Ce travail de thèse a été réalisé au sein du laboratoire des Sciences Fondamentales et aux laboratoires du département de biologie (Université Amar Telidji, Laghouat). De plus, plusieurs séjours scientifiques effectués au sein de l’Equipe des Mycotoxines de Fusarium, de l’Unité Mycologie et Sécurité des Aliments (INRA de Bordeaux-Aquitaine, France) nous ont permis d’entreprendre des travaux fructueux. Résumé Les extraits de plantes sont connus depuis longtemps comme agents antifongiques. Cependant, leur utilisation comme préservateurs des céréales a été rarement rapportée. Ce travail a pour objectif l'étude de la variabilité chimique, l'activité antifongique ainsi que l’activité antimycotoxinogène des huiles essentielles de Rhanterium adpressum, vis-à-vis de quatre champignons mycotoxinogènes du genre Fusarium. L’échantillonnage a été fait dans quatre localités différentes (échantillonnage dans l’espace) et chaque trois mois, pendant trois ans (échantillonnage dans le temps). Les huiles essentielles des feuilles et des fleurs ont été obtenues, séparément, par hydrodistillation. L’analyse (CG/SM) a révélé trente-six constituants chimiques, au total, répartis en quatre classes de composés chimiques ; avec des pourcentages variables, selon le mois d'extraction. Dans ces huiles essentielles, les hydrocarbures monoterpéniques forment la principale classe et, dans les échantillons collectés le mois de mai, les composés oxygénés présentent les pourcentages les plus élevés. L’ACP et l’AHC ont mis en évidence trois classes chimiques dans l’ensemble des échantillons d’HE. Le pouvoir antifongique des HE des feuilles et des fleurs a été évalué par la méthode de contact direct, en milieux solide et liquide. Cependant, L'effet de ces HE sur le la production de trichothécènes de type B (TCTB) a été évaluée, en milieu liquide, par UPLC. L’activité antifongique et antimycotoxinogène des composés antifongiques bioactifs volatils (α-Pinène, β-Myrcène, Limonène, Géraniol et Linalool) ont été également évaluées sur les deux types de milieux et comparés à l’activité de l’HE. Les résultats des activités ont montré que l'extrait des fleurs présente une activité antifongique forte ; avec des concentrations intéressantes de CI50 et CMI, sur les deux milieux. Cette activité pourrait être attribuée à leur composition, relativement élevée, de monoterpènes oxygénés. De plus, d’un effet inhibiteur significatif, des deux extraits d’HE, sur la production de TCTB, l’inhibition était de 66 à 97 et 76 à 100 % de FX pour 3-ADON et 15-ADON, respectivement.

Recent attention on medicinal plants emanates from their long historical utilisation in folk medicine as well as their prophylactic properties. However, there is a dearth of scientific data on the efficacy and stability of the bioactive chemical constituents in medicinal plants after prolonged storage. This is a frequent problem in African Traditional Medicine. Methods: The phytochemical, antioxidant and acetylcholinesterase-inhibitory properties of 21 medicinal plants were evaluated after long-term storage of 12 or 16 years using standard in vitro methods in comparison to freshly harvested materials. Results: The total phenolic content of Artemisia afra, Clausena anisata, Cussonia spicata, Leonotis intermedia and Spirostachys africana were significantly higher in stored compared to fresh materials. The flavonoid content were also significantly higher in stored A. afra, C. anisata, C. spicata, L. intermedia, Olea europea and Tetradenia riparia materials. With the exception of Ekebergia capensis and L. intermedia, there were no significant differences between the antioxidant activities of stored and fresh plant materials as measured in the β-carotene-linoleic acid model system. Similarly, the EC50 values based on the 2,2-diphenyl-1-picrylhydrazyl (DPPH) free radical scavenging assay were generally lower for stored than fresh material. Percentage inhibition of acetylcholinesterase was generally similar for both stored and fresh plant material.

This enzyme is further implicated in the involvement of melanin in malignant melanoma, the most life- threatening skin tumors. Although, synthetic products were found effective to combat this menace, nevertheless, overtime detrimental effect on human skin is a challenge. Investigation of natural tyrosinase inhibitors from methanol extracts of medicinal plants of Lamiaceae family using L-tyrosine as substrate on three different complementary assays (TLC bio-autography, spectrophotometry and cyclic voltammetry) was carried out accordingly. The result indicated Salvia chamelaeagnea, Salvia dolomitica, Plectranthus ecklonii, Plectranthus namaensis, and Plectranthus zuluensis, with significant zone of inhibition against tyrosinase on TLC bio-autography, spectrophotometry result showed that extracts of Plectranthus ecklonii (IC50 = 21.58 휇g/mL), Plectranthus zuluensis (IC50 = 23.99 휇g/mL), Plectranthus madagascariensis (IC50 = 23.99 휇g/mL) and Salvia lanceolata (IC50 = 28.83) demonstrated good anti-tyrosinase activity when compared with kojic acid (IC50 = 3.607 μg/mL). On the other hand, cyclic voltammetry are in consonant with above results thereby supported the nomination of some of the extracts as strong anti-tyrosinase agents.

Their ability to produce aromatic volatile oils has made them valuable sources of materials in the cosmetic, culinary, and pharmaceutical industries. A thorough account of the taxonomic diversity, chemistry and ethnobotany is lacking for southern African Lamiaceae, which feature some of the region’s most notable medicinal and edible plant species. We provide a comprehensive insight into the Lamiaceae flora of southern Africa, comprising 297 species in 42 genera, 105 of which are endemic to the subcontinent. We further explore the medicinal and traditional uses, where all genera with documented uses are covered for the region. A broad review of the chemistry of southern African Lamiaceae is presented, noting that only 101 species (34%) have been investigated chemically (either their volatile oils or phytochemical characterization of secondary metabolites), thus presenting many and varied opportunities for further studies. Recently, a study showed that T. riparia essential oil (TrEO) obtained in summer has antileishmanial effects, although these results could be influenced by seasonal variation. This study evaluated the activity of the TrEO obtained in different seasons against Leishmania (Leishmania) amazonensis, in vitro and in vivo. The compounds in the TrEO were analysed by gas chromatography-mass spectrometry; terpenoids were present and oxygenated sesquiterpenes were the majority compounds (55.28%). The cytotoxicity and nitric oxide (NO) production were also tested after TrEO treatment. The TrEO from all seasons showed a 50% growth inhibitory concentration for promastigotes of about 15 ng/mL; at 30 ng/mL and 3 ng/mL, the TrEO reduced intracellular amastigote infection, independently of season. The TrEO from plants harvested in summer had the highest 50% cytotoxic concentration, 1,476 ng/mL for J774.A1 macrophages, and in spring (90.94 ng/mL) for murine macrophages.

Metal nanoparticles have important applications in medicine and agriculture. The leaves of Tetradenia riparia (Iboza), an important medicinal species in South Africa, are reported to contain various terpenoids and pyrones which can be used in the reduction of silver nitrate (AgNO3) to nano-silver particles. Materials and Methods: Fresh leaves and stems of Iboza were oven-dried, crushed, extracted in water and methanol and filtered followed by incubation with AgNO3. Synthesized nano-silver particles were characterised using ultraviolet visible (UV-Vis) spectroscopy, scanning electron microscopy (SEM), energy dispersive X-ray (EDX) spectroscopy and Fourier transform infrared (FTIR) spectroscopy. Results: The bio-reduction of metal ion to base metal was rapidly conducted in a single step and at room temperature and pressure. UV-Vis spectra showed the characteristic surface plasmon resonance band of the synthesized nano-silver particles at 410 nm for all extracts. SEM analysis revealed predominantly aggregated spherically-shaped nano-silver particles with a size range of 20-50 nm and an average diameter of 26 nm.
