Western Blot Immunocytochemistry Detection of Human, Mouse, and Rat Enolase 2/Neuronspecific Enolase in SH Enolase 2/Neuronspecific Enolase by SY5Y Human Cell Line. Enolase 2/Neuron Western Blot. Western blot shows lysates of specific Enolase was detected in immersion Jurkat human acute T cell leukemia cell line, fixed SHSY5Y human neuroblastoma cell SHSY5Y human neuroblastoma cell line, line using Mouse AntiHuman Enolase human brain tissue (cerebellum), mouse 2/Neuronspecific Enolase Monoclonal brain tissue (cerebellum), and rat brain tissue Antibody (Catalog # MAB51691) at 1 µg/mL (cerebellum). PVDF membrane was probed for 3 hours at room temperature. Cells were with 0.5 µg/mL of Mouse AntiHuman stained using the NorthernLights™ 557 Enolase 2/Neuronspecific Enolase conjugated AntiMouse IgG Secondary Monoclonal Antibody (Catalog # MAB51691) Antibody (red; Catalog # NL007) and followed by HRPconjugated AntiMouse IgG counterstained with DAPI (blue). Specific Secondary Antibody (Catalog # HAF018). A staining was localized to cytoplasm and specific band was detected for Enolase plasma membrane. BACKGROUND Enolase 2 (2phosphoDglycerate hydrolyase; also Neuronspecific Enolase, NSE, neural enolase and γenolase) is a 46 kDa member of the Enolase family of enzymes. It is expressed in developing neurons and glia, is known to catalyze the generation of phosphoenolpyruvate, and is suggested to possess neurotrophic activity for neurons, likely through an extracellular mechanism. Human Enolase 2 is 434 amino acids (aa) in length. The enzymatic site spans most of the length of the molecule. Enolase 2 exists as both a noncovalentlylinked homodimer, or heterodimer with αenolase. Fulllength human Enolase 2 shares 99% aa identity with both mouse and canine Enolase 2. It shares 83% aa identity with human enolases 1 and 3.
