Janet Oesterling Novozymes North America, Inc. 77 Perry Chapel Church Road Franklinton, NC 27525 Re: GRAS Notice No. GRN 000675 Dear Ms. Oesterling: The Food and Drug Administration (FDA, we) completed our evaluation of GRN 000675. We received the notice, dated October 10, 2016, that you submitted in accordance with the agency’s proposed regulation, proposed 21 CFR 170.36 (62 FR 18938; April 17, 1997; Substances Generally Recognized as Safe (GRAS); the GRAS proposal) on July 17, 2016, and filed it on October 14, 2016. We received amendments containing clarifying information on February, 22, 2017 and March 09, 2017. FDA published the GRAS final rule on August 17, 2016 (81 FR 54960), with an effective date of October 17, 2016. As GRN 000675 was pending on the effective date of the GRAS final rule, we requested some additional information consistent with the format and requirements of the final rule. We received an amendment responding to this request on October 24, 2016. The subject of the notice is xylanase enzyme preparation produced by Trichoderma reesei carrying a xylanase gene from Talaromyces leycettanus (xylanase enzyme preparation). The notice informs us of the view of Novozymes North America, Inc. (Novozymes) that xylanase enzyme preparation is GRAS, through scientific procedures, for use as an enzyme at levels up to 48.33 mg Total Organic Solids (TOS) per kg raw material in brewing, cereal beverage processing, baking, and processing of cereal grains such as corn, wheat, barley, and oats. Commercial enzyme preparations that are used in food processing typically contain an enzyme component that catalyzes the chemical reaction as well as substances used as stabilizers, preservatives, or diluents. -

Abstract Prevotella copri DSM18205 is a bacterium, classified under Bacteroidetes that can be found in the human gastrointestinal tract (GIT). The role of P. copri in the GIT is unclear, and elevated numbers of the microbe have been reported both in dietary fiber- induced improvement in glucose metabolism but also in conjunction with certain inflammatory conditions. -

Mutants of ArthβDG: D207A and E517Q were designed to determine the significance of specific residues and to enable formation of complexes with lactulose and sucrose and to shed light onto the structural basis of the transglycosylation reaction. The catalytic assays proved loss of function mutation E517 into glutamine and a significant drop of activity for mutation of D207 into alanine. Solving crystal structures of two new mutants, and new complex structures of previously presented mutant E441Q enables description of introduced changes within active site of enzyme and determining the importance of mutated residues for active site size and character. -

GCase deficiency results in the accumulation of its substrate glucosylceramide (GlcCer) within the lysosomes leading to various severities of hepatosplenomegaly, bone disease and neurodegeneration. For most forms of Gaucher disease, the mutations in the GBA gene cause the enzyme to misfold but retain catalytic activity. However, the misfolded mutant enzyme is recognized and degraded by the endoplasmic reticulum-associated degradation (ERAD) pathway prior to delivery into the lysosome. Symptoms begin to show in patients when the function of the defective enzyme drops below 10-20% residual enzyme activity. There are currently three therapeutic approaches to treat Gaucher disease: enzyme replacement therapy (ERT), substrate reduction therapy (SRT), and a relatively recent addition, enzyme enhancement therapy (EET) through the use of pharmacological chaperones. Many pharmacological chaperones are competitive inhibitors that are capable of enhancing lysosomal GCase activity by stabilizing the folded conformation of GCase enabling it to bypass the ERAD pathway. -

LCT gene lactase Normal Function The LCT gene provides instructions for making an enzyme called lactase. This enzyme helps to digest lactose, a sugar found in milk and other dairy products. Lactase is produced by cells that line the walls of the small intestine. These cells, called intestinal epithelial cells, have finger-like projections called microvilli that absorb nutrients from food as it passes through the intestine so they can be absorbed into the bloodstream. Based on their appearance, groups of these microvilli are known collectively as the brush border. Lactase functions at the brush border to break down lactose into smaller sugars called glucose and galactose for absorption. Health Conditions Related to Genetic Changes Lactose intolerance At least nine LCT gene mutations cause congenital lactase deficiency, also called congenital alactasia. In this disorder, infants are unable to break down lactose (lactose intolerance) in breast milk or formula. The LCT gene mutations change single protein building blocks (amino acids) in the lactase enzyme or result in an enzyme that is abnormally short. The mutations are believed to interfere with the function of the lactase enzyme, leading to undigested lactose in the small intestine and causing severe diarrhea. Lactose intolerance in adulthood is caused by gradually decreasing activity (expression) of the LCT gene after infancy, which occurs in most humans. Pompe’s Disease and the Effects ofAlpha-Glucosidase Deficiency Aaron Richler Overview: The energy that the body needs in order to function is obtained from carbohydrates that we get through our diet. These carbohydrates are monosacharides, disaccharides and polysacharides. The polysaccharides and disaccharides are hydrolyzed to monosaccharide’s such glucose (which comprises roughly 80%) fructose and galactose. Most cells convert the fructose and galactose to glucose. The body can use the glucose or store it. If energy is needed, glucose can be oxidized through the many reactions of glycolysis which gives a net production of 2 ATP and 2 NADH from one molecule of glucose. In the presence of oxygen the product of glycolysis pyruvic acid will be decarboxilated to Acetyl Coenzyme A (Acetyl COA) in the mitochondrial matrix forming carbon dioxide and 2 NADH in the process.
