Kingsley Donkor, Ken Wagner, Julian Davies and Laura Lamb Abstract The discovery of new and more effective antibiotics continues to be a priority given the frequency of the emerging multi-drug resistant pathogenic microorganisms. Thus, scientists are searching for new antibiotics from microorganisms selected from extreme habitats such as very old caves. Various cave bacteria species were isolated and could be sources of new antibiotics. The objective of our work is to isolate cave bacteria from a volcanic cave in Wells Gray Provincial Park in BC and to test if they produce metabolites with antimicrobial activity against some microorganisms including multi- drug resistant pathogens. This study used 16 cave strains previously isolated and screened against a panel of microorganisms including drug resistant pathogens. Upon retesting, 4 out of 16 cave bacterial isolates, RA001, RA003, RA004, and PM58B-RA, demonstrated antimicrobial activity against Mycobacterium smegmatis, Micrococcus luteus, Acinetobacter baumannii and MDR-Staphylococcus aureus. To study the conditions for best growth and antimicrobial production, these four bacteria were cultured in different fermentation media (namely Hickey-Tresner, R2A, V-8 juice and ISP-2) and incubated at 12 and 25°C for 14 days. During the course of fermentation, the percentage of packed cell volume (%PCV), antimicrobial activity and pH were observed and recorded daily. It was found that each of the bacteria demonstrated antimicrobial activity against different microorganisms at various times of fermentation and temperature. Overall, R2A broth 1 medium and the lower temperature of 12°C appear to be best for antimicrobial production by the cave bacteria used in this study. Isolation and purification of the antimicrobial compounds produced by these isolates is under investigation using the best growth conditions determined in this study. We identified these bacteria using chemotaxonomic studies; 16S rRNA sequencing and Matrix-assisted laser desorption/ionization-time of flight (MALDI-TOF), all isolates were identified to the species level. PM58B was found to be Bacillus licheniformis, RA001, and RA004 were identified as Arthrobacter agilis that may be of different variants. While RA003 was identified as Sphingopyxis terrae. Active compounds from RA003 fermentation broth were further studied by extraction and purification. In conclusion, cave bacteria are promising sources of potential novel antimicrobial compounds. Isolation, optimization of screening, growth media and conditions of cave bacteria may be useful in the discovery of new antimicrobial drugs. Additionally, the knowledge obtained from this study with respect to cave bacteria and their roles in cave formation and degradation will add to existing information on cave conservation. 2 Table of Therefore, our research has been aimed to fulfil this lack of knowledge and improve the understanding of functioning of edaphic bacterial communities of the Central Mojave Desert soil. We characterized aerobic heterotrophic soil bacterial communities of the central region of the Mojave Desert. A high total number of prokaryotic cells and a high proportion of culturable forms in the soil studied were observed. Prevalence of Actinobacteria, Proteobacteria, and Firmicutes was discovered. The dominance of pigmented strains in culturable communities and high proportion of thermotolerant and pH-tolerant bacteria were detected. Resistance to a number of salts, including the ones found in Martian regolith, as well as antibiotic resistance, were also estimated.

Contrary to human probiotics, they belong to many different phyla and have usually had each genus analysed independently, which has resulted in lack of a complete taxonomic analysis as a group. This review scrutinizes the plant probiotic concept, and the taxonomic status of plant probiotic bacteria, based on both traditional and more recent approaches. Phylogenomic studies and genes with implications in plant-beneficial effects are discussed. This report covers some representative probiotic bacteria of the phylum Proteobacteria, Actinobacteria, Firmicutes and Bacteroidetes, but also includes minor representatives and less studied groups within these phyla which have been identified as plant probiotics. In this study, we compared the genomes of sixteen strains within the Arthrobacter group, including strain PAMC25564, to identify genomic features that adapt and survive life in the cold environment. Results: The genome of Arthrobacter sp. PAMC25564 comprised 4,170,970 bp with 66.74 % GC content, a predicted genomic island, and 3,829 genes. This study provides an insight into the redundancy of CAZymes for potential cold adaptation and suggests that the isolate has glycogen, trehalose, and maltodextrin pathways associated to CAZyme genes. This strain can utilize polysaccharide or carbohydrate degradation as a source of energy. Moreover, this study provides a foundation on which to understand how the Arthrobacter strain produces energy in an extreme environment, and the genetic pattern analysis of CAZymes in cold-adapted bacteria can help to determine how bacteria adapt and survive in such environments.

In addition, a collection of bacterial strains from wasted gums was set, and the biodegradation capability of diferent gum ingredients by the isolates was tested. Our results reveal that the oral microbiota present in gums after being chewed, characterised by the presence of species such as Streptococcus spp. or Corynebacterium spp., evolves in a few weeks to an environmental bacteriome characterised by the presence of Acinetobacter spp., Sphingomonas spp. and Pseudomonas spp. Wasted chewing gums collected worldwide contain a typical sub-aerial bioflm bacteriome, characterised by species such as Sphingomonas spp., Kocuria spp., Deinococcus spp. and Blastococcus spp. Our fndings have implications for a wide range of disciplines, including forensics, contagious disease control, or bioremediation of wasted chewing gum residues. Chewing gums may have been used for thousands of years, since wood tar from the Mesolithic and Neolithic periods have been found with tooth impressions, which suggests a role in teeth cleaning as well as its usage as early adhesives1, 2. Te frst modern chewing gum was introduced in the market in the late 19th 3 and chewing gums are today vastly consumed worldwide: it is estimated that Iran and Saudi Arabia are the countries with the highest chewing gum consumption, where 80% of the population are regular chewing gum consumers4. Moreo- ver, global online surveys on gum intake conducted in Europe5 and United States6 displayed similar chewing gum patterns among them, where more than 60% of adolescents and adults had chewed gums in the last 6 months before the survey and the mean intakes ranged from 1 to 4 pieces of chewing gum per day.

Due to their animal origin, parchments easily undergo biodeterioration: the most common biological damage is characterized by isolated or coalescent purple spots, that often lead to the detachment of the superfcial layer and the consequent loss of written content. Although many parchments with purple spot biodegradative features were studied, no common causative agent had been identifed so far. In a previous study a successional model has been proposed, basing on the multidisciplinary analysis of damaged versus undamaged samples from a moderately damaged document. Although no specifc sequences were observed, the results pointed to Halobacterium salinarum as the starting actor of the succession. In this study, to further investigate this topic, three dramatically damaged parchments were analysed; belonging to a collection archived as Faldone Patrizi A 19, and dated back XVI-XVII century A.D. With the same multidisciplinary approach, the Next Generation Sequencing (NGS, Illumina platform) revealed DNA sequences belonging to Halobacterium salinarum; the RAMAN spectroscopy identifed the pigment within the purple spots as haloarchaeal bacterioruberin and bacteriorhodopsine, and the LTA technique quantifed the extremely damaged collagen structures through the entire parchments, due to the biological attack to the parchment frame structures.

Box 59, 1790 AB Den Burg, The Netherlands 1 Summary Bacterial populations inhabiting the sea surface microlayer from two contrasted Mediterranean coastal stations (polluted vs. oligotrophic) were examined by culturig and genetic fingerprinting methods and were compared with those of underlying waters (50 cm depth), for a period of two years. More than 30 samples were examined and 487 strains were isolated and screened. Proteobacteria were consistently more abundant in the collection from the pristine environment whereas Gram-positive bacteria (i.e., Actinobacteria and Firmicutes) were more abundant in the polluted site. Cythophaga-Flavobacter–Bacteroides (CFB) ranged from 8% to 16% of total strains. Overall, 22.5% of the strains showed a 16S rRNA gene sequence similarity only at the genus level with previously reported bacterial species and around 10.5% of the strains showed similarities in 16S rRNA sequence below 93% with reported species. The CFB group contained the highest proportion of unknown species, but these also included Alpha- and Gammaproteobacteria. Such low similarity values showed that we were able to culture new marine genera and possibly new families, indicating that the sea-surface layer is a poorly understood microbial environment and may represent a natural source of new microorganisms.

However, their survival mechanisms in cold regions such as Antarctica are not yet fully understood. In this study, we compared the genomes of 16 strains within the Arthrobacter group, including strain PAMC25564, to identify genomic features that help it to survive in the cold environment. Results: Using 16 S rRNA sequence analysis, we found and identified a species of Arthrobacter isolated from cryoconite. We designated it as strain PAMC25564 and elucidated its complete genome sequence. The genome of PAMC25564 is composed of a circular chromosome of 4,170,970 bp with a GC content of 66.74 % and is predicted to include 3,829 genes of which 3,613 are protein coding, 147 are pseudogenes, 15 are rRNA coding, and 51 are tRNA coding. In addition, we provide insight into the redundancy of the genes using comparative genomics and suggest that PAMC25564 has glycogen and trehalose metabolism pathways (biosynthesis and degradation) associated with carbohydrate active enzyme (CAZymes). We also explain how the PAMC26654 produces energy in an extreme environment, wherein it utilizes polysaccharide or carbohydrate degradation as a source of energy.
