EP 2 185 011 B1 Printed by Jouve, 75001 PARIS (FR) EP 2 185 011 B1 Description TECHNICAL FIELD 5 [0001] This invention relates to a tablet or capsule which comprises an extract of a plant of the genus Salacia and a flavonoid. BACKGROUND ART 10 [0002] Recently, the number of persons having obesity or increased blood sugar level caused by the ingestion of high calorie food and overeating or lack of exercise, so-called reserve group of diabetes mellitus, has been increasing. To prevent this, supplements aimed at suppressing sugar absorption and fat absorption have been placed on the market. In addition, for the purpose of suppressing high blood sugar level and preventing diabetes mellitus,α-glucosidase inhibitors which suppress formation of glucose from starch and oligosaccharides are also on the market. 15 [0003] To prevent obesity, prevention of overeating by controlling consumption of calories and exercise are necessary, but their everyday practice is difficult in reality. In addition, theα-glucosidase inhibitors are effective not only as a therapeutic agent for diabetes mellitus but in dissolving obesity caused by overeating and lack of exercise and diet control. However, these agents (e.g., acarbose (mfd. by Bayer, trade name Gulcobay)), voglibose (mfd. by Takeda Chemical Industries, trade name Basen) and the like) are difficult to obtain, and when these are taken, absorption of 20 polysaccharides into the body can be suppressed but absorption of glucose and the like monosaccharides cannot be suppressed, so that significant sugar absorption-suppressing effect cannot be expected. In addition, problems of various side effects occur sometimes after their taking. Because of this, concern has been directed for the development of a safe and inexpensive natural-origin composition. [0004] As a natural material effective in satisfying such a requirement for obesity and sugar absorption suppression, 25 there are plants of the genus Salacia. The root and trunk of a plant of the genus Salacia have been used as a natural drug by a traditional medical science, aayurveda, in India and Sri Lanka. It has been handed down in Sri Lanka that the root skin of Salacia reticulata is effective in treating rheumatism, gonorrhea and a skin disease and is also used in the treatment of initial stage diabetes mellitus. In India, a root of Salacia oblonga is used in similar treatments, and it is said that Salacia chinensis is also used in the treatment of diabetes mellitus (FOOD Style 21, vol. 6, no. 5, pp. 72 - 78). 30 [0005] Thus, it has been handed down that plants of the genus Salacia are effective in the prevention and early stage treatment of diabetes mellitus. In recent years, it has been reported that a plant of the genus Salacia has the action to suppress increase of blood sugar level, and its action mechanism is the sugar absorption suppressing action based on the α-glucosidase activity inhibition (FOOD Style 21, vol. 6, no. 5, pp. 72 - 78). [0006] In addition, there are patents on the compounds which are contained in the extraction components of the genus 35 Salacia and have the action to inhibitα -glucosidase activity (Japanese Patent No. 3030008, JP-A-2004-323420 and JP-A-2000-86653), and their application examples and patents as anti-diabetic agents based on theα-glucosidase activity inhibitory action (JP-A-9-301882 and Japanese Patent No. 3261090). [0007] Since the sucrase inhibitory activity of most of the supplements on the market which use Salacia extract powder is standardized as 250 mg/ml or less, and when their sucrase inhibitory activity is actually measured, it is mostly about 40 350 mg/ml, so that it is necessary to increase the number of tablets in order to ingest effective amount. [0008] Concern has been directed toward the development of a high concentration Salacia extract powder, for the purpose of decreasing the number of tablets by increasing blending amount of Salacia extract powder per tablet. [0009] In addition, it is known that a large quantity of catechins as a kind of polyphenols are contained in the flavonoid- containing tea extract to be used in the invention. Anti-oxidation action, bactericidal action, anticancer action, blood 45 pressure lowering action, blood sugar level suppressing action and the like many physiological actions have been reported on such catechins, and in recent years, effectiveness of the fat combustion for its somatic fat reducing effect has further been reported (Shimotoyodome A., et al., Med. Sci. Sport Exerc., 37, 1884 - 1892, 2005 and Murase T., et al., Int. J. Obes. Relat. Metab. Disord., 30, 561 - 568, 2006). [0010] It has been reported that the effect to decrease body weight and somatic fat (abdominal total fat area, visceral 50 fat area) can be seen by the ingestion of high concentration catechin for a prolonged period of time. It is considered that a fat combusting enzyme in cells of the liver is activated by the high concentration catechin and thereby increase energy consumption by activating the fat metabolism, thus resulting in the reduction of somatic fat (Tsuchida T., Itakura H., Nakamura H., Prog. Med., 22, 2189 - 2203, 2002, Hackett A.M., et al., Eur. J. Drug Metab. Pharmacokinet, 8, 35 - 42, 1983, Murase T., et al., Int. J. Obes. Relat. Metab. Disord., 26, 1459 - 1464, 2002 and Ota N., et al., J. Health. Sci., 51, 55 233 - 236, 2005). [0011] It is possible to ingest catechin by drinking a green tea or the like tea, but since the content per cup is small, it is necessary to ingest a tea extract powder in the form of tablets or capsules for ingesting a desired amount. [0012] In addition, a tea extract contains catechin, epicatechin, gallocatechin, epigallocatechin, catechin gallate, epi- 2 EP 2 185 011 B1 catechin gallate, gallocatechin gallate and epigallocatechin gallate as catechins, or epigallocatechin gallate, epigallo- catechin, epicatechin gallate and gallocatechin in order of the content. [0013] In addition, the flavonoid to be used in the invention is roughly divided into flavonols, isoflavones and catechins. Among these, flavonols are a general term of polyphenols, and particularly among them, anti-oxidation action, bactericidal 5 action, anticancer action, immune action, fat combustion and the like many physiological actions have been reported on resveratrol, and further, its effectiveness for fat combustion has been confirmed. Also, effectiveness of the life- prolonging effect of mice which ingested a high calorie feed supplemented with resveratrol has been reported (Science. 275, 218, (1997) and Nature. Nov. 16th. (2006)). [0014] It is possible to ingest resveratrol by drinking or eating a grape extract or drinking red wine or the like, but since 10 the content per once is small, it is necessary to drink a large volume of red wine or ingest it in the form of tablets or capsules for ingesting a desired amount. [0015] In addition, vitamin C absorption support, anti-oxidation action, immune action and the like physiological actions have been reported particularly on quercetin among polyphenols, and it was further confirmed that this is effective in suppressing fat absorption (JP-T-2001-524119 (the term "JP-T" as used herein means a published Japanese translation 15 of a PCT patent application) and WO 01/076580). [0016] It is possible to ingest quercetin by eating an onion, but since the content per once is very small because the skins are not generally used for food.

It is widely used in treating diabetes, a chronic disorder in metabolism of carbohydrates, proteins and fat due to absolute or relative deficiency of insulin secretion with/without varying degree of insulin resistance. The decoction of S. reticulata roots is also used in the treatment of gonorrhea, rheumatism, skin diseases, haemorrhoids, itching and swelling, asthma, thirst, amenorrhea and dysmenorrheal. Presence of mangiferin (a xanthone from the roots), kotalanol and salacinol (from the roots and stems) have been identified as the antidiabetic principles of S. reticulata. Chemical constituents such as 1,3- diketones, dulcitol and leucopelargonidin, iguesterin, epicatechin, phlobatannin and glycosidal tannins, triterpenes, and 30-hydroxy-20(30) dihydroisoiguesterin, hydroxyferruginol, lambertic acid, kotalagen- in 16-acetate, 26-hydroxy-1,3-friedelanedione, maytenfolic acid have also been detected in the roots of S. reticulata. The antidiabetic property of Salacia is basically attributed to the inhibitory activity of in- testinal enzymes (α-glucosidase and α-amylase). Inhibition of intestinal enzymes delays glucose absorp- tion into the blood and suppresses postprandial hyperglycemia, resulting in improved glycemic control. Furthermore, mangiferin has been reported to inhibit aldose reductase activity delaying the onset or progression of diabetic complications. Though diabetes has now become an epidemic affecting millions of people worldwide, neither insulin nor other modern pharmaceuticals has been shown to modify the course of diabetic complications mainly due to the multifactorial basis that involves both genetic and environmental risk factors.

It was found that on an average, the plant extract with higher levels of phenol content had higher antioxidant activity. The antioxidant activities were found to be highest and lowest at methenolic and hexane extracts, respectively in both the assays. At 1mg/ml concentration, DPPH and ABTS assays showed highest activity in the methanol extract of 84 and 99% respectively. The presence of phenolic compounds was confirmed by HPLC. INTRODUCTION In Siddha and Ayurveda, plants and food extracts were used as preventive measures and rejuvenating agents for the diseases. During the past decade there has been an ever increased demand especially from the developed countries for more and more drugs from the plant sources. This revival of interest in plant-derived drugs is mainly due to current widespread belief that green medicine is safe and more dependable than the costly synthetic drugs, many of which having the adverse side effects.

Hence, the present study was aimed to estimate three important secondary metabolites such as phenol, flavonoid and tannin. Antioxidant assays such as DPPH assay and ABTS assay and Total Antioxidant Capacity were also studied for this medicinal genus. Seven species of Salacia were considered for this present study. They are Salacia beddomei Gamble, Salacia chinensis L, Salacia fruticosa Heyne ex Lawson, Salacia gambleana Whiting & Kaul, Salacia macrosperma Wight, Salacia malabarica Gamble, and Salacia oblonga Wall. Various concentrations such as 20µg, 40µg, 60 µg, 80 µg and 100 µg were taken for all studies and the values are entered in terms of Mean±SD. In case of DPPH assay and ABTS assay, IC50 values are calculated using ANNOVA and Total Antioxidant Capacity was calculated using a calibration curve.

It is a placebo controlled randomized crossover design study. Subjects were given oral sucrose solution along with capsules and plasma glucose and insulin responses were analyzed. Blood samples were collected at 0, 30, 60, 90, 120, and 180 minutes after administration. AUC insulin significantly lowered after ingestion of SCE. No significant adverse events were observed. Datar Biodiversity and Palaeobiology group, Agharkar Research Institute, Pune - 411 004, Maharashtra, India. __________________________________________________________________ ABSTRACT In Traditional Indian Medicine three species of genus Salacia viz. S. oblonga Wall., S. chinensis L. and S. reticulata Wight are known as 'Saptarangi'. There is no detailed pharmacgonostic profile of S. oblonga; hence study of its stem and root was carried out. Macroscopic characters were documented. Microscopically stem and root showed distinct stone cells and red colouring matter. Total and water soluble ash were higher in root (1.57±0.012) while stem showed higher acid insoluble ash (0.26±0.03). Methanolic extract was higher in root (0.092) and stem (0.057) than other solvents. Primary metabolites: stem showed higher total carbohydrates (71.381±5.1) while root showed higher total proteins (10.561±0.5). Secondary metabolites were higher in root (Polyphenols - 0.79±0.04, Tannins - 0.12±0.0005). Antioxidant activity was observed to be more in the root which could be due its higher concentration of polyphenols. These developed parameters may prove significant in identification of the crude drug ‘Saptarangi'. India _____________________________________________________________________________________________ ABSTRACT The study was conducted to determine the antioxidant activity, total phenol and flavonoid content of the stem of Salacia oblonga, collected from five geographically distant regions of Western Ghat, Karnataka were examined using extracts of aqueous, ethanol, methanol, chloroform and petroleum ether. The five different solvent extracts of S. oblonga stem were evaluated for antioxidant activities by DPPH (1,1 – diphenyl -2- picryl-hydrazyl) radical scavenging activity using Butylated Hydroxy Toluene (BHT) as standard. Among five accessions with different solvents used, maximum antioxidant activity was found in aqueous stem extract (84.3 %) from Hubli(2) followed by others. Total phenol and flavonoid contents were quantitatively estimated. The aqueous stem extract of Salacia oblonga (Hubli(2) - Karnataka) was found maximum in total phenol and flavonoid contents were 34.63 mg GAE /g and 18.72 mg QE /g respectively. The aqueous stem extracts of S. oblonga had superior level of antioxidant activity. The powerful free radical scavenging effect is attributed to the greater amount of phenol and flavonoid compounds in the aqueous stem extract.
