Ltd. Properties and Use of DMDC 3.1 Overview DMDC is widely used in beverage and wine as low-temperature bactericide in the world. After been dosed into drinks, DMDC will decompose into trace of CO2 and Methanol ( When the temperature is 20℃, the half-life period in drinks is 2.13 hours ,completely decomposition is 7 hours), CO2 and Methanol are the natural composition of beverage and grape wine which are made of fruits and fruit juice. No There will be no danger to drinkers when hydrolysis is finished. 3.2 Mechanism of function By inactivating enzyme inside microbe-react with imidazole and amidogen, DMDC will eliminate all microbes. Imidazole and amidogen are the basic element of enzyme in microbes, so the effects of antibiotics is extensive. The primary target of DMDC including: yeast (brewer's yeast, Zygosaccharomyces, rhodotorula, Pichia pastoris, Torulopsis, etc.), mold(blue mold, hairy mold, Byssochlamys, Fusarium, etc.), bacteria(Pasteurianus, Pseudomonas aeruginosa, Staphylococcus aureus, Lactobacillus, etc.) In water, DMDC can kill 99.999%~99.9999% E. coli and yeast in 2 hours. Thus, the bactericidal ability of DMDC is very outstanding. 3.3 Product feature High potency Minute dosage can wipe out the microbe which spoil beverage. Safe DMDC has been approved by more than 110 countries. No residue Completely decomposed into trace of CO2 and Methanol. (Both are natural component of beverage) Highly-efficient on retain freshness DMDC has no odor at a low-concentration, it has no effect on beverage’s flavor, not to mention loss of nutritional ingredient. 2 Compatible with various packing materials DMDC is compatible with various packing materials such as glass, metal, plastic, paper box and so on. High-temperature PET bottle can be replaced by a cheaper one, for DMDC is capable of low-temperature bottling. Use and Dosing 4.1 Use in beverage and wine An appropriate dosing machine is indispensable for the use of DMDC, it can assure DMDC evenly dispersed in the beverage and wine. Operator must be trained by professional personnel. Eye and skin contact with DMDC should be avoided, Vapor and mist must not be inhaled. Keep DMDC away from heat source. Smoking is definitely prohibited. Take action to prevent static focus. For DMDC will decompose into beverage’s natural ingredient in a short time, and the bactericidal effect will not exist after decomposition. So the dosing step better be operated just before bottling. To ensure the bactericidal effect, placing the beverage bottle upside down is suggested after bottling, so DMDC can eliminate the microbes exist in the upper space and bottle cap. If interruption (over 1 hour) occurs in bottling process, in consideration of DMDC’s decomposition has already been on for a while, we suggest that empty the beverage tubes and bottling equipment, then restart bottling step. When High temperature sterilization and DMDC are used together, temperature of beverage should be under 35℃ before dosing DMDC into the beverage, in case that DMDC decomposes too fast. 4.2 Applications in other industries Other industries like plant extracts, industrial fermentation, consult with DMDC manufacturer. 4.3 Matters need attention Though DMDC performs magnificently in sterilization, but high-standard sanitary condition is still irreplaceable (including Contact us: Phone: +86 23 6308-6956 Fax: +86 23 6308-6956 Email: [email protected] Page 3 of 8 Chongqing Changfeng biotechnology Co. Ltd. hardware and management). For better effect of sterilization and ensure the product’s quality, regular cleaning and sterilization of equipment and tube are still necessary. Storage and Transportation 5.1 Storage condition and warranty period Store DMDC in cool, dry and well-ventilated area. The suggested temperature is 18-25℃. Temperature above 30℃ (no more than a week )does not affect the product quality. DMDC will decompose if exposed to high temperature for a long time. When the temperature is below 17℃ , DMDC will crystallize. Crystallization is significantly good for the extension of warranty period. But it need some time to melt DMDC before dosing. Unfreezing temperature must be below 40℃, dose DMDC as soon as it be unfrozen, in case of decomposition. Finish the use of DMDC as soon as possible after the original container is opened, if there is rest, seal the container with original bottle cap, and keep the container upright in case of leakage. If the stored time is too long or the temperature is always higher than suggested standard .CO2 may appear (for DMDC’s decomposition).If this happens, there will be the risk of container explosion. So be very careful, when open the container .Watch out for vapor at the same time. Inhaling product in a high concentration for a short time may also cause irritation to the nose, throat and respiratory system and cause suppurative bronchitis. Anaphase symptoms (1-5 hours) include discomfort, headache, severe irritating cough and irritation of the skin. Asthma may occur in the staff those are at the pretreatment place. When ventilation at the work place is not in good condition and DMDC is scattered in a large area, high vapor pressure of DMDC can enhance the concentration in the air (When accident happens or disposal is not adequate). Long-term inhalation will damage lungs, may lead to death in extreme situation. Caution: Never feed the insensible guy with anything or water. 7.2 Matters need attention for physician 4 Medical measures: When symptoms started, emesis is recommended after 15 to 30 minutes. If inhaled DMDC, inhale cortisone spray immediately. After the first aid is finished, 24-hour observation on the victim is necessary. 7.3 Firefighting measures Extinguish media Water spray, alcohol-resistant foam, dry chemical or carbon dioxide. Accidental Release Measures 8.1 Personal precautions, protective equipment and emergency procedures Wear respiratory protection. Avoid inhaling vapors, mist or gas. Ensure adequate ventilation. Remove all sources of ignition. Evacuate personnel to safe areas. Vapors can accumulate in low areas. Beware of vapors accumulating to explosion limit. For personal protection see section 8. 8.2 Environmental precautions Prevent further leakage or spillage if safe to do so. The issue of preventing the re-fermentation and protection against undesirable malolactic fermentation (MLF) in order to safe content of acids in wine is very complicated. In this paper the saturated higher fatty acids (HFA) – C8, C10 and C12, dimethyldicarbonate (DMDC) and sulphur dioxide (SO2) were tested. The re-fermentation test showed the strongest inhibition power at ratio 2:8, 1:9 and 0:10 as C8:C10 acids – 65 days without re-fermentation. MLF experiments confi rmed that addition of SO2 into the fermenting media causes rapid inhibition of lactic acid bacteria metabolic activity. Malic acid concentrations were proportionally decreasing during 6 days of experiment and at the end the content of this acid varied between 0.16 and 0.22 g/L, the only exception formed a variant with the addition of SO2 (1.57 g/L of malic acid). A er calculation of the average consumption rate of malic acid, the results showed the inhibition power – SO2 (81.05%) followed by variant of 40 mg/L mixture of HFA (40.76%), a variant of 200 mg/L of DMDC (31.98%) and a variant of 20 mg/L mixture of HFA (12.59%).

Cornell University 2014 Wine is subjected to many sources of microbial contamination throughout the wine making process, including but not limited to fermentation, barrel maturation, bottling, etc. In wineries, sanitation protocols should consider not only the type of microorganisms that need to be challenged, but also the type of surface that is going to be sanitized, since contact surfaces need to be treated differently according to physical and chemical properties. In the past, chlorinated compounds were used as sanitizers in wine industry, however we now know that they can be involved in the formation of trichloroanisoles (TCA), resulting in wine defects. Chlorine dioxide unlike other chlorinated compounds does not form TCA, or at least at very low levels. However, this research concluded its poor efficacy to sanitize wine barrels, likely due to the organic nature of the barrels. Alternative sanitizers in wine industry also include: sulfur dioxide, peroxyacetic acid, hot water, steam, ozone, etc. On the other hand, Velcorin ® (Dimethyl dicarbonate or DMDC) is currently used as a wine sterilant, however, due to its high disinfection effectiveness against yeast, we investigated its potential as a sanitizer for wine contact surfaces.

Ultraviolet radiation (UV) was used to control patulin in apple juice from concentrate (UV exposure from 14.2 to 99.4 mJ/cm2) and kinetic degradation models between apple cider and apple juice from concentrate were compared. UV radiation was shown to effectively reduce patulin with minimal changes in physicochemical properties and sensory characteristics of apple juice from concentrate. Patulin reduction by UV radiation in apple juice from concentrate followed first-order kinetics. Dimethyl dicarbonate (DMDC) with 0, 50, 125, and 250 ppm, and papain and bromelain (0, 100, and 1000 ppm) were used to investigate the inhibitory effect of vegetative cells and spores of Alicyclobacillus acidoterrestris strains VF, WAC, and SAC in potato dextrose broth (PDB), apple juice, and orange juice. DMDC showed a significant reduction in vegetative cells and spores of Alicyclobacillus in all samples, with no changes in juice quality, whereas, papain and bromelain showed antimicrobial activity only against vegetative cells of all strains with all samples, and minimal alterations in juice quality and sensory attributes.

    15 December 2010 [25-10] APPLICATION A1025 CLASSIFICATION OF DIMETHYL DICARBONATE APPROVAL REPORT Executive Summary Purpose Food Standards Australia New Zealand (FSANZ) received an Application (A1025 - Classification of Dimethyl Dicarbonate) from Lanxess Deutschland GmbH (Germany, formerly Bayer Chemicals AG) and Victus International, on 3 June 2009. Lanxess is the manufacturer of Dimethyl Dicarbonate (DMDC) (brand name Velcorin®) and Victus International is the Australian distributor of Velcorin®. Application A1025 seeks a reassessment of the regulatory classification of DMDC. DMDC is currently listed as a food additive for use in some beverages in Standard 1.3.1 – Food Additives in the Australia New Zealand Food Standards Code (the Code). The Application states that DMDC is currently regulated as a food additive despite having a mode of action which is more consistent with the definition of a processing aid under Standard 1.3.3 – Processing Aids. The Applicant asserts that DMDC is neither present nor technologically functional in the food as sold. Further, that by classifying DMDC as a food additive and therefore requiring it to be labelled as a preservative in the statement of ingredients, the Code does not reflect that DMDC has no ongoing antimicrobial function in the finished product. This assessment has considered the technological function and the mode of action of DMDC and its appropriate classification in the Code. This Application was assessed under the General Procedure. Risk and Technical Assessment A safety assessment of DMDC and its breakdown products was undertaken by the then Australia New Zealand Food Authority (ANZFA) in 1996, with no public health and safety issues identified.

    TRIOLI, MICROBIAL CONTAMINATION IN WINE, P. 1 MICROBIAL CONTAMINATION IN WINE Gianni TRIOLI Vinidea, Italy Extract from Technical Notes of Code of Best Practice for Organic Winemaking, produced under the EU FP6 STREP project ORWINE Microbial contamination or spoilage occurs with the development of micro-organisms whose metabolism can negatively affect wine quality. Grape juice, rich in sugars and nutrients, is a suitable substrate for the growth of many species of micro-organisms, including yeasts, bacteria and moulds. After alcoholic fermentation, the ethanol presence reduces the potential for development of many micro-organisms but even under final wine conditions some yeasts and bacteria can still be active. SPOILAGE AGENTS The low pH of juice and wine does not permit the growth of human pathogens which are thus not a concern in the wine industry. Many micro-organisms can however adversely affect wine quality by producing unwanted chemicals resulting from the degradation of favourable ones. Oxidative yeasts This group includes yeasts from the genera Hansaenula, Hanseniaspora, Pichia, Candida. These yeasts have a predominant oxidative metabolism, but some species can survive quite high levels of alcohol. They can metabolise sugars and organic acids in the presence of oxygen. Unwanted by-products resulting from this activity are acetic acid, ethyl-acetate and acetaldehyde, together with many other compounds whose high presence can initiate faults and off flavours in wine. Oxidative yeasts are found on grapes, in the juices and in the wine. Apiculata yeasts The name of these yeasts refers to the lemon-shaped appearance of Kloeckera apiculata . This yeast is predominant in grape juice before the complete onset of alcoholic fermentation and can growth fast at low temperatures.
