before the coming harvest season. This will ensure no further interruption of business to Driscoll’s or SaKuma Brothers and more importantly ensure decent jobs for hundreds of farmworKers whose tireless worK contributes to the ability of consumers to enjoy fresh berries year round. However, the maintenance of mitochondrial variation within species indicates a possible selective advantage conferred by favourable mtDNA mutations. I hypothesised that mtDNA variation may be maintained through interactions of mtDNA with environmental factors, including diet, temperature and the presence of stressors. I further hypothesised that experimentally manipulating the interactions of mtDNA and environmental factors may allow context specific favourable mutations in mtDNA to be discovered. Diet provides a strong environmental factor to identify favourable mtDNA mutations as the macronutrients of diet provide substrate to the mitochondria at different stages of the electron transport system to produce cellular energy. Thus modulating diet and other environmental factors may elucidate favourable mutations as these mutations may be context dependent, being favourable in one context, but deleterious in another. The overall goal of this thesis is to determine whether a single mtDNA mutation can have favourable effects on organismal fitness and health through interactions with environmental factors.
