Although the mechanisms behind the pathogenic effects of LRRK2 mutations are still not clear, data emerging from in vitro and in vivo models suggests roles in regulating neuronal polarity, neurotransmission, membrane and cytoskeletal dynamics and protein degradation. We created mice lacking exon 41 that encodes the activation hinge of the kinase domain of LRRK2. We have performed a comprehensive analysis of these mice up to 20 months of age, including evaluation of dopamine storage, release, uptake and synthesis, behavioral testing, dendritic spine and proliferation/neurogenesis analysis. Our results show that the dopaminergic system was not functionally comprised in LRRK2 knockout mice. However, LRRK2 knockout mice displayed abnormal exploratory activity in the open-field test. Moreover, LRRK2 knockout mice stayed longer than their wild type littermates on the accelerated rod during rotarod testing. Finally, we confirm that loss of LRRK2 caused degeneration in the kidney, accompanied by a progressive enhancement of autophagic activity and accumulation of autofluorescent material, but without evidence of biphasic changes. These include the use of vanillymandelic acid and homovanillic acid in screening for neuroblastoma in newborn infants, AFP in screening for hepatocellular cancer in high‐risk subjects, CA 125 in combination with transvaginal ultrasound (TVU) in screening for ovarian cancer, PSA in screening for prostate cancer and fecal occult blood testing (FOBT) in screening for CRC. Of these markers, only the use of FOBT in screening for CRC has been shown to reduce mortality from cancer. Large randomized prospective trials are currently in progress aimed at evaluating the potential value of PSA screening in reducing mortality from prostate cancer and CA 125 in combination with TVU in reducing mortality form ovarian cancer.

This traditional medicinal plant has been used for treating rheumatoid arthritis and lung disorders as well as for its anti-inflammation and anticancer activities. This study aimed to investigate the anti-proliferative and apoptotic-inducing activities of HCUA in oral cancer cells. HCUA exhibited anti-proliferative activity in oral cancer cell lines (Ca9-22 and SAS cells), but not in normal oral fibroblasts. The inhibitory concentration of HCUA that resulted in 50% viability was 24.0 µM and 17.8 µM for Ca9-22 and SAS cells, respectively. Moreover, HCUA increased the number of cells in the sub-G1 arrest phase and apoptosis in a concentration- dependent manner in both oral cancer cell lines, but not in normal oral fibroblasts.
