[1] Muscarinic acetylcholine receptor antagonism decreases sign-tracking behavior in rats Christopher J. Fitzpatrick1* and Jonathan D. Morrow2 1Neuroscience Graduate Program and 2Department of Psychiatry, University of Michigan, Ann Arbor, MI *[email protected] Muscarinic acetylcholine receptors (mAChRs) facilitate dopamine (DA) transmission within the mesolimbic reward system. Our laboratory uses a Pavlovian conditioned approach (PCA) paradigm to investigate this system and addiction vulnerability in rats. Briefly, rats are repeatedly presented with a conditioned stimulus (CS; a lever) followed by the response-independent presentation of an unconditioned stimulus (US; a food pellet). Over multiple training sessions, two phenotypes can develop: sign-tracking (CS-directed behavior) and goal-tracking (US-directed behavior). Sign-trackers (STs) attribute incentive- motivational value to reward-related cues and are more vulnerable to addiction-like behaviors, such as cue-induced reinstatement of drug-seeking. In addition, sign-tracking behavior requires DA transmission within the mesolimbic reward system, therefore we hypothesized that mAChR antagonism would attenuate
