Directions provided in order from northern Illinois at the Iowa border, to the southern tip of Illinois.

Jo Daviess County Take U.S. Highway 20 from the Illinois state line in East Dubuque, through Galena to approximately 12 miles east of Galena to the intersection of U.S. Highway 20 and . Go south (turn right) on Illinois Route 84, through Hanover, to the southern border of Jo Daviess County.

Carroll County The route continues from the northern border of Carroll County on Illinois Route 84 through Savanna, through Thomson, to the southern border of Carroll County.

Whiteside County The route continues from the northern border of Whiteside County on Illinois Route 84 through Fulton, past East Clinton, and through Albany to the southern border of Whiteside County.

Rock Island County The route continues from the northern border of Rock Island County on Illinois Route 84 through Cordova, Port Byron, Rapids City, and into East Moline. Turn west (right) off Illinois Route 84 (19th Street) onto Highway 92 at their intersection in East Moline. Continue through East Moline, Moline, Rock Island, and Andalusia on Highway 92. Approximately 7 miles past Andalusia, turn right onto County Road 59 (Loud Thunder Road). Follow to Illinois City and turn right to rejoin Highway 92. At the intersection of Highway 92 and County Road 11 (322nd Street W), turn south (left) onto County Road 11(322nd Street W). Follow County Road 11 to the southern border of Rock Island County.

Mercer County The route continues from the northern border of Mercer County where County Road 11 becomes County Road 14 (45th Street). Stay on this to just north of New Boston. County Road 14 will intersect Illinois Route 17. At this intersection, turn left onto Illinois Route 17. After approximately 4 miles on Illinois Route 17, the route turns south (right) onto County Road 25 (76th Street) and continues through Keithsburg to the southern border of Mercer County.

Henderson County The route continues from the northern border of Henderson County on the same road, now called County Road 3 (1350 E), to the intersection with Illinois Route 164 in Oquawka. Turn west (right) onto Illinois Route 164 in Oquawka. The route follows Route 164 through Oquawka and Gladstone to the junction of Illinois Route 164 and U.S. Route 34. Turn right at the intersection of 164 and 34. Follow U.S. Route 34 west approximately 7 miles to its intersection with Carman Road. Turn south (left) on Carman Road. The route follows Carman Road south to Lomax, where it intersects Illinois Route 96. Turn west (right) onto Illinois Route 96 at Lomax. The route continues on Illinois Route 96 through Dallas City to the southern border of Henderson County.

Hancock County The route continues from the northern border of Hancock County on Illinois Route 96 through Dallas City, past Pontoosuc, through Niota, Nauvoo, and Hamilton. In Hamilton, turn west (veer right) onto U.S. Highway 136, for 0.7 miles, to County Road 32/Warsaw Road. Turn south (left) onto County Road 32 (Warsaw Road) and continue south to Warsaw. Once in Warsaw, the route will continue west (right) on Main Street to Water Street (County Road 12). Follow County Road 12 to the southern border of Hancock County.

Adams County The route continues from County Road 12 at the northern border of Adams County and turns into County Road 7 (East 500th Street). Follow County Road 7 (East 500th Street) to the intersection of County Road 7 (East 500th Street) and North 2150th Avenue. Turn east (left) onto North 2150th Avenue. Follow County Road 7 (North 2150th Avenue) to its intersection with East 603rd Lane and turn south (right) onto County Road 7 (East 603rd Lane). The route will remain on County Road 7 until it reaches County Road 1400 North (Kochs Lane/Bluff Road) outside of Quincy. Turn left onto County Road 1400 North (Kochs Lane/Bluff Road) and follow to its intersection with U.S. Highway 24. Turn south (right) onto U.S. 24 (Southbound – 3rd Street, Northbound – 4th Street) and follow U.S. 24 through Quincy to Illinois Route 57. The route follows Illinois Route 57 south through Marblehead and Fall Creek to the southern border of Adams County.

Pike County The route continues from the northern border of Pike County along Illinois Route 57 to its intersection with Illinois Route 96. Turn right at the intersection and follow Illinois Route 96 to the intersection of Illinois Route 106 and Illinois Route 96. Turn east (left) on Illinois Route 96 and continue south past Hull, through Kinderhook, New Canton, Rockport, and Pleasant Hill to the southern border of Pike County.

Calhoun County The route continues on Illinois Route 96 at the northern border of Calhoun County. At the intersection of Illinois Route 96 and County Road 2 ( Road), turn east (left) and follow Illinois Route 96 to its intersection with in Kampsville. Take a right onto Illinois Route 100 and head south toward Hardin and to the eastern border of Calhoun County.

Greene County Turn east (left) at Hardin and continue to follow Illinois Route 100 briefly through Greene County.

Jersey County The route continues on Illinois Route 100 at the northwestern border of Jersey County past Rosedale, through Grafton, and past Elsah to the eastern border of Jersey County.

Madison County The route continues on Illinois Route 100 at the western border of Madison County. Follow Route 100 to Alton and continue onto U.S. Highway 67. Follow U.S. 67 for 0.8 of a mile and continue onto . Follow Illinois Route 143 to Wood River and head south on through Hartford. Exit left off Illinois Route 3 just north of Granite City onto Interstate 270 East. The route continues east on Interstate 270 to the intersection of Interstate 270 and . Take Exit 6A onto Illinois Route 111. The route continues on Illinois route 111 for approximately 6 miles, then exit onto Interstate 70 Westbound (St. Louis). Follow this for approximately 4 miles to the southern border of Madison County.

St. Clair County The route continues from the northern border of St. Clair County on Interstate 70 Westbound and travels to the junction with Illinois Route 3 Southbound. Take Exit 1 for I-70West/South Illinois Route 3. Continue on Illinois Route 3 through Cahokia and Dupo to the southern border of St. Clair County.

Monroe County The route continues from the northern border of Monroe County on Illinois Route 3 through Columbia and Waterloo to the southern border of Monroe County.

Randolph County The route continues from the northern border of Randolph County on Illinois Route 3 through Red Bud and Ruma, past Evansville and Ellis Grove, and through Chester and Rockwood to the southern border of Randolph County.

Jackson County The route continues from the northern border of Jackson County on Illinois Route 3 through Glenn, past the Route 149 intersection and past Grand Tower approximately 5 miles to the southern border of Jackson County.

Union County The route continues from the northern border of Union County on Illinois Route 3 through Wolf Lake, past Ware and Reynoldsville, to the southern border of Union County.

Alexander County The route continues from the northern border of Alexander County on Illinois Route 3 past McClure, through Gale, past Thebes, and through Olive Branch to the intersection of Illinois Route 3 and . At the intersection of Route 3 and Route 127, turn south (right) onto Illinois Route 3. Follow through Cache and Klondike and continue on to U.S. Highway 51. On U.S. 51, the byway continues south through Cairo to U.S. Highway 60/62. It then continues on U.S. 60/62 beside Fort Defiance Park and exits the state of Illinois.
