Camilla Di Marcantonio Supervisor Co-Supervisor Prof. Agostina Chiavola Prof. Maria Rosaria Boni Abstract Abstract Organic MicroPollutants (OMPs) – also called Emerging Contaminants or Contaminants of Emerging Concern – include a wide number of chemicals belonging to different classes, e.g. pharmaceuticals and personal care products (PPCPs), drugs of abuse and their metabolites, steroids and hormones, endocrine- disrupting compounds, surfactants, perfluorinated compounds, phosphoric ester flame retardants, industrial additives and agents, siloxanes, artificial sweeteners, and gasoline additives (Barbosa et al., 2016; Bletsou et al., 2015; Chiavola et al., 2019). In the last two decades, increasing attention has been dedicated to OMPs, as a matter of high risk for public health and environment. (Naidu et al., 2016; Rodriguez-Narvaez et al., 2017; Thomaidi et al., 2016; Vilardi et al., 2017). OMPs are characterized by low environmental concentrations (about ng/L or µg/L), high toxicity, very low biodegradability and resistance to degradation and to conventional treatments. Consequently, they tend to be bioaccumulated in aquatic environments, and to enter the food chain through agriculture products and drinking water (Clarke and Smith, 2011). Measurement of OMPs in the aquatic medium became possible only in the last 20 years, thanks to the improvement of sensitivity and accuracy of the analytical methods; among the different methods, liquid chromatography coupled with high-resolution tandem mass spectrometry (LC-HRMS/MS) is increasingly applied for the analysis of some known and unknown emerging contaminants in water.

Introduction The term amphetamine type stimulants (ATS) refer to a range of drugs mostly derived from the phenethylamines. Amphetamines are central nervous system (CNS) stimulants that were first synthesised more than a century ago for medical applications, but are currently mostly found on illicit drug markets. Globally, ATS use occurs in a range of contexts, and for a variety of purposes. Both recreational and occupational reasons may determine initial use; and use may occur in public settings (e.g., nightclubs), private parties, work environments or as sex aids. In this respect, ATS have demonstrated their attractiveness in a very wide range of situations that seem to differ across countries. ATS are used recreationally to experience the drug’s effects of increased sociability, loss of inhibitions, a sense of escape, or to enhance sexual encounters30‐34. In many high income countries, users typically have a history of other drug use and may use other substances in combination with M/A35‐38. M/A is sometimes used in occupational settings to sustain long work hours and to increase energy and productivity. Examples of this include use by jade‐mine workers in Myanmar, sex workers in Cambodia (Chouvy & Meissonnier, 2004), truck drivers73 159‐161 and even pilots in armed forces, where it has been provided by Governments (Emonson & Vanderbeek, 1995). Amphetamine (AMPT) and methamphetamine (MAMPT) both increase the release of dopamine, noradrenalin, adrenaline and serotonin (Seiden, Sobol, & Ricaurte, 1993; World Health Organization, 2004), stimulate the central nervous system, and have a range of effects including increased energy, feelings of euphoria, decreased appetite, elevated blood pressure and increased heart rate.

It is the common structural element of all tropane alkaloids (Lounasmaa and Tamminen, 1993). Tropane does not occur naturally in free form rather it is found as part of esters in plant species. Esters of tropane are generally secondary metabolites of these plants. 2.4 II.2.4 Cocaine and its metabolites by Kenichi Takekawa Introduction Cocaine is an alkaloid being contained in the leaves of Erythroxylon coca Lam. or Erythroxylon novogranatense (Morris) Hieronymus of Erythroxylaceae plants. Th e compound is being used as a local anaesthetic. It also shows stimulating action on the central nervous system; when it is used continuously, psychic dependence on its use appears, resulting in its chronic toxicosis [1]. Cocaine is being abused worldwide; its abuse is most serious in the north and south American continents. Fortunately, in Japan, its abuse is not so many; but it cannot be overlooked. In this chapter, the methods of extractiona and GC/MS analysis for cocaine and its main metabolites, such as ecgonine methyl ester ( EME) and benzoylecgonine ( BE), in urine and blood, are presented; a fatal cocaine poisoning case, which the author experienced, is also demonstrated. Chemical synthesis of the metabolite standards [2] • Cocaine hydrochloride (Shionogi & Co., Ltd., Osaka, Japan; Takeda Chem. Ind. Ltd., Osaka, Japan; or Sigma, St. Louis, MO, USA) is dissolved in 0.2 M hydrochloric acid aqueous solu- tion and heated at 90 °C with refl uxing (gentle mixing) for 24 h to produce ecgonine. Th e solution is extracted with ethyl ether to remove benzoic acid. Th e dried residue of ecgonine hydrochloride is dissolved in 10 % hydrochloric acid methanolic solution for its methyla- tion to synthesize EME [2]. • Aqueous solution of cocaine hydrochloride is carefully adjusted to pH 7 by repeated addi- tion of 0.1 M NaOH solution with refl uxing for 3 days; precipitated BE can be obtained.
