Cristina, nunca pensé que un cuerpo tan chiquitito pudiera albergar tanto, humana y profesionalmente. A pesar de las listas de “cosas por hacer en tus ratos libres” que (aún no sé cómo) no mermaba nunca, sabes que repetiría contigo una y mil veces. No olvidaré nuestras aventuras con y sin pipeta en mano, así como tu compromiso, alegría y entusiasmo. Ha sido un placer “jefa”. Y sí, lo recuerdo, te debo un barril de cerveza, o dos. Gracias por haberme tratado con cariño y, sobretodo, haberme considerado una igual. CUAK. Chesco, tú fuiste el primero de todos que apostó por mí y te estaré eternamente agradecida, gracias a ti empecé a pensar que valía para esto. Aún recuerdo la lucha y correos electrónicos mandados para que nos hicieran entender por qué un 7,14 no era un 2 en la escala del 0-4, mi gratitud por acompañarme en esa y tantas otras “guerras burocráticas”. Ejemplo de “pasito a pasito” pero lo que bien hecho está, bien parece. Gracias por intentar siempre facilitar mi tarea y sobre todo por aceptar aquello de “dos mujeres contra un hombre” que a veces te ha dejado en desventaja. A las Dras. This study aims to investigate whether the genetic variation in NPC1 is associated with risk of coronary heart disease (CHD) and to detect whether NPC1 might interact with smoking on the risk of CHD. Methods: We performed a case-control study, including 873 patients with coronary heart disease (CHD) and 864 subjects without CHD as control. Polymorphisms of NPC1 gene were genotyped by polymerase chain reaction (PCR) -restriction fragment length polymorphism (RFLP). Results: A tag-SNP rs1805081 (+644A > G) in NPC1 was identified. The G allele of the +644 locus showed reduced risk of CHD than wild-type genotype in Chinese population (recessive model GG vs. AG+AA: odds ratio [OR] 0.647, 95% CI 0.428 to 0.980, P = 0.039; additive model GG vs. AG vs. AA: OR 0.847, 95% CI 0.718 to 0.998, P = 0.0471). Moreover in smokers, the G-allele carriers had reduced risk of CHD compared with A-allele carries (OR 0.552, 95% CI 0.311 to 0.979, P = 0.0421). Conclusions: The results of the present study suggest that NPC1 variants seem to be contributors to coronary heart disease occurrence in Chinese population.

The increased amount of pituitary TSH was caused by reduced expression of type 2 iodothyronine deiodinase (Dio2), a gene that is required for T4-mediated negative feedback regulation of TSH expression. These data establish a molecular mechanism that accounts for the regulation of energy expenditure and the development of obesity by the JNK signaling pathway. The serpin peptidase inhibitor SERPINB5 is a tumour-suppressor gene that promotes the development of various cancers in humans. However, whether SERPINB5 gene variants play a role in HCC susceptibility remains unknown. In this study, we genotyped 6 SNPs of the SERPINB5 gene in an independent cohort from a replicate population comprising 302 cases and 590 controls. Additionally, patients who had at least one rs2289520 C allele in SERPINB5 tended to exhibit better liver function than patients with genotype GG (Child-Pugh grade A vs. B or C; P = 0.047). Next, haplotype blocks were reconstructed according to the linkage disequilibrium structure of the SERPINB5 gene. A haplotype “C-C-C” (rs17071138 + rs3744941 + rs8089204) in SERPINB5-correlated promoter showed a significant association with an increased HCC risk (AOR = 1.450; P = 0.031). Haplotypes “T-C-A” and “C-C-C” (rs2289519 + rs2289520 + rs1455555) located in the SERPINB5 coding region had a decreased (AOR = 0.744; P = 0.031) and increased (AOR = 1.981; P = 0.001) HCC risk, respectively. Finally, an additional integrated in silico analysis confirmed that these SNPs affectedSERPINB5 expression and protein stability, which significantly correlated with tumour expression and subsequently with tumour development and aggressiveness.

The single marker and haplotype analyses were performed by Family-based Association Tests (FBAT). Three SNPs had P-values o0.05 in at least one inherited model survived with the correction. Several haplotypes composed of these SNPs were also associated with MR. The in silico analyses identified that one of the SNPs, rs1388378, may be a functional SNP. However, further in vitro studies of this SNP should be considered in elucidating its effect on gene expression and the possible role in MR susceptibility. Although this method was previously used to analyze one dataset at a time; but now, a vast amount of mouse phenotypic data is now publicly available, which could lead to many more genetic discoveries. Results: HBCGM and a whole genome SNP map covering 53 inbred strains was used to analyze 8462 publicly available datasets of biomedical responses (1.52M individual datapoints) measured in panels of inbred mouse strains. As proof of concept, causative genetic factors affecting susceptibility for eye, metabolic and infectious diseases were identified when structured automated methods were used to analyze the output.
