FORM This affinity-purified mouse monoclonal antibody is supplied as a 200 µL aliquot at a concentration of 0.5 mg/mL in PBS, pH 7.4, containing 0.1% sodium azide. This antibody is highly purified from mouse ascites by protein A chromatography.

IMMUNOGEN Peptide corresponding to amino acid residues from the C-terminal region of human cathepsin V protein (accession # O60911, NP_001324), which is 99% homologous with chimpanzee, 94% rhesus monkey, 82% canine, and 79% with bovine.

SPECIFICITY This antibody is specific for the cathepsin V (, cathepsin U) protein. On Western Blots of human recombinant pro-cathepsin V, it identifies the target band at ~40 kDa. This antibody also can identify a 24 kDa mature two-chain cathepsin V. This antibody does not cross react with closely related human cathepsin L which has 78% sequence identity, and is negative to human B, F, H, K, S, and W.

REACTIVITY Reactivity has been confirmed with recombinant human pro-cathepsin V protein by Western Blotting, and with normal human keratinocytes by immunofluorescence. Based on amino acid sequence homology, reactivity with chimpanzee, rhesus monkey, canine and bovine is also expected.

BACKGROUND Cathepsins are a large family of lysosomal cysteine that have a vital role in mammalian cellular turnover, e.g. bone resorption. They degrade polypeptides and are distinguished by their substrate specificities. There are approximately a dozen members of this family, which are distinguished by their structure and which proteins they cleave. Most of the members become activated at the low pH found in lysosomes. Thus, the activity of this family lies almost entirely within those organelles.1 Cathepsins can be used as biological markers of malignant tumors and have been associated with Alzheimer and stroke. They can be used as targets for cancer therapy and prognosis of cancer.2

Degradation of the arterial elastin matrix contributes to the loss of elasticity of the arterial wall resulting in atherosclerosis. A novel human elastolytic cysteine , cathepsin V, also known as cathepsin L2 and cathepsin U has been detected in atherosclerotic plaque specimens. Cathepsin V is expressed by macrophages and exhibits the most potent elastase activity yet described among human proteases.3 Cathepsin V, specifically expressed in the thymus, testis, and corneal epithelium, can be autocatalytically converted to the mature form at pH 4.0.
