Strains were identified by ITS rDNA sequencing, and Exophiala cancerae species borderlines were established by multilocus sequencing and AFLP analysis. Fonsecaea brasiliensis Virulence Fonsecaea brasiliensis proved to be closely related to the pathogenic species Cladophia- lophora devriesii which originally was isolated from a systemic infection in a human patient. The virulence of F. brasiliensis is lower than that of E. cancerae, as established by artificial inoculation of mangrove crabs. ß 2012 Elsevier B.V. All rights reserved. 1. The ) main signs of disease include increasingly weak motor control, particularly of pereiopods and chelae, causing lethargy and poor balance, followed by the death of the BT2, TEF1 affected crab. Tetany of the claws is also observed in many crabs with other signs of the disease. Despite the importance of the crab species for low- income populations in the coastal regions of Brazil, only JF747063, JN112443, – JF747064, JN112444, – JN173784, JN368478, JN187943 JN173785, JN368479, JN187944 JN173787, JN368481, – JN173788, JN368480, JN187945 few scientific studies addressing LCD are available (e.g. Pie et al., 2011; Ore´lis-Ribeiro et al., 2011; Boeger et al., 2005). All evidence from a variety of sources (light and electron microscopy, behavioral and experimental tests, and molecular phylogenetics) indicate that LCD signs were associated with the presence of a black yeast-like fungus, Exophiala cancerae (de Hoog et al., 2011). Exophiala and Fonsecaea are members of the ascomy- cete order Chaetothyriales, comprising the black yeasts and allies, which are regularly encountered as causative agents Grande city, mangrove Goiana city, mangrove Grande city, mangrove Grande city, mangrove Grande city, mangrove Grande city, mangrove of disorders in humans and animals (Nyaoke et al., 2009; de Hoog et al., 2011; Kano et al., 2000). In humans, infections by chaetothyrialean fungi range from commensalism or mild cutaneous infection to fatal neurotropism with severe mutilation. All infections are preponderantly found in patients without immune disorder or underlying metabolic disease. Outside humans, particularly cold-blooded water- borne vertebrates are susceptible to a diversity of Exophiala species (de Hoog et al., 2011). Some of these are host-specific to particular fish taxa (Nyaoke et al., 2009). As virulence factors the ability to assimilate alkylbenzenes, present in sweat and nervous tissues of mammals and in the toxic skin of amphibians, has been suggested (Prenafeta-Boldu´ et al., Toad liver Israel JF747065, JN112445, – 2006; de Hoog et al., 2011). Many reports of epizootics from the older literature clearly demonstrate that black yeast infection is a relatively common phenomenon in cold-blood vertebrates (e.g. Fijan, 1969; Richards et al., 1978; Reuter et al., 2003). Recent studies have shown that different pathogenic species are involved (e.g. Manharth et al., 2005; Nyaoke et al., 2009; de Hoog et al., 2011) which are morpholo- gically very similar. A total of 313 melanized fungi were isolated at fve locations in Guangzhou, China. Internal transcribed spacer (rDNA ITS) sequencing showed a preponderance of 27 species belonging to 10 genera; 64.22% (n = 201) were known as human opportunists in the orders Chaetothyriales and Venturiales, potentially causing cutaneous and sometimes deep infections. Knufa epidermidis was the most frequently encountered species in bathrooms (n = 26), while in kitchens Ochroconis musae (n = 14), Phialophora oxyspora (n = 12) and P. europaea (n = 10) were prevalent. Since the majority of species isolated are common agents of cutaneous infections and are rarely encountered in the natural environment, it is hypothesized that indoor facilities explain the previously enigmatic sources of infection by these organisms. Black yeast-like and other melanized fungi are frequently isolated from clinical specimens and are known as etiologic agents of a gamut of opportunistic infections, but for many species their natural habitat is unknown and hence the source and route of transmission remain enigmatic. Te majority of clinically relevant black yeast-like fungi belong to the order Chaetothyriales, while some belong to the Venturiales. Propagules are mostly hydro- philic1 and reluctantly dispersed by air, infections mostly being of traumatic origin.

Its traits, potentially linked to pathogenesis, have never been system- atically studied. Here, we examine 49 A. melanogenum strains for growth at 37 ◦C, siderophore production, hemolytic activity, and assimilation of hydrocarbons and human neurotransmitters and report within-species variability. All but one strain grew at 37 ◦C. All strains produced siderophores and showed some hemolytic activity. The largest differences between strains were observed in the assimilation of hydrocarbons and human neurotransmitters. Sybren de Hoog2,7* & Vania Aparecida Vicente1,2* Melanized fungi and black yeasts in the family Herpotrichiellaceae (order Chaetothyriales) are important agents of human and animal infectious diseases such as chromoblastomycosis and phaeohyphomycosis. The oligotrophic nature of these fungi enables them to survive in adverse environments where common saprobes are absent. Due to their slow growth, they lose competition with common saprobes, and therefore isolation studies yielded low frequencies of clinically relevant species in environmental habitats from which humans are thought to be infected. This problem can be solved with metagenomic techniques which allow recognition of microorganisms independent from culture. The present study aimed to identify species of the family Herpotrichiellaceae that are known to occur in Brazil by the use of molecular markers to screen public environmental metagenomic datasets from Brazil available in the Sequence Read Archive (SRA). Species characterization was performed with the BLAST comparison of previously described barcodes and padlock probe sequences. A total of 18,329 sequences was collected comprising the genera Cladophialophora, Exophiala, Fonsecaea, Rhinocladiella and Veronaea, with a focus on species related to the chromoblastomycosis. The data obtained in this study demonstrated presence of these opportunists in the investigated datasets. The used techniques contribute to our understanding of environmental occurrence and epidemiology of black fungi.

The species are excluded from the genus Cladosporium and are classified in the genus Cladophialophora. Taeniolella boppii is also transferred to this genus. Cladosporium bantianum (= Xylohypha emmonsii) and C. trichoides are considered conspecific and are now referred to as Cladophialophora bantiana. Meso-erythritol, L-arabinitol, ethanol and growth at 40 °C are found to be the most useful criteria for species distinction. The species Cladosporium carrionii is found to be heterogeneous. The anamorph of the saprophytic ascomycete Capronia pilosella is morphologically similar to an authentic strain of Cladosporium carrionii, but physio- logically distinct.
