Testosterone undecanoate (TU), an orally administered ester prodrug of testosterone, is significantly subjected to first-pass metabolism. However, the individual contribution of intestinal and hepatic first-pass metabolism is not well determined. Therefore, the aim of the current study was to predict the metabolic contribution of each site. The hydrolysis–time profiles of TU incubation in human liver microsomes and Caco-2 cell homogenate were used to predict hepatic and intestinal first-pass metabolism, respectively. The in vitro half-life (t1/2 inv) for the hydrolysis of TU in microsomal mixtures was 28.31 3.51 min. By applying the “well-stirred” model, the fraction ± of TU that could escape hepatic first-pass metabolism (F ) was predicted as 0.915 0.009. -

It is the fourth most common cause of infection. Clotrimazole, a broad spectrum imidazole antifungal agent is widely used to treat fungal infections. Conventional topical formulations of clotrimazole are intended to treat infections by effective penetration of drugs into the stratum corneum. However, drawbacks such as poor dermal bioavailability, poor penetration, and variable drug levels limit the efficiency. The present study aims to load clotrimazole into ufosomes and evaluate its topical bioavailability. Clotrimazole loaded ufosomes were prepared using cholesterol and sodium oleate by thin film hydration technique and evaluated for size, polydispersity index, and entrapment efficiency to obtain optimized formulation. -

NEW ZEALAND DATA SHEET 1. PRODUCT NAME MIRENA 52 mg intrauterine contraceptive device (release rate: 20 microgram/24 hours) 2. QUALITATIVE AND QUANTITATIVE COMPOSITION MIRENA is an intrauterine system (IUS) containing 52 mg levonorgestrel. For details of release rates, see Section 5.2. For the full list of excipients, see Section 6.1. 3. PHARMACEUTICAL FORM MIRENA consists of a white or almost white drug core covered with an opaque membrane, which is mounted on the vertical stem of a T-body. The vertical stem of the levonorgestrel intrauterine system is loaded in the insertion tube at the tip of the inserter. Inserter components are an insertion tube, plunger, flange, body and slider. The white T-body has a loop at one end of the vertical stem and two horizontal arms at the other end. Brown coloured removal threads are attached to the loop. The T-body of MIRENA contains barium sulfate, which makes it visible in X-ray examination. The IUS and inserter are essentially free from visible impurities. 4. Therefore, the therapeutic standard dose of a drug, which is based on trails in healthy volunteers and patients, is not suitable for every patient. Variability exists in both pharmacokinetics and pharmacodynamics. For a typical drug, one standard deviation in the values observed for bioavailability (F), clearance (CL) and volume of distribution (Vd) would be about 20%, 50% and 30% respectively. Therefore, 95% of the time, the average concentration (Cav) will be between 35% and 270% of the target value.
