In histology, different staining methods can be used for the differentiated visualisation of connective tissue. Most of these stainings for connective tissue belong to the group of the so-called trichrome staining methods in which three different dyes are used. They function according to the following basic principle: The dyes used have different molecular sizes. The smaller dye molecule penetrates tissue most rapidly and stains a structure, whereas the larger dye molecule penetrates tissue more slowly and displaces the smaller molecule from the structure to which it attached.

Trichrome staining according to Masson-Goldner combines azophloxine, Orange G and light green SF whereby muscle fibre, collagenous fibre, fibrin and erythrocytes can be selectively visualised. Due to the carefully adjusted concentrations in the dyes used, the individual tissue components can be clearly differentiated by brilliant colours.

Since trichrome stainings use acid dyes, nuclei have to be stained with Weigert's iron hematoxylin. Nuclei stained with ordinary hematoxylin solutions are decoloured. For nucleus staining we recommend the ready-to-use, very stable Weigert's iron hema- toxylin solution which is offered separately (see additional reagents).

Procedure of Masson-Goldner trichrome staining kit Staining procedure 1. Deparaffinate the sections in the typical manner and rehydrate 2. Hematoxylin acc. to Weigert 5min 3. Running tap water 5min 4. Rinse in 1% acetic acid 30 sec 5. Azophloxine solution 10 min 6. Rinse in 1% acetic acid 30 sec 7. Tungstophosphoric acid Orange G solution 1min 8. Rinse in 1% acetic acid 30 sec 9. Light green SF solution 2min 10. Rinse in 1% acetic acid 30 sec 11. Ascending alcohol series, 2 x Neo-Clear® or xylene 12. Mount with Neo-Mount® or Entellan® new Material: formalin-fixed paraffin sections. Preparation: dilute the 10 % acetic acid 1:10 with distilled water.
