Simple Summary: Paradise fish (Macropodus opercularis) has been a favored subject of behavioral research during the last decades of the 20th century. Lately, however, with a massively expanding genetic toolkit and a well annotated, fully sequenced genome, zebrafish (Danio rerio) became a central model of recent behavioral research. But, as the zebrafish behavioral repertoire is less complex than that of the paradise fish, the focus on zebrafish is a compromise. Roberts Research Associate, Smithsonian Tropical Research Institute Email: [email protected] ABSTRACT. - Celestichthys margaritatus, a new genus and species of Danioinae, is described from a rapidly developing locality in the Salween basin about 70-80 km northeast of Inle Lake in northern Myanmar. Males and females are strikingly colouful. It is apparently most closely related to two danioins endemic to Inle, Microrasbora rubescens and "Microrasbora" erythromicron. The latter species may be congeneric with the new species. The new genus is identified as a danioin by specializations on its lower jaw and its numerous anal fin rays. The colouration, while highly distinctive, seems also to be characteristically danioin. The danioin notch (Roberts, 1986; Fang, 2003) is reduced or absent, but the danioin mandibular flap and bony knob (defined herein) are present. The anal fin has iiiSVz-lOV: rays. In addition to its distinctive body spots and barred fins the new fish is distinguished from other species of danioins by the following combination of characters: snout and mouth extremely short; premaxillary with an elongate and very slender ascending process; mandible foreshortened; body deep, with rounded dorsal and anal fins; modal vertebral count 15+16=31; caudal fin moderately rather than deeply forked; principal caudal fin rays 9/8; scales vertically ovoid; and pharyngeal teeth conical, in three rows KEY WORDS. - Hopong; principal caudal fin rays; danioin mandibular notch, knob, and pad; captive breeding. -

Armbruster1 Abstract Background: Cypriniformes (minnows, carps, loaches, and suckers) is the largest group of freshwater fishes in the world (~4300 described species). Despite much attention, previous attempts to elucidate relationships using molecular and morphological characters have been incongruent. In this study we present the first phylogenomic analysis using anchored hybrid enrichment for 172 taxa to represent the order (plus three out-group taxa), which is the largest dataset for the order to date (219 loci, 315,288 bp, average locus length of 1011 bp). Results: Concatenation analysis establishes a robust tree with 97 % of nodes at 100 % bootstrap support. Species tree analysis was highly congruent with the concatenation analysis with only two major differences: monophyly of Cobitoidei and placement of Danionidae. Conclusions: Most major clades obtained in prior molecular studies were validated as monophyletic, and we provide robust resolution for the relationships among these clades for the first time. These relationships can be used as a framework for addressing a variety of evolutionary questions (e.g. phylogeography, polyploidization, diversification, trait evolution, comparative genomics) for which Cypriniformes is ideally suited. Risk Assessment to add Kryptopterus vitreolus to, and remove Kryptopterus bicirrhis from, the Environment Protection and Biodiversity Conservation Act 1999 List of Specimens taken to be Suitable for Live Import August 2020 Introduction Purpose of proposed amendments The Australian Government Department of Agriculture, Water and the Environment (the Department) proposes to amend the List of Specimens Taken to be Suitable for Live Import (Live Import List) to: 1. include Kryptopterus vitreolus, commonly known as the Glass Catfish, and 2. remove Kryptopterus bicirrhis, which was known previously as the Glass Catfish. The Department has recently become aware that the commonly traded Glass Catfish which appears on the Live Import List has been misidentified. The species imported into Australia (and around the world) is a previously unnamed species which has been globally misidentified for over 80 years before being identified as Kryptopterus vitreolus. The species on the Live Import List with the common name Glass Catfish (Kryptopterus bicirrhis) is not imported or common in the aquarium industry. The Department proposes to include Kryptopterus vitreolus on, and remove Kryptopterus bicirrhis from, the Live Import List. The Department’s objective is to enable legal imports of the species Kryptopterus vitreolus into Australia under its correct taxonomical name. Background Under s303EC of the EPBC Act, the Minister may amend the Live Import List by including a specimen in the list. There are two parts to the list—Part 1 comprises specimens that can be imported without a permit under the Act and Part 2 comprises specimens, including CITES species, that require a permit under the Act to be imported. -

The dataset comprises 1 individual from each of 15 species, 13 Danios and two outgroups. These data are from McClusckey and Postlethwait, 2015. • Software - All are open source software • Stacks (http://catchenlab.life.illinois.edu/stacks/) - A set of interconnected open source programs designed initially for the de novo assembly of RAD sequences into loci for genetic maps, and extended to be used more flexibly in studies of organisms with and without a reference genome. • RAxML (http://sco.h-its.org/exelixis/web/software/raxml/index.html) - A software program written by the Exelixis Lab for the construction of maximum likelihood phylogenetic trees. !1 Exercise IV. Building a RADseq-based Phylogenetic Tree 1. In this exercise we will be working on a set of RAD-seq data taken from a number of Danio species, and two outgroups to Danio. We want to understand how these species relate to one another phylogenetically by examining variable sites between the genomes of these 15 species. The data set includes one sample per species. Our goal is to process the raw RAD data and reconstruct loci shared across the species. -

Introduction Diploidic germplasms such as embryos, compared to haploidic gametes, are theoretically a better choice for preservation of an animal species. However, there are significant challenges in cryopreservation of multicellular materials due to their size and physical complexity which affect the permeation of cryoprotectants and water, sensitivity to chilling and toxicity of cryoprotectants. While cryopreservation technologies are well developed and found feasible in embryos/larvae of some species, embryos of other species such as zebrafish failed to be cryopreserved. In addition, cryopreservation in many other emerging model organisms have not been developed at all. Hence, the limited cryopreservation technology has become a bottleneck in the development of various research areas, especially those relying on molecular genetics of emerging model organisms. Thorough understanding of the embryonic development and critical stages tolerant to cryopreservation needs to be identified so as to facilitate expansion of model systems available for specific biological and experimental interrogations. 1.2 Traditional and emerging animal models Classical animal models, including species that represent major branches of the tree of life, are being used in biological studies. They include Caenorhabditis elegans (a nematode), Drosophila melanogaster (an arthropod), Danio rerio (a teleost fish), Gallus gallus (an avian), and Mus musculus (a mammal). They have been widely used in scientific research, primarily due to the ease of maintenance and specific features that facilitate experimental manipulations, genetic study and observation. As knowledge from these models has accumulated over the years, they offer important insights into the overall organization and functional composition of the general form of life. -
