Storage: 2 years -80°C in solvent; 3 years -20°C powder; DMSO 10 mg/mL, warmed&ultrasonic Solubility ( < 1 mg/ml refers to the product slightly soluble or insoluble ) In vivo Activity Acrivastine (usually 8mg three times daily) is an effective and well-tolerated antihistamine in the treatment of chronic urticaria and allergic rhinitis. Acrivastine causes less drowsiness than clemastine, the incidence of adverse effects being indistinguishable from that with placebo or terfenadine. In the treatment of dermatoses in which histamine has a pathogenetic role, the efficacy of acrivastine is superior to that of placebo and similar to that of usual dosages of clemastine, hydroxyzine, chlorpheniramine, cyproheptadine or terfenadine [1]. Both 4 mg and 8 mg acrivastine alleviate the symptoms of seasonal allergic rhinitis with significant improvements in the symptom scores for sneezing, running nose and the calculated overall score. In addition, 8 mg acrivastine reduces the symptom scores for watery eyes and itchy throat. Acrivastine is both well tolerated and effective in the treatment of seasonal allergic rhinitis [2]. Until specific studies have been carried out Acrivastine capsules should not be given to patients with significant renal impairment. Interaction with other medicinal products and other forms of interaction It is usual to advise patients not to undertake tasks requiring mental alertness whilst under the influence of alcohol and other CNS depressants.

Elderly: It has been shown in clinical studies with a total daily dosage of 24 mg acrivastine plus 180 mg pseudoephedrine that no special precautions are required, although it may be advisable to monitor renal function (see section 4.3). 4.3 Contraindications Acrivastine + pseudoephedrine is contra-indicated in individuals with known hypersensitivity to acrivastine, triprolidine or pseudoephedrine. Acrivastine+ pseudoephedrine is contra-indicated in patients with severe hypertension or severe coronary artery disease. The concomitant use of a pseudoephedrine-containing product and monoamine oxidase inhibitors can occasionally cause a rise in blood pressure. Acrivastine + pseudoephedrine is therefore contra-indicated in patients who are taking or have taken monoamine oxidase inhibitors within the preceding two weeks. The antibacterial agent furazolidone is known to cause a dose-related inhibition of monoamine oxidase. Although there are no reports of hypertensive crises caused by the concurrent administration of acrivastine + pseudoephedrine and furazolidone, they should not be taken together. Renal excretion is the main route of elimination of the active ingredients of acrivastine + pseudoephedrine. Until specific studies have been carried out acrivastine+ pseudoephedrine should not be given to patients with significant renal impairment (creatinine clearance less than 50 ml/min or serum creatinine greater than 150 micromol/litre).

However, there is still a lack of collective evidence regarding the antihistamine effects and safety profiles of fexofenadine relative to other antihistamine drugs and placebo. Therefore, we aimed to systematically evaluate the antihistamine effects and safety of fexofenadine. Methods: An electronic literature search of randomized controlled trials (RCTs) was performed using Embase, Cochrane and PubMed from establishment to January 1st, 2018. RCTs comparing the antihistamine effects or safety (adverse events, sedative effects, and cognitive/psychomotor function) of fexofenadine with either other antihistamines or placebo for healthy subjects and patients with allergy were selected. Results: Fifty-one studies of 14,551 participants met the inclusion criteria. When compared with the first-generation antihistamines, fexofenadine produced significantly lower adverse events frequency (OR = 0.446; 95% CI: 0.214 to 0.929, P = 0.031), significantly lower sedative effects frequency (OR = 0.265; 95% CI: 0.072 to 0.976, P = 0.046) and significantly less change of all cognitive/psychomotor function.
