RAPID DETECTION OF POLYCYCLIC AROMATIC HYDROCARBONS FROM COOKING OIL USING ON-CHIP ULTRA-THIN LAYER CHROMATOGRAPHY WITH SURFACE ENHANCED RAMAN SPECTROSCOPY by JING CHEN (Under the Direction of YAO-WEN HUANG) ABSTRACT Polycyclic aromatic hydrocarbons (PAHs) are a class of potent environmental pollutants which exhibit carcinogenic, mutagenic, and teratogenic properties. High levels of PAHs are found in food products cooked at high temperatures, such as fried foods and repeatedly used or illicitly recycled oils. Screening and detection of PAH compounds are an urgent demand to ensure cooking oil safety, but current techniques require labor-intensive and time-consuming sample preparation procedures. To shorten the detection time and avoid complicated sample pretreatment, a new technique combining ultra-thin layer chromatography (UTLC) and surface enhanced Raman spectroscopy (SERS) has been proposed and evaluated for rapid screening of PAHs in oil samples. The UTLC-SERS method utilizes the nanoporous and Raman enhancing properties of the silver nanorod (AgNR) substrates for chromatographic separation of mixture samples directly on the sensing surface within minutes. In this dissertation, the UTLC-SERS principles were demonstrated using model Raman reporter molecules, and subsequently applied in the detection of PAHs. The AgNR substrate was modified with mercaptoethanol (ME), which served as the stationary phase in UTLC. The mobile phase was optimized for the separation of three representative PAHs, benz(a)anthracene (BaA), benzo(a)pyrene (BaP), and pyrene (P). PAHs were extracted from artificially contaminated vegetable oil samples through a 1- min acetonitrile extraction procedure, and the organic phase was directly used for UTLC-SERS without further treatment. The UTLC process served to elute target PAHs from the oil matrix to near the solvent front, where the SERS signal of BaA, BaP, and P could be identified at concentrations as low as 50 µg PAH/mL oil. -

Premier Li Keqiang, Head of the National Food Safety Commission, State Council, People Republic of China.1 Melamine contaminated milk, toxic bean sprouts, aluminum dumplings, glow-in-the-dark pork, gutter oil, cadmium rice, toxic preserved fruits, fluorescent bleached mushrooms, fake eggs and the list goes on and on – this has been the sad reality of the Chinese food industry for the past several years.2 Millions of Chinese citizens are paralyzed with fear of another food safety scare and doubt about the quality and safety of their next meal. More and more parents and caretakers around China are justifiably concerned about feeding their infants baby formula produced in China. -

Turner (China Environment Forum) April 17, 2019 Prepared by Eastern Research Group (ERG) and the Wilson Center’s China Environment Forum under Contract EP-BPA-18-H-0009 for the United States Environmental Protection Agency 1 Introduction In 2016, the Wilson Center’s China Environment Forum (CEF) produced a scoping paper for the Global Methane Initiative (GMI) entitled Scaling China’s Sludge Mountains: Challenges and Opportunities to Improve Sludge Treatment and Disposal in China’s Tier-2 Cities. In the report, we highlighted the potential of sludge-to-energy (StE) technologies for methane capture and utilization in China as a solution to address water-energy choke points. With our extensive research and reporting experience on water-energy, sludge, and wastewater issues in China, CEF then conducted out additional research and meetings to help GMI identify potential tier-2 cities and partners in China to help catalyze 1 to 2 StE pilots in China at either wastewater treatment or stand-alone sludge treatment plants. Our Sludge Mountains report highlighted that while China has 50+ StE plants, most are not in use or are underutilized. One of the biggest obstacles is the problem of excessive stormwater runoff diluting the urban wastewater and the resulting sludge lacks sufficient organic material to produce methane. Many Chinese cities are working to address the stormwater runoff under the central government’s “sponge city” campaign, which is promoting green urban infrastructure such as green rooftops, porous pavements, and rainwater harvesting. -
