Pfister Danny Haelewaters STUDIES OF THE LABOULBENIOMYCETES: DIVERSITY, EVOLUTION, AND PATTERNS OF SPECIATION ABSTRACT CHAPTER 1: Laboulbeniales is one of the most morphologically and ecologically distinct orders of Ascomycota. These microscopic fungi are characterized by an ectoparasitic lifestyle on arthropods, determinate growth, lack of asexual state, high species richness and intractability to culture. DNA extraction and PCR amplification have proven difficult for multiple reasons. DNA isolation techniques and commercially available kits are tested enabling efficient and rapid genetic analysis of Laboulbeniales fungi. Success rates for the different techniques on different taxa are presented and discussed in the light of difficulties with micromanipulation, preservation techniques and negative results. CHAPTER 2: The class Laboulbeniomycetes comprises biotrophic parasites associated with arthropods and fungi. -

Department of Biology, Imperial College at Silwood Park, Ascot, Berkshire, SL5 7PY, U.K. August 1999 1 THESIS ABSTRACT Populations of the sycamore aphid Drepanosiphum platanoidis Schrank (Homoptera: Aphididae) have been shown to undergo regular two-year cycles. It is thought this phenomenon is caused by an inverse seasonal relationship in abundance operating between spring and autumn of each year. It has been hypothesised that the underlying mechanism of this process is due to a plant factor, intra-specific competition between aphids, or a combination of the two. This thesis examines the population dynamics and the life-history characteristics of D. platanoidis, with an emphasis on elucidating the factors involved in driving the dynamics of the aphid population, especially the role of bottom-up forces. Manipulating host plant quality with different levels of aphids in the early part of the year, showed that there was a contrast in aphid performance (e.g. duration of nymphal development, reproductive duration and output) between the first (spring) and the third (autumn) aphid generations. This indicated that aphid infestation history had the capacity to modify host plant nutritional quality through the year. However, generalist predators were not key regulators of aphid abundance during the year, while the specialist parasitoids showed a tightly bound relationship to its prey. The effect of a fungal endophyte infecting the host plant generally showed a neutral effect on post-aestivation aphid dynamics and the degree of parasitism in autumn. -

Pfster1 Accepted: 8 October 2018 Our understanding of fungal diversity is far from complete. Species descriptions generally focus on Published: xx xx xxxx morphological features, but this approach may underestimate true diversity. Using the morphological species concept, Hesperomyces virescens (Ascomycota, Laboulbeniales) is a single species with global distribution and wide host range. Since its description 120 years ago, this fungal parasite has been reported from 30 species of ladybird hosts on all continents except Antarctica. These host usage patterns suggest that H. virescens could be made up of many diferent species, each adapted to individual host species. Using sequence data from three gene regions, we found evidence for distinct clades within Hesperomyces virescens, each clade corresponding to isolates from a single host species. We propose that these lineages represent separate species, driven by adaptation to diferent ladybird hosts. Our combined morphometric, molecular phylogenetic and ecological data provide support for a unifed species concept and an integrative taxonomy approach. What is a species? Tis is a perennial question in evolutionary biology. Te answer is complex and has been intensely argued for decades. Diferent species concepts corresponding to multiple biological properties pro- vide a means to recognize, delineate and describe species. Tese properties include diferences in morphological traits, nucleotide divergence and monophyly, reproductive isolation, ecological niches or adaptive zones, mate recognition or mating systems, geographic range, exclusive coalescence of alleles, etc. However, biologists from various research felds have advocated diferent and sometimes incompatible species concepts, leading to varying conclusions regarding delimitation of species and their numbers. -

The harlequin ladybird is a global invader that causes the decline of native ladybirds in some countries. Therefore, it is advisable to monitor the ladybird fauna in regions recently occupied by this species. We analyzed the dynamics of the fauna at the main sea resort of Russia over a period of 120 years to determine the following: (1) how the ladybird biodiversity changed during the intensive landscape transformation; (2) what alien species introduced for pest control have occurred to date; and (3) what the impact is of the harlequin ladybird on the ladybird fauna. We examined specimens collected by us and 54 other collectors including specimens from old museum collections and reconstructed the history of the biodiversity like a picture from puzzle pieces. Surprisingly, landscape transformation did not cause a decrease but rather an increase in ladybird biodiversity; most of the species recorded before 1930 have occurred to date, and 23 other species have spread to the region. -

Kingley Vale NNR is managed by Natural England. My dissertation deals with the biodiversity in these woodlands. During my stay in England I learned many things about the different aspects of nature conservation in England. First of all I want to thank Katherine Birch (manager of Kingley Vale NNR) for giving guidance through my dissertation project and for creating lots of interesting days during my internship. All the specimens of the coccinellid ladybird beetles were collected from five major localities: City centre, Linnunlahti, Karsikko, Utra and Pilkko areas. In the family Coccinellidae, 16 species belonging to 10 genera were found in our experiment time. Most of ladybird’s common host–plant was Rosa rugosa and also other different species of plants. All the host-plants were growing naturally in the local ecosystem of the experimental area. Out of the 16 species of ladybird beetles from Joensuu, there were two species (Cryptolaemus montrouzieri and Chilocorus stigma) reported for the first time from North Karelia and is therefore, a new addition to the ladybird beetles fauna of Finland. Museomics in combination with morphological analysis is useful for systematic studies. Eriopis is a genus of ladybird beetles (Coccinellidae) that lives in South America. This study presents Eriopis patagonia, a new species of ladybird beetle discovered with two old specimens collected in Patagonia at least 100 years ago and deposited in a natural history collection. DNA was extracted from the specimens by a non-destructive method, allowing the specimens to be preserved again. The total gDNA was sequenced using Next-Generation Sequencing (NGS) technologies. The genetic information obtained allows us to reconstruct and describe its mitochondrial genome and examine its phylogenetic position. Abstract: Natural history collections house an important source of genetic data from yet unexplored biological diversity. Molecular data from museum specimens remain underexploited, which is mainly due to the degradation of DNA from specimens over time. However, Next-Generation Sequencing (NGS) technology can now be used to sequence “old” specimens. Indeed, many of these specimens are unique samples of nomenclatural types and can be crucial for resolving systematic or biogeographic scientific questions. -
