MOLECULAR SYSTEMATICS OF CYANOBACTERIA Submitted by Elvina Lee Mei Hui, BSc (Hons) This thesis is presented for the degree of Doctor of Philosophy of Murdoch University i DECLARATION I declare that this thesis is my own account of my research and contains as its main content work, which has not been previously submitted for a degree at any tertiary education institution. ______________________________ Elvina Lee January 2016 ii ABSTRACT Cyanobacteria constitute a phylum of ubiquitous cosmopolitan bacteria with the ability to perform oxygenic photosynthesis. Their ancient origins, ecological and economic potential, biotechnological applications and impact on water systems have attracted much interest from the academia, industry, health authorities and regulators. Despite this, cyanobacteria classification and nomenclature still remains difficult. One of the aims of this project was to further our understanding of cyanobacteria systematics by (1) testing polyphasic characterization methods and (2) examining the effect of various phylogenetic reconstruction strategies. Additionally, (3) Next Generation Sequencing (NGS) assays using novel cyanobacteria 16S rDNA targeted primers were implemented to provide better taxa resolution than that offered by “universal” 16S rDNA primers. Cyanobacteria strains from various water sources in Australia were isolated, characterised at four loci commonly used for cyanobacteria molecular classification, and tested for the presence of genes implicated in toxin and terpene (odour) production. A total of 55 novel cyanobacterial strains were isolated and maintained in culture, forming the first known collection of cyanobacteria isolates from Western Australia. Comparison of molecular– and morphology– based identifications not only showed the limitations of the current methods (only 45% of the isolates showed agreement) but also provided the opportunity to suggest guidelines and conceive a way forward towards more effective identification approaches. -

The whole classification of cyanobacteria (species, genera, families, orders) has undergone exten- sive restructuring and revision in recent years with the advent of phylogenetic analyses based on molecular sequence data. Several recent revisionary and monographic works initiated a revision and it is anticipated there will be further changes in the future. However, with the completion of the monographic series on the Cyanobacteria in Süsswasserflora von Mitteleuropa, and the recent flurry of taxonomic papers describing new genera, it seems expedient that a summary of the modern taxonomic system for cyanobacteria should be published. In this review, we present the status of all currently used families of cyanobacteria, review the results of molecular taxonomic studies, descriptions and characteristics of new orders and new families and the elevation of a few subfamilies to family level. -
Phenotypic and Ecological Diversity of Freshwater Coccoid Cyanobacteria from Maritime Antarctica and Islands of NW Weddell Sea

Jiří Komárek Institute of Botany AS CR, Třeboň, and Faculty of Science, University of South Bohemia, České Budějovice, Czech Republic Abstract The review of freshwater natural populations of coccoid cyanobacteria from the ecosystems of the coastal Antarctica in the vicinity of Antarctic Peninsula is presented in two consequent articles. The natural cyanobacterial populations were studied from deglaciated regions with aquatic and terrestrial habitats, mainly from the islands Nelson and King George (South Shetland Islands) and from the northern deglaciated part of the James Ross Island (NW part of the Weddell Sea). Majority of identified morphospecies was distinctly ecologically restricted and their cultivation was not successful; the molecular evaluation was therefore not possible. All morphotypes appeared also in small quantities, and only few morphospecies created locally more intense populations. However, the knowledge of phenotype variation is important for the classification of Antarctic cyanobacterial diversity. This first article contains 11 taxa (morphospecies) from 9 genera from the simple phylogenetic clade, classified recently in the order Synechococcales (the second part will contain species from more complicated unicellular cyanobacterial orders). Our results were compared with literary data, but several populations occurred only rarely -

This thesis is devoted to the systematic revisions of selected terrestrial cyanobacteria at several taxonomic levels. The methodology included thorough morphological characterization of cultured cyanobacterial strains using light and electron microscopy complemented with analyses of the molecular data: DNA sequencing, phylogenetic analyses based on 16S rRNA gene and the adjacent 16S-23S ITS region, and comparison of the predicted secondary structures of this region. Descriptions of new species, genera, families and an in-depth characterization of a previously poorly known family were achieved. Biodiversity Data Journal 4: e10084. https://doi.org/10.3897/BDJ.4.e10084 Abstract Background The checklist of Greek Cyanobacteria was created in the framework of the Greek Taxon Information System (GTIS), an initiative of the LifeWatchGreece Research Infrastructure (ESFRI) that has resumed efforts to compile a complete checklist of species reported from Greece. This list was created from exhaustive search of the scientific literature of the last 60 years. All records of taxa known to occur in Greece were taxonomically updated. New information The checklist of Greek Cyanobacteria comprises 543 species, classified in 130 genera, 41 families, and 8 orders. The orders Synechococcales and Oscillatoriales have the highest number of species (158 and 153 species, respectively), whereas these two orders along with Nostocales and Chroococcales cover 93% of the known Greek cyanobacteria species. It is worth mentioning that 18 species have been initially described from Greek habitats. The marine epilithic Ammatoidea aegea described from Saronikos Gulf is considered endemic to this area. Our bibliographic review shows that Greece hosts a high diversity of cyanobacteria, suggesting that the Mediterranean area is also a hot spot for microbes. -

ABSTRACT – The research aimed to study the cyanobacterial flora of an artificial pond and marsh at the Lutheran University of Brazil (Canoas, Rio Grande do Sul State), while understanding the distribution and environmental conditions where these species occur. Samples were collected monthly between November 2009 and October 2010. Twenty- four species were identified, highlighting the colonial forms (50%), with the predominance of the order Chroococcales (67%). The identified filamentous cyanobacteria belonged to Oscillatoriales (25%), Nostocales (17%) and Pseudanabaenales (8%). Out of all the identified species, only six [Microcystis aeruginosa (Kützing) Kützing M. wesenbergi (Komárek) Komárek, Oscillatoria curviceps Agardh ex Gomont, O. tenuis Agardh ex Gomont, Cuspidothrix issatschenkoi (Usačev) Rajaniemi et al. and Cylindrospermopsis raciborskii (Woloszynska) Seenayya & Subba Raju] were observed in both analyzed water bodies. Over the study period, Cylindrospermopsis raciborskii blooms occurred in the pond, while blooms of Microcystis aeruginosa were observed in the marsh in spring and summer. Cyanobacteria, also known as blue-green algae, are a very ancient group of photosynthetic bacteria. As probably the first photosynthetic organisms releasing oxygen, they altered the primitive, virtually oxygen-free atmosphere leading to the evolution of today’s living world. They are of universal occurrence in fresh and salt water, from the Antarctic to the arctic, in permanently frozen lakes and in hot springs, in soil, deserts and tundra. -
