Improving the bioavailability of pharmacologically active peptides requires in-depth investigation into the noncovalent interactions between the protease and the peptide substrate under consideration. To this end, different strategies such as the use of unnatural building blocks and fluorinated amino acids have become standard approaches in protein engineering. The incorporation of fluorine into amino acids has attracted much interest in recent years due to the unique stereoelectronic properties of fluorine, which have already proven useful in the development of therapeutically active small molecules. In this manner, the current thesis presents data on the systematic investigation of the influence of side chain fluorination on proteolytic stability of peptide sequences that are based on ideal protease substrates. Several model peptides were designed according to the specificities of serine and aspartic proteases; three different control sequences were modified by introducing either 2-aminobutyric acid (Abu) or one of its fluorinated derivatives at the P2, P1'or P2' positions. Through the use of an RP- HPLC assay with fluorescence detection, the proteolytic stabilities of these peptides toward α- chymotrypsin, pepsin, elastase, proteinase K and human blood plasma proteases were determined. Molecular modeling was used to support the interpretation of the structure-activity relationship based on the analysis of potential ligand-enzyme interactions. In all cases, increases and decreases in proteolytic stability were observed for the different isolated enzymes and the human blood plasma, and these effects depend upon the particular peptide sequence, the fluorine content of the side chain, and the position of substitution relative to the cleavage site. Interestingly, in some cases fluorination leads to dramatic improvements in resistance to degradation: namely, TfeGly and DfeGly at the P2’ position with pepsin; DfeGly at the P2 position with chymotrypsin; TfeGly and DfeGly at the P2 and P2’ positions with proteinase K; and TfeGly at the P2 and P1’ positions with elastase. Our observations indicate that although the fluorination of peptide substrates does not always have predictable effects on proteolytic stability, this strategy for developing more bioavailable peptide therapies is promising; in particular, part of this thesis was devoted to establishing an analytical approach for the identification of fluorinated peptide-based HIV-1 entry inhibitors. I II Kurzzusammenfassung Die Proteasestabilität ist ein wichtiges Kriterium bei der Erntwicklung von Peptid-basierten Medikamenten. Daher umfasst die Verbesserung der Bioverfügbarkeit von pharmakologisch aktiven Peptiden eine gründliche Untersuchung der nichtkovalenten Interaktionen zwischen Protease und Peptidsubstrat. In diesem Zusammenhang gehören verschiedene Strategien, wie beispielsweise die Verwendung von nicht-natürlichen Aminosäuren mittlerweile zu etablierten Methoden im Protein Engineering. Der Einbau fluorierte Aminosäuren hat in den letzten Jahren besonders an Bedeutung gewonnen, da das Element Fluor über einzigartige stereoelektroische Eigenschaften verfügt, die sich bei der Entwicklung von Pharmazeutikern als nützlich erwiesen haben. Intrapancreatic trypsin activation is an early event in experimental acute pancreatitis in rodents, suggesting that trypsin is a key driver of pathology. In contrast with trypsin, the pancreatic protease chymotrypsin serves a protective function by mitigating trypsin activation through degradation. In humans, loss-of-function mutations in chymotrypsin C (CTRC) are common risk factors for chronic pancreatitis; however, the pathogenic effect of CTRC deficiency has not been corroborated in animal models yet. Here we report that C57BL/6 mice that are widely used for genetic manipulations do not express functional CTRC because of a single-nucleotide deletion in exon 2 of the Ctrc gene. We restored a functional Ctrc locus in C57BL/6N mice and demonstrated that in the Ctrc+ strain, the severity of cerulein- induced experimental acute and chronic pancreatitis was significantly ameliorated. Improved disease parameters were associated with reduced intrapancreatic trypsin activation, suggesting a causal link between CTRC-mediated trypsinogen degradation and protection against pancreatitis.

Besides a stabilizing effect and protection against proteolysis, several proteases show a significant influence of glycosylation on the catalytic activity. Glycans can alter the substrate recognition, the specificity and binding affinity, as well as the turnover rates. However, there is currently no known general pattern, since glycosylation can have both stimulating and inhibiting effects on activity. Thus, a comparative analysis of individual cases with sufficient enzyme kinetic and structural data is a first approach to describe mechanistic principles that govern the effects of glycosylation on the function of proteases. The understanding of glycan functions becomes highly significant in proteomic and glycomic studies, which demonstrated that cancer-associated proteases, such as kallikrein-related peptidase 3, exhibit strongly altered glycosylation patterns in pathological cases. Such findings can contribute to a variety of future biomedical applications. Among the many pathologic events observed in the progression of AD, changes in amyloid β peptide (Aβ) metabolism proceed fastest, and precede clinical symptoms. BACE1 (β-secretase 1) catalyzes the initial cleavage of the amyloid precursor protein to generate Aβ. Therefore inhibition of BACE1 activity could block one of the earliest pathologic events in AD. However, therapeutic BACE1 inhibition to block Aβ production may need to be balanced with possible effects that might result from diminished physiologic functions BACE1, in particular processing of substrates involved in neuronal function of the brain and periphery. Potentials for beneficial or consequential effects resulting from pharmacologic inhibition of BACE1 are reviewed in context of ongoing clinical trials testing the effect of BACE1 candidate inhibitor drugs in AD populations. We demonstrate that a reduction in Reelin expression results in enhanced amyloidogenic APP processing, as indicated by the precocious production of amyloid-␤ peptides, the significant increase in number and size of amyloid-␤ plaques, as well as age-related aggravation of plaque pathology in double mutant compared with single AD mutant mice of both sexes. Numerous amyloid-␤ plaques accumulate in the hippocampal formation and neocortex of double mutants, precisely in layers with strongest Reelin expression and highest accumulation of Reelin plaques in aged wild-type mice. Moreover, concentric accumulations of phosphorylated Tau-positive neurons around amyloid-␤ plaques were evident in 15-month-old double versus single mutant mice.

We found two highly expressed isoforms, anionic and cationic trypsinogen. When compared to human cationic trypsinogen (PRSS1), ferret anionic trypsinogen autoactivated only in the presence of high calcium concentrations but not in millimolar calcium, which prevails in the secretory pathway. Ferret cationic trypsinogen was completely defective in autoactivation under all conditions tested. However, both isoforms were readily activated by enteropeptidase and cathepsin B. We conclude that ferret trypsinogens do not autoactivate as their human paralogs and cannot be used to model the efects of trypsinogen mutations associated with human hereditary pancreatitis. Intra-pancreatic trypsinogen activation by cathepsin B can occur in ferrets, which might trigger pancreatitis even in the absence of trypsinogen autoactivation. Te digestive protease precursor trypsinogen is synthesized and secreted by the pancreas to the duodenum where it becomes activated to trypsin1. Te activation process involves limited proteolysis of the trypsinogen activation peptide by enteropeptidase, a brush-border serine protease specialized for this sole purpose. Te activation peptide is typically an eight amino-acid long N-terminal sequence, which contains a characteristic tetra-aspartate motif preceding the activation site peptide bond, which corresponds to Lys23-Ile24 in human trypsinogens.

Plasmin, and other host 15 proteases, may cleave the furin site of SARS-CoV-2 S protein and subunits of epithelial sodium channels ( 16 ENaC), resulting in an increment in virus infectivity and channel activity. As for the importance of ENaC in 17 the regulation of airway surface and alveolar fluid homeostasis, whether SARS-CoV-2 will share and 18 strengthen the cleavage network with ENaC proteins at the single-cell level is urgently worthy of consideration.

They have no common phylogenetic origin but can be found in most protist evolutionary branches. Acanthamoeba is a common FLA that can be found worldwide and is capable of infecting humans. The main disease is a life altering infection of the cornea named Acanthamoeba keratitis. Additionally, Acanthamoeba has a close relationship to bacteria. Acanthamoeba feeds on bacteria. At the same time, some bacteria have adapted to survive inside Acanthamoeba and use it as transport or protection to increase survival. When conditions are adverse, Acanthamoeba is capable of differentiating into a protective cyst.

Lacking any verified substrate its role remained ambiguous. MASP-3 was shown to compete with a key lectin pathway enzyme MASP-2 for MBL binding, and was therefore considered to be a negative complement regulator. Later, knock-out mice experiments suggested that MASP-1 and/or MASP-3 play important roles in complement pro-factor D (pro-FD) maturation. However, studies on a MASP-1/MASP-3-deficient human patient produced contradicting results. In normal resting blood unperturbed by ongoing coagulation or complement activation, factor D is present predominantly in its active form, suggesting that resting blood contains at least one pro-FD activating proteinase that is not a direct initiator of coagulation or complement activation. We have recently showed that all three MASPs can activate pro-FD in vitro. In resting blood, however, using our previously evolved MASP-1 and MASP-2 inhibitors we proved that neither MASP-1 nor MASP-2 activates pro-FD. Other plasma proteinases, particularly MASP-3, remained candidates for that function. For this study we evolved a specific MASP-3 inhibitor and unambiguously proved that activated MASP-3 is the exclusive pro-FD activator in resting blood, which demonstrates a fundamental link between the lectin and alternative pathways.
