Jones, MD ABSTRACT Purpose: To determine the risk factors and clinical signs of Curvularia keratitis and to evaluate the management and out- come of this corneal phæohyphomycosis. Methods: We reviewed clinical and laboratory records from 1970 to 1999 to identify patients treated at our institution for culture-proven Curvularia keratitis. Descriptive statistics and regression models were used to identify variables associ- ated with the length of antifungal therapy and with visual outcome. In vitro susceptibilities were compared to the clini- cal results obtained with topical natamycin. Results: During the 30-year period, our laboratory isolated and identified Curvularia from 43 patients with keratitis, of whom 32 individuals were treated and followed up at our institute and whose data were analyzed. Trauma, usually with plants or dirt, was the risk factor in one half; and 69% occurred during the hot, humid summer months along the US Gulf Coast. Presenting signs varied from superficial, feathery infiltrates of the central cornea to suppurative ulceration of the peripheral cornea. A hypopyon was unusual, occurring in only 4 (12%) of the eyes but indicated a significantly (P = .01) increased risk of subsequent complications. The sensitivity of stained smears of corneal scrapings was 78%. Curvularia could be detected by a panfungal polymerase chain reaction. Fungi were detected on blood or chocolate agar at or before the time that growth occurred on Sabouraud agar or in brain-heart infusion in 83% of cases, although colonies appeared only on the fungal media from the remaining 4 sets of specimens. -

The first patient presented for investigation of a lesion at the right hilum on chest radiograph and a raised blood eosinophil count. A bronchogram showed complete block of the apical segmental bronchus which at operation was shown to be caused by inspissated material. The second patient was investigated because of a cough productive of plugs of sputum and irregular opacities in both upper zones on chest radiograph and a raised blood eosinophil count. This only cleared after one month on high dose oral prednisone therapy. The third patient with a previous history ofleft lingular pneumonia and bronchiectasis ofthe lingular segment ofthe left upper lobe was investigated three years later for right basal shadowing and a raised blood eosinophil count. The radio- graph cleared after one month on high dose oral prednisone treatment. The aetiological agents in these cases were dematiaceous hyphomycetes, fungi ubiquitous in nature, and also agents of plant disease. The causal fungi, Curvularia hlnata and Drechslera hawaiiensis, have on a few occasions been reported as causing human disease but in cases quite dissimilar to the three reported here. Our bioassays had shown that extracts of Paraphoma sp. VIZR 1.46, a pathogen of Cirsium arvense, are phytotoxic. In this study, two phytotoxically active metabolites were isolated from Paraphoma sp. VIZR 1.46 liquid and solid cultures and identified as curvulin and phaeosphaeride A, respectively. The latter is reported also for the first time as a fungal phytotoxic product with potential herbicidal activity. Both metabolites were assayed for phytotoxic, antimicrobial and zootoxic activities. Curvulin and phaeosphaeride A were tested on weedy and agrarian plants, fungi, Gram-positive and Gram-negative bacteria, and on paramecia. Curvulin was shown to be weakly phytotoxic, while phaeosphaeride A caused severe necrotic lesions on all the tested plants. -

Abstract Aquatic hyphomycetes are a diverse, polyphyletic group of asexually reproducing fungi involved in the decomposition of litter in freshwater ecosystems. Curvularia eragrostidis, C. verruculosa and Phragmocephala atra were identified from sub- merged wood collected from freshwater streams in Yunnan Province, Southwestern China. They were characterised based on morphology and LSU, ITS and SSU sequence data. Phylogenetic analysis of LSU sequences placed the isolates within the order Pleosporales. Curvularia eragrostidis and C. verruculosa are reported from freshwater habitats for the first time. An epitype is designated for Curvularia verruculosa. This is the first phylogenetic placement of the genus Phragmocephala in the family Melanommataceae in Dothideomycetes, providing new DNA sequence data. -

A biotic cause of the disease is most likely since soil disinfection can restore plant growth. Fungi have appeared to contribute to the complex of biotic factors, but up to now the actual cause of the disease remains unknown. Further, environmentally friendly and practically applicable mitigation strategies are missing. Fungal root endophytes were isolated in two central experiments of the ORDIAmur consortium. Dark septate endophytes (Leptodontidium spp.) were frequently isolated from apple roots. An abundant occurrence of Nectriaceae fungi (Dactylonectria torresensis and Ilyonectria robusta) was found in ARD roots. Reference sites displayed a different characteristic fungal community. In roots grown in irradiated soil, a reduction of the number of isolated fungi and a changed composition of the fungal community was found. To investigate the effect of fungal endophytes on apple plants a quick and soil-free bio test in Petri dishes was developed using perlite. Inoculated fungi isolated from ARD roots induced neutral (Plectosphaerella, Pleotrichocladium, and Zalerion) to negative (Cadophora, Calonectria, Dactylonectria, Ilyonectria, and Leptosphaeria) plant reactions. After re-isolation, most of the Nectriaceae isolates were confirmed as pathogens. Microscopic analyses of ARD-affected roots revealed necroses caused by an unknown fungus that forms cauliflower-like (CF) structures in diseased cortex cells. -

Abstract: Fungal endophytes are the most ubiquitous plant symbionts on earth and are phylogenetically diverse. Studies on the fungal endophytes in tobacco have shown that they are widely distributed in the leaves, stems, and roots, and play important roles in the composition of the microbial ecosystem of tobacco. Herein, we analyzed and quantified the endophytic fungi of healthy tobacco leaves at the seedling (SS), resettling growth (RGS), fast-growing (FGS), and maturing (MS) stages at three altitudes [600 (L), 1000 (M), and 1300 m (H)]. We sequenced the ITS region of fungal samples to delimit operational taxonomic units (OTUs) and phylogenetically characterize the communities. The result showed the number of clustering OTUs at SS, RGS, FGS and MS were greater than 170, 245, 140, and 164, respectively. At the phylum level, species in Ascomycota and Basidiomycota had absolute predominance, representing 97.8% and 2.0 % of the total number of species. We also found the number of unique fungi at the RGS and FGS stages were higher than those in the other two stages. -

Current Research in Environmental & Applied Mycology 6(3), 231– 237, Doi 10.5943/cream/6/3/11 Abstract Bipolaris rostratae, a new foliicolous anamorphic fungus discovered on living leaves of Heliconia rostrata (Heliconiaceae), is described and illustrated. The species was compared with closely related species of Bipolaris and similar fungi recorded on Heliconia spp. This species is different from other Bipolaris spp. reported on Heliconia due to its shorter, thinner and less septate conidia. A key is provided to all species of Bipolaris reported on Heliconia. Key words − fungal diversity – morphotaxonomy – Foliicolous fungi – Bipolaris – new species Introduction After several taxonomic refinements, graminicolous Helminthosporium were segregated into several genera including Bipolaris, Curvularia, Drechslera and Exserohilum (Sivanesan 1987). These genera belong to Ascomycota, Dothideomycetes, Pleosporales, Pleosporaceae. These genera can be distinguished on the basis of characters such as conidial shape and size, hilum morphology, origin of the germ tubes from the basal or other conidial cells, and the location and sequence in the development of the conidial septa. Illustrations of different hilum morphologies in graminicolous Helminthosporium species were given by Alcorn (1988). -
