Prof. Dr. Pornsilp Pholpunthin) Major Advisor ..………….....………………Signature (Dr. Hendrik Segers) Co-Advisor .………….....………………Signature (Miss Phannee Sa-ardrit) Candidate iv I hereby certify that this work has not been accepted in substance for any other degree, and is not being currently submitted in candidature for any degree. ………………………………….Signature (Miss Phannee Sa-ardrit) Candidate v ชื่อวิทยานิพนธ์ ชีววิทยาเชิงโมเลกุลและสัณฐานวิทยาของชนิดทับซ้อนของโรติเฟอร์ในวงศ์ Brachionidae และวงศ์ Lecanidae (Eurotatoria: Monogononta: Brachionidae and Lecanidae) ในประเทศไทย ผู้เขียน นางสาวพรรณี สอาดฤทธิ์ สาขาวิชา ชีววิทยา ปีการศึกษา 2559 บทคัดย่อ การศึกษาครั้งนี้มีวัตถุประสงค์เพื่อวิเคราะห์หาโรติเฟอร์ชนิดซ่อนเร้นที่อยู่ภายในโรติเฟอร์ ชนิดทับซ้อนในวงศ์ Brachionidae และวงศ์ Lecanidae ที่พบในประเทศไทย โดยศึกษาความ แตกต่างของนิวคลีโอไทด์ของยีน COI และ ITS1 ด้วยวิธีการทางอนุกรมวิธานชีวโมเลกุล 3 วิธี ได้แก่ ABGD, PTP และ GMYC ในการก าหนดขอบเขตของชนิดและประมาณจ านวนชนิดที่พบในโรติเฟอร์ ชนิดทับซ้อน ความสอดคล้องของจ านวนชนิดที่ประมาณได้จากทุกวิธีการ จะใช้ยืนยันการมีอยู่ของ ชนิดซ่อนเร้นภายในโรติเฟอร์ชนิดทับซ้อน การประมาณจ านวนชนิดที่แท้จริงภายในชนิดทับซ้อน นอกจากจะสามารถน าไปใช้ประมาณความหลากหลายทางชีวภาพของโรติเฟอร์ที่แท้จริงในประเทศ ไทยแล้ว ยังน าไปสู่ความเข้าใจทางนิเวศวิทยา และวิวัฒนาการของโรติเฟอร์ในกลุ่มที่มีความสัมพันธ์ ใกล้ชิดกันมากขึ้น นอกจากนี้การศึกษาความแตกต่างทางพันธุกรรมกับการแพร่กระจายทางภูมิศาสตร์ ของชนิดซ่อนเร้นในประเทศไทย ยังถูกศึกษาโดยวิเคราะห์ด้วยวิธีการทางสถิติ Mantel test ผลการศึกษาโดยอาศัยวิธีการทางอนุกรมวิธานชีวโมเลกุล ชี้ให้เห็นถึงชนิดซ่อนเร้นภายใน โรติเฟอร์ชนิดทับซ้อนทั้งสองวงศ์ที่พบในประเทศไทย โดยพบจ านวนชนิดซ่อนเร้นมากที่สุดในกลุ่ม Lecane bulla โดยพบชนิดซ่อนเร้นอย่างน้อย 20 ชนิด อยู่ภายในชนิดทับซ้อนของ Lecane bulla การศึกษาครั้งนี้ได้น าลักษณะทางสัณฐานวิทยาของ Lecane bulla มาวิเคราะห์ร่วมด้วย พบว่า ลักษณะทางสัณฐานวิทยาสามารถน ามาใช้อธิบายความแตกต่างของบางชนิดได้เพียงเล็กน้อย แต่ไม่ สามารถใช้จ าแนกชนิดซ่อนเร้นทั้งหมดที่อยู่ภายใน Lecane bulla นอกจากนี้ยังพบชนิดซ่อนเร้น ภายในชนิดทับซ้อนอื่น ๆ ได้แก่ Brachionus angularis, Brachionus calyciflorus, Brachionus caudatus, Brachionus quadridentatus, Keratella cochlearis, Keratella tropica, Lecane bulla, Lecane closterocerca, Lecane crenata, Lecane curvicornis, Lecane hamata, Lecane leontina, Lecane ludwigii, Lecane luna, Lecane lunaris, Lecane nitida, vi Lecane quadridentata, Lecane signifera และ Lecane unguitata และจากการวิเคราะห์ด้วย DNA taxonomy พบว่าผลที่ได้จากการวิเคราะห์ล าดับนิวคลีโอไทด์ของยีน COI มีความสอดคล้องกัน ของจ านวนชนิดที่ประมาณได้ในแต่ละวิธี ขณะที่การประมาณจากยีน ITS1 มีความแตกต่างกันในแต่ ละวิธี และจากการวิเคราะห์เปรียบเทียบความแตกต่างทางพันธุกรรมภายในชนิดซ่อนเร้นในแต่ละ พื้นที่ของประเทศไทย พบว่าไม่มีความแตกต่างทางพันธุกรรมระหว่างพื้นที่ทางภูมิศาสตร์ ยกเว้น Lecane curvicornis ชี้ให้เห็นว่าโรติเฟอร์ชนิดซ่อนเร้นที่พบในประเทศไทยมีการกระจายทาง ภูมิศาสตร์ที่ซ้อนทับกันและกระจายตัวได้ในบริเวณกว้างในประเทศไทย vii Thesis Title Combined molecular-morphological approach to species complexes of Brachionid and Lecanid rotifers (Eurotatoria: Monogononta: Brachionidae and Lecanidae) in Thailand Author Miss Phannee Sa-ardrit Major Program Biology Academic Year 2016 ABSTRACT This study aims to analyze potential species complexes of rotifers within the families Brachionidae and Lecanidae, to test the hypothesis whether cryptic diversity is indeed as common as is being put forward. Moreover, this study analyzed the geographic distribution and genetic divergence within species complexes in Thailand. The tests for the presence of cryptic species were applied using three methods on both COI and ITS1 datasets: Automatic Barcode Gap Discovery (ABGD), a Poisson Tree Processes (PTP) model, and the Generalized Mixed Yule Coalescent (GMYC) model. Genetic divergence and geographic distance were analyzed using Mantel test. The results from three DNA taxonomy approaches, ABGD, PTP, and GMYC from COI and ITS1 markers clearly indicated the existence of cryptic species in both families. The highest estimate of species diversity in this study was Lecane bulla with at least 20 species existing in the complex. Morphological characters were examined. However, morphology cannot be used to separate some cryptic species in Lecane bulla complex. Moreover, this study reveals the existence of cryptic species in Brachionus angularis, Brachionus calyciflorus, Brachionus caudatus, Brachionus quadridentatus, Keratella cochlearis, Keratella tropica, Lecane bulla, Lecane closterocerca, Lecane crenata, Lecane curvicornis, Lecane hamata, Lecane leontina, Lecane ludwigii, Lecane luna, Lecane lunaris, Lecane nitida, Lecane quadridentata, Lecane signifera, and Lecane unguitata. The species estimated using COI marker is congruent among different DNA taxonomy methods, while there is incongruence among different methods based on ITS1 marker. Only Lecane curvicornis shows significant correlation between genetic and geographic distances. No correlation occurs between genetic and geographic distances in other species complexes. Thus, there is a geographic overlap in rotifer distribution in Thai waters. viii ACKNOWLEDGEMENTS This thesis would not have been completed without some assistance from many people. The present analysis of alpha diversity of Rotifera of the floodplain wetlands of Barak valley, south Assam, northeast India (NEI) reveals 170 species, belonging to 39 genera and 19 families, with three species new to the Indian sub-region and two species new to Assam state. The rich and diverse rotifer assemblages comprise ~70%, ~57% and ~39% of total species of the Phylum known till date from Assam, NEI and India, respectively and thus affirm habitat and ecological heterogeneity of the Barak River floodplain wetlands. The elements of global biogeographic importance include one Australasian, eight Oriental, 10 Paleotropical, one Indo-Chinese, one cosmo (sub) tropical and four other species, and ~37% species merit regional distribution interest for India with ~13% exclusively reported from NEI. Lecanidae (~30%) and Brachionidae ≈ Lepadellidae (~32%) largely contribute to the rotifer richness; Trichocercidae > Testudinellidae are notable (~15%), and Notommatidae, Euchlanidae, Mytilinidae, Trichotriidae and Trochosphaeridae collectively form 16.5%. Lecane is the most speciose genus; Lepadella > Brachionus = Trichocerca include ~32% species, while Testudinella > Keratella deserve mention. Rotifera of the Barak floodplains exhibit the littoral-periphytonic nature, several small sized species and tropical character. Keywords. There are 35 species of sessile rotifers from three families Atrochidae, Collothecidae, and Flosculariidae, but to that four more species in the planktonic family Conochilidae may be added, as recent molecular research indicates that this family lies within Flosculariidae.

We identified 44 different taxa of sessile rotifers, including thirty-nine fixosessile species and three planktonic colonial species. This corresponds with about 40 % of the global sessile rotifer diversity, and is the highest alpha-diversity of the group ever recorded from a single lake. The record further includes a new genus, Lacinularoides n. gen., containing a single species L. coloniensis (Colledge, 1918) n. comb., which is redescribed, and several possibly new species, one of which, Ptygura thalenoiensis n. spec. is formally described here. Ptygura noodti (Koste, 1972) n. comb. is relocated from Floscularia, based on observations of living specimens of this species, formerly known only from preserved, contracted specimens from the Amazon region.

The speciose rotifers of small lentic biotopes of Arunachal Pradesh, Mizoram, Nagaland, Meghalaya, Manipur and Tripura, the floodplain lakes beels( ) and small wetlands (dobas and dubies) of the Brahmaputra and the Barak floodplains of Assam, and the floodplain lakes (pats) of Manipur are noteworthy. Deepor Beel and Loktak Lake (two Ramsar sites) are the globally rich rotifer `hotspots’. Interesting assemblages per sample of 80+ species in certain beels and pats, and up to 50 species in dobas and dubies depict the `Rotifera paradox’.

Schmitt Abstract The serotonergic nervous systems of two non-colonial species of Conochilus were examined to obtain the first immunohistochemical insights into the neuroanatomy of species of Flosculariacea (Rotifera, Monogononta). Species of Conochilus, subgenus Conochiloides, were examined using serotonin (5-HT) immunohistochemistry, epifluorescence and confocal laser scanning microscopy, and 3D computer imaging software. In specimens of C. coenobasis and C. dossuarius, the serotonergic nervous system is defined by a dorsal cerebral ganglion, apically directed cerebral neurites, and paired nerve cords. The cerebral ganglion contains approximately four pairs of small 5-HT- immunoreactive perikarya; one pair innervates the posterior nerve cords and three pairs innervate the apical field. The most dorsal pair innervates a coronal nerve ring that encircles the apical field. Within the apical field is a second nerve ring that outlines the inner border of the coronal cilia. Together, both the inner and outer nerve rings may function to modulate ciliary activity of the corona. The other two pairs of perikarya innervate a region around the mouth. Specific differences in the distribution of serotonergic neurons between species of Conochilus and previously examined ploimate rotifers include the following: (a) a lack of immunoreactivity in the mastax; (b) a greater number of apically directed serotonergic neurites; and (c) a complete innervation of the corona in both species of Conochilus.

This list has been produced by the Department of Sustainability, Environment, Water, Population and Communities (SEWPC) for the Natural Resource Management Spatial Information System. The list was produced using the AustralianAustralian Natural Natural Heritage Heritage Assessment Assessment Tool Tool (ANHAT), which analyses data from a range of plant and animal surveys and collections from across Australia to automatically generate a report for each NRM region. Data sources (Appendix 2) include national and state herbaria, museums, state governments, CSIRO, Birds Australia and a range of surveys conducted by or for DEWHA. For each family of plant and animal covered by ANHAT (Appendix 1), this document gives the number of species in the country and how many of them are found in the region. It also identifies species listed as Vulnerable, Critically Endangered, Endangered or Conservation Dependent under the EPBC Act. A biodiversity summary for this region is also available. To date, 402 species of rotifers have been recorded from Mexico, besides a few infraspecific taxa such as subspecies and varieties. The rotifers from Mexico represent 27 families and 75 genera. Molecular analysis showed about 20 cryptic taxa from species complexes. The genera Lecane, Trichocerca, Brachionus, Lepadella, Cephalodella, Keratella, Ptygura, and Notommata accounted for more than 50% of all species recorded from the Mexican territory. The diversity of rotifers from the different states of Mexico was highly heterogeneous. Only five federal entities (the State of Mexico, Michoacán, Veracruz, Mexico City, Aguascalientes, and Quintana Roo) had more than 100 species. Extrapolation of rotifer species recorded from Mexico indicated the possible occurrence of more than 600 species in Mexican water bodies, hence more sampling effort is needed. In the current review, we also comment on the importance of seasonal sampling in enhancing the species richness and detecting exotic rotifer taxa in Mexico. Thirteen species of Lecanidae were identified, 6 of which are new records for Iran. The new records include Lecane flexilis (Gosse 1886), L. hamata (Stokes 1896), L. hastata (Murray 1913), L. lamellata (Daday 1893), L. papuana (Murray 1913), and L. punctata (Murray 1913). Drawings are provided, and both the seasonal and spatial distributions of the identified taxa are discussed. A brief comparison is made between the present study’s results and those reported from Turkey. Key words: Rotifera, Lecanidae, Iran Introduction Lecane is among the best-represented genera in The Lecanidae constitutes a group of common Southeast Asia (Segers, 2001). In the Middle East the fresh and saline-water monogonont rotifers. The most intensive work has been performed in Turkey family’s single genus, Lecane, with about 200 species, (Ustaoğlu, 2004).
