Identificação de proteases de leptospira envolvidas com mecanismos de escape do sistema complemento humano. [tese (Doutorado em Imunologia)]. São Paulo: Instituto de Ciências Biomédicas, Universidade de São Paulo; 2014. A leptospirose é uma zoonose mundialmente disseminada que representa um grave problema de saúde pública. Microrganismos patogênicos, notadamente os que atingem a circulação sanguínea como a leptospira, desenvolveram múltiplas estratégias de evasão ao sistema imune do hospedeiro, em especial ao sistema complemento. Neste contexto, o principal objetivo deste trabalho foi analisar a secreção de proteases capazes de clivar moléculas do sistema complemento por leptospiras patogênicas, o que constituiria um novo mecanismo de evasão imune para este patógeno. Nove estirpes de leptospira foram selecionadas para este trabalho: sete patogênicas e duas saprófitas. Para a obtenção dos sobrenadantes de cultura, bactérias cultivadas em meio EMJH modificado foram transferidas para PBS pH 7,4 e incubadas a 37 oC por diferentes tempos. Após incubação, os sobrenadantes foram coletados e analisados quanto à atividade inibitória e proteolítica sobre componentes do sistema complemento. O efeito sobre a ativação do complemento foi quantificado por ELISA, onde a atividade das três vias foi medida separadamente. Verificamos que o sobrenadante de leptospiras patogênicas foi capaz de inibir a ativação de todas as vias do complemento, enquanto o da espécie saprófita não inibiu nenhuma delas. -

A major drawback of serology in diagnosis is that it needs sufficient levels of anti-Leptospira antibodies, thus jeopardizing confirmation in the early acute phase of disease. The cross agglutinin absorption test (CAAT) that determinesLeptospira serovars is a technically demanding and laborious method and therefore is only performed at a few laboratories. Novel molecular diagnostic tests mainly rely on the polymerase chain reaction (PCR). PCR perfectly complements serological testing, since it is especially sensitive in the first 5 days after onset of symptoms. Real time PCR is rapid and has been validated for their high clinical accuracy. The introduction of molecular techniques for definingLeptospira species revolutionized the categorization of strains in this genus, as species and serogroups appeared to show little correlation. -

Zuerner, and Ben Adler Summary The complete nucleotide sequences of three and, in the not too distant future, six strains from the genus Leptospira will be available. Managing and maintaining these data will be a perpetual problem unless a system is devised to address this issue. We propose a central role for the Interna- tional Union of Microbiological Societies Subcommittee on the Taxonomy of Leptospira in main- taining and updating the annotated leptospiral genome sequences. The first step in this process is provided as part of this publication, namely a revision of the annotation of the three published genomes, and an internet location for the current versions of these genomes. Key Words: Leptospira; comparative genomics; spirochetes; genome annotation. 1. Introduction Leptospirosis is a zoonosis of worldwide distribution. Significant efforts aimed at raising the standard of diagnostics (1) and the collection and collation of disease data (via LeptoNet; www.leptonet.net) will correct what is likely to be a significant under- estimation of the contribution of this disease to human morbidity and mortality. Lepto- spirosis is caused by pathogenic serovars from the genus Leptospira and in the current species classification schema these pathogenic serovars are usually classified as Lepto- spira interrogans, Leptospira borgpetersenii, Leptospira kirschneri, Leptospira noguchii, Leptospira meyeri, Leptospira weilii, or Leptospira santarosai. Analysis based on 16S ribosomal RNA (rRNA) gene analysis shows that these serovars are found in one of the three phylogenic clades into which the genus can be divided (2).
