Endothelin is synthesized in vascular endothelial and smooth muscle cells, as well as in neural, renal, pulmonic, and inflammatory cells. It acts through a seven transmembrane endothelin receptor A (ETA) and endothelin receptor B (ETB) receptors belongs to G protein-coupled rhodopsin-type receptor superfamily. This peptide involved in pathogenesis of cardiovascular disorder like (heart failure, arterial hypertension, myocardial infraction and atherosclerosis), renal failure, pulmonary arterial hypertension and it also involved in pathogenesis of cancer. Potentially endothelin receptor antagonist helps the treatment of the above disorder. Currently, there are a lot of trails both per-clinical and clinical on endothelin antagonist for various cardiovascular, pulmonary and cancer disorder. Some are approved by FAD for the treatment. CHLORZOXAZONE- chlorzoxazone tablet DIRECT RX ---------- CHLORZOXAZONE DESCRIPTION SECTION Chlorzoxazone USP is a centrally acting skeletal muscle relaxant, available as tablets of 500 mg for oral administration. Its chemical name is 5-Chloro-2-benzoxazolinone, and its structural formula is: C7H4CINO2 MW 169.57 Chlorzoxazone USP is a white or practically white, practically odorless, crystalline powder. Chlorzoxazone is slightly soluble in water; sparingly soluble in alcohol, in isopropyl alcohol, and in methanol; soluble in solutions of alkali hydroxides and ammonia. Chlorzoxazone tablets contain the inactive ingredients Docusate Sodium, Lactose (hydrous), Magnesium Stearate, Microcrystalline Cellulose, Pregelatinized Starch, Sodium Benzoate, and Sodium Starch Glycolate. CLINICAL PHARMACOLOGY SECTION Chlorzoxazone is a centrally-acting agent for painful musculoskeletal conditions. Data available from animal experiments as well as human study indicate that chlorzoxazone acts primarily at the level of the spinal cord and subcortical areas of the brain where it inhibits multisynaptic reflex arcs involved in producing and maintaining skeletal muscle spasm of varied etiology. The clinical result is a reduction of the skeletal muscle spasm with relief of pain and increased mobility of the involved muscles. Blood levels of chlorzoxazone can be detected in people during the first 30 minutes and peak levels may be reached, in the majority of the subjects, in about 1 to 2 hours after oral administration of chlorzoxazone. Chlorzoxazone is rapidly metabolized and is excreted in the urine, primarily in a conjugated form as the glucuronide. Less than one percent of a dose of chlorzoxazone is excreted unchanged in the urine in 24 hours. INDICATIONS & USAGE SECTION Chlorzoxazone is indicated as an adjunct to rest, physical therapy, and other measures for the relief of discomfort associated with acute, painful musculoskeletal conditions. -

It seriously affects the quality of life of epileptic patients, and nearly 30% of individuals are refractory to treatment of antiseizure drugs. Therefore, there is an urgent need to develop new drugs to manage and control refractory epilepsy. Cannabinoid ligands, including selective cannabinoid receptor subtype (CB1 or CB2 receptor) ligands and non- selective cannabinoid (synthetic and endogenous) ligands, may serve as novel candidates for this need. Cannabinoid appears to regulate seizure activity in the brain through the activation of CB1 and CB2 cannabinoid receptors (CB1R and CB2R). An abundant series of cannabinoid analogues have been tested in various animal models, including the rat pilocarpine model of acquired epilepsy, a pentylenetetrazol model of myoclonic seizures in mice, and a penicillin-induced model of epileptiform activity in the rats. The accumulating lines of evidence show that cannabinoid ligands exhibit significant benefits to control seizure activity in different epileptic models. Since the introduction of tretinoin over 40 years ago, topical retinoid products have been a mainstay of acne treatment. The retinoids are very effective in addressing multiple aspects of the acne pathology as they are comedolytic and anti-inflammatory, and do not contribute to antibiotic resistance or microbiome disturbance that can be associated with long-term antibiotic therapies that are a common alternative treatment. However, topical retinoids are associated with skin dryness, erythema and pain, and may exacerbate dermatitis or eczema. Thus, there is a clear need to target delivery of the retinoids to the pilosebaceous units to increase efficacy and minimise side effects in surrounding skin tissue. -

Oral health is an important part of our overall health. Oral health issues can cause oral pain, increased costs in healthcare, and less productivity. Recent developments in oral hygiene have led to improved outcomes for patients. Many oral health issues are still prevalent today, but the good news is that many are preventable with daily healthy habits. Healthy habits are required to maintain good oral health and many oral health issues are still prevalent today. One of the most common oral health problems is decay in the tooth also known as a cavity. The outer layer of the tooth is a tough mineral layer called the enamel. Bacteria group together on teeth to form plaques that can erode the enamel and the deeper layers of your teeth, causing cavities. Cavities are present and untreated in up to 26% up American adults. (1) If left untreated, they can lead to pain in the tooth and more severe infections known as an abscess. Another common oral health problem is gum disease. When bacteria group together on the teeth, it causes the immune system to respond and causes inflammation in the mouth. The initial inflammation is called gingivitis and makes your gums swell and bleed more easily. Untreated gingivitis can progress to a more severe gum disease called periodontitis. Periodontitis can damage the structure that holds your teeth in place and is a common cause of tooth loss. -
Public Safety Aspects of the Heroin Abuse Epidemic
A brief from June 2015 Public Safety Aspects of the Heroin Abuse Epidemic Overview Policymakers at all levels of government are attempting to address a nationwide increase in heroin abuse.1 Use of the drug has tripled over the past decade, paralleling widespread misuse of prescription opioids, and overdose deaths have increased nearly threefold in the past seven years. Available research suggests that the most effective response to the growth in heroin abuse is a combination of law enforcement to curtail trafficking and limit the emergence of new markets; alternative sentencing to divert nonviolent drug offenders from costly incarceration; treatment to reduce dependency and recidivism; and prevention efforts that can help identify individuals at high risk for addiction. These strategies are most effective when tailored to the specific nature of the heroin problem in a given community. Research further suggests that increasing criminal penalties for heroin-related offenses is unlikely to reverse the growth in heroin use and tends to have a poor return on investment. Summary Povidone Iodine (Iodine with water soluble polymer Polyvinylpyrolidone) or PVP-I is a proven and time trusted antiseptic agent having best possible (99.99%) virucidal effect in it‟s only 0.23% concentration, against all viruses including SARS-Co, MERS-CoV; even in SARS-COV-2 due to it‟s nonspecific mode of action for virus killing and having no resistance [1,2]. Corona virus is transmitted by/via respiratory droplets or aerosol, produced from sneezing or coughing of infected persons to healthy individual through mouth and nose mainly [5, 6]. The routes of entry of coronavirus in human body are mouth, nose and eye. PVP-I products for gargling the throat and spraying or washing the nose may have a preventive effect on COVID-19 and if it is proved in this study following human trial, this will be a landmark research in COVID-19 pandemic. In line of this, PVP-I containing oro-nasal spray, proposed Bangasafe, which should be regarded as PONS (Povidone Iodine oro-nasal spray) in this protocol, has been developed and proposed to use against corona virus disease.
