No. C-3881.) B. 10.4% (w/v) Milk Solution with 10 mM Calcium Chloride (Milk) (Prepare by dissolving 20.9 g of Carnation Instant Nonfat Dry Milk Powder in 200 ml of deionized water or use skimmed cow's milk. Then add 2 ml of Reagent A.) C. 0.1% (w/v) Rennet Standard Solution (Rennet Std) (Prepare 10 ml in deionized water using Rennet, Sigma Prod. No. R-3376.) D. Rennin Enzyme Solution (Rennin) (Immediately before use, prepare a solution containing 0.01 - 0.05 mg/ml of Rennin in cold deionized water. Hébert, 2003 i RÉSUME COURT La préséniline-1 (PS1) mutée est associée à un développement très précoce de la maladie (chez les individus de 24-55 ans) d’Alzheimer. Dans la cellule, la PS1 existe principalement sous forme de fragments (N-terminaux et C-terminaux, respectivement NTFs et CTFs) qui semblent nécessaires à sa fonction et à l’activité γ- secrétase étroitement liée à la formation des peptides β-amyloïdes neurotoxiques. Nous avons étudié l’organisation structurale et fonctionnelle de la PS1, ses fragments, et leurs relations avec les protéines impliquées dans la production des peptides β- amyloïdes. Dans un premier temps, des expériences en levure, confirmées avec des techniques d’immunobuvardage et d’immunoprécipitation en cellules de mammifères, nous ont permis de mettre en évidence un rôle de l’holoprotéine de la PS1 dans la génération des fragments NTFs et CTFs. De plus, il a été possible de démontrer une interaction directe entre PS1 et BACE (enzyme de type aspartyl protéase aussi appelée β-secrétase), les deux joueurs clés dans la production des peptides β- amyloïdes. Finalement, l’étude du rôle fonctionnel de cette interaction nous a fournis des résultats permettant de suggérer une régulation de la PS1 par BACE. -

Among the many pathologic events observed in the progression of AD, changes in amyloid β peptide (Aβ) metabolism proceed fastest, and precede clinical symptoms. BACE1 (β-secretase 1) catalyzes the initial cleavage of the amyloid precursor protein to generate Aβ. Therefore inhibition of BACE1 activity could block one of the earliest pathologic events in AD. However, therapeutic BACE1 inhibition to block Aβ production may need to be balanced with possible effects that might result from diminished physiologic functions BACE1, in particular processing of substrates involved in neuronal function of the brain and periphery. Potentials for beneficial or consequential effects resulting from pharmacologic inhibition of BACE1 are reviewed in context of ongoing clinical trials testing the effect of BACE1 candidate inhibitor drugs in AD populations. Treatment is limited to therapeutics that alleviate symptoms of memory loss, but are effective for a relatively short duration during and after which disease progression continues. -

Their coagulation efficiency was measured in bovine, buffalo, goat and sheep milk incubated at different temperatures. The milk-clotting and proteolytic activities as well as the lactodynamographic parameters were determined considering animal rennet as a reference coagulant. The vegetable coagulant, extracted from C. cardunculus pistils, proved to be the most suitable milk-clotting enzyme for cheesemaking, since it possesses similar milk clotting properties to conventional calf rennet. F. carica latex, but seemed to be a promising alternative coagulant at higher temperatures. The strong proteolytic activity of papain caused poor milk coagulation in all milk samples. To conclude, this result also supports the original hypothesis of this study that the excessive proteolytic nature of plant coagulants can negatively affect the cheesemaking process. The optimization of using a plant rennet in a dairy application can be done by selecting the appropriate plant rennet with a consistent clotting efficiency. -

In vitro data revealed a biochemical link between Reelin-mediated signaling, Tau phosphorylation, and amyloid precursor protein (APP) processing. To directly address the role of Reelin in amyloid- plaque and Tau pathology in vivo, we crossed heterozygous Reelin knock-out mice (reeler) with transgenic AD mice to investigate the temporal and spatial AD-like neuropathology. We demonstrate that a reduction in Reelin expression results in enhanced amyloidogenic APP processing, as indicated by the precocious production of amyloid- peptides, the significant increase in number and size of amyloid- plaques, as well as age-related aggravation of plaque pathology in double mutant compared with single AD mutant mice of both sexes. Numerous amyloid- plaques accumulate in the hippocampal formation and neocortex of double mutants, precisely in layers with strongest Reelin expression and highest accumulation of Reelin plaques in aged wild-type mice. Moreover, concentric accumulations of phosphorylated Tau-positive neurons around amyloid- plaques were evident in 15-month-old double versus single mutant mice. -

They have no common phylogenetic origin but can be found in most protist evolutionary branches. Acanthamoeba is a common FLA that can be found worldwide and is capable of infecting humans. The main disease is a life altering infection of the cornea named Acanthamoeba keratitis. Additionally, Acanthamoeba has a close relationship to bacteria. Acanthamoeba feeds on bacteria. At the same time, some bacteria have adapted to survive inside Acanthamoeba and use it as transport or protection to increase survival. When conditions are adverse, Acanthamoeba is capable of differentiating into a protective cyst. -

Its mostly described function is intracel- lular catabolism in lysosomal compartments, other physiological effect include hormone and antigen processing. For almost two decades, there have been an increasing number of data describing additional roles imparted by cathepsin D and its pro-enzyme, resulting in cathepsin D being a specific biomarker of some diseases. These roles in pathological conditions, namely elevated levels in certain tumor tissues, seem to be connected to another, yet not fully understood functionality. However, despite numerous studies, the mechanisms of cathepsin D and its precursor’s actions are still not completely understood. From results discussed in this article it might be concluded that cathepsin D in its zymogen status has additional function, which is rather dependent on a “ligand-like” function then on proteolytic activity. Des solutions de rechange à l'utilisation de la bentonite sont activement recherchées en raison des problèmes technologiques, organoleptiques et de durabilité associés à son utilisation. Dans cette étude, MpAPr1, une protéase aspartique extracellulaire préalablement isolée et partiellement caractérisée à partir de la levure Metschnikowia pulcherrima, a été clonée et exprimée de manière hétérologue dans la levure Komagataella pastoris. Les propriétés enzymatiques de MpAPr1 ont été initialement caractérisées dans un extrait brut. Après plusieurs essais faisant appel à différentes techniques, MpAPr1 a été purifié avec succès par chromatographie échangeusede cations.
