BMC Genetics 2013, 14:73 http://www.biomedcentral.com/1471-2156/14/73 RESEARCH ARTICLE Open Access Insights into the European rabbit (Oryctolagus cuniculus) innate immune system: genetic diversity of the toll-like receptor 3 (TLR3) in wild populations and domestic breeds Joana Abrantes1,3, Helena Areal1,2 and Pedro J Esteves1,4* Abstract Background: Toll-like receptors (TLRs) belong to the innate immune system and are a major class of pattern recognition receptors representing the first line of the innate immune response. The TLR molecule is structurally composed by an ectodomain that contains leucine rich repeats (LRRs) that interact with pathogen associated molecular patterns (PAMPs), a transmembrane domain and a conserved cytoplasmic domain designated TIR (Toll-IL1 receptor) that is responsible for the intracellular signaling. TLR3 has been associated with the direct recognition of double-stranded viral RNA resulting from viral replication, while TLR7 and TLR8 target single-stranded viral RNA. In the European rabbit (Oryctolagus cuniculus), TLR7 and TLR8 were reported to be absent and pseudogenised, respectively, making TLR3 the only available TLR for the recognition of viral RNA. Thus, the levels of diversity of TLR3 were evaluated in the European rabbit by analysing different genetic backgrounds and exposure to pathogen pressures. Results: We detected 41 single nucleotide polymorphisms (SNPs) in the coding sequence of TLR3. The highest diversity was observed in the wild populations of Iberian Peninsula, between 22–33 polymorphic positions. In the French population, 18 SNPs were observed and only 4 polymorphic positions were detected in the domestic breeds. 14 non-synonymous substitutions were observed, most of them in the LRR molecules.

Here, we provide the first comprehensive survey of nucleotide polymorphism and linkage disequilibrium (LD) within and among rabbit breeds. We resequenced 16 genomic regions in population samples of both wild and domestic rabbits and additional 35 fragments in 150 rabbits representing six commonly used breeds. Patterns of genetic variation suggest a single origin of domestication in wild populations from France, supporting historical records that place rabbit domestication in French monasteries.

Consumers’ attitude was evaluated in Spain, Italy, France, Poland, Hungary, China, Brazil and Mexico based on the respondents’ opinion regarding origin, production conditions, slaughtering methods and breed. Results revealed that breed was an inessential factor. The origin of rabbits was the most important in Italy, France, Hungary and Brazil (in the latter country the slaughtering method and the level of processing were also highly valued), feeding of animals received the highest scores in Spain, Poland and Mexico (in the latter also the slaughtering method), while the level of processing was the foremost factor in China. The preference of fresh meat was the highest in Spain, France and Mexico, and that of frozen in Brazil and Mexico. The highest preference of a whole carcass was found in France and Mexico. The thigh was mostly favored in France and that of the loin in Mexico. The prepared meat products (roasted, smoked and semi-finished) were the most popular in Mexico.

Cuniculture, 87a Chemin de Lassère, 31450 Corronsac, France [email protected] ABSTRACT In this invited communication the author proposes a list of nutritional recommendations for rabbits of different categories: growing from 18 to 42 days, from 42 to 80 days, for breeding does according to productivity (40-50 kits weaned per doe/year or more than 50) and for a single diet suitable for all rabbits. Recommendations taking account the last published data, are divided in 2 groups. The first corresponds to nutrients which contribute mainly to feed efficiency: digestible energy, crude and digestible protein, amino acids, minerals, and fat-soluble vitamins. The second group corresponds to nutrients which contribute mainly to nutritive security and digestive health: different fibre components (lignins, cellulose, hemicelluloses) and their equilibrium, starch and water soluble vitamins. In a second part, 387 papers published during the last 30 years on raw material utilisation in rabbit feeding were analysed. In a total of 14 tables, the 542 corresponding experiments were summarised each by the identification of the raw material, by the highest level of incorporation used in the experiment, by the highest acceptable level, by the main ingredient(s) replaced by the raw material studied, and finally by the authors reference. RASHWAN 2 Rabbits behavioural response to climatic and managerial conditions – a review Abstract The domestic rabbit is deprived of the protection of burrows and of a social hierarchy and live in a limited space which is a fraction of a metre of a hard sanitized space, removed from odours, markers and social interaction. Hot climate was the main cause for abnormal maternal and sexual behaviour. Exposure to high ambient temperature induces rabbits to try to balance their excessive heat load by using different means. The doe that was capable to produce 10 litters a year may give only 4 to 5 litters in hot climate Noise in rabitries causes adverse effects including nervous and behavioural abnormalities and can cause a startled response and traumatic injuries to limbs and back. Particularly, most concern about noise effects has traditionally focused on impairment of reproductive and maternal behaviours, although few controlled studies have been done to support the observations of animal caretakers that noise inhibits production. Moon phases are one of the main causes of abnormal sexual behaviour in females. Crowdness causes that rabbits become aggressive and bite one another during the first few days of nest sharing, while successive litters live together. Keeping rabbits singly in cages is not compatible with the demand of housing with respect to animal welfare, since the singly caged rabbit is exposed to natural external stimulus, has no social contact and has no conditions for suitable locomotion.
