at MAFFs Fisheries Laboratory at Conwy, North Wales A general introductory section on marine algae and their uses is followed by an explanation of factors affecting their growth, general maintenance, types of culture and examples of the small-scale batch culture methods and large-scale continuous and semi-continuous methods developed at Conwy D J Garrod Director Ministry of Agriculture, Fisheries and Food Directorate of Fisheries Research Laboratory Leaflet (67) 6 1 INTRODUCTION Unicellular, marine algae are widely used as food in the hatchery production of commercially valuable fish and shellfish Oysters and clams feed by filtering them from sea water Rotifers and brine shrimps also ingest algae, and are then themselves used as food for larval fish and prawns In some systems, algae are added to the water containing fish or prawns to improve the quality, but the way in which this works is not yet fully understood Marine algae are simple-celled plants and, like all plants, contain chlorophyll, which traps the energy from light and uses it to convert nutrients and carbon dioxide dissolved in the sea water into organic growth In the laboratory or hatchery, a collection of algal cells, which are growing and dividing, is known as a culture Of the very many types of algae which live in the sea, only a few can be cultured, and only certain types will give good growth when fed to oysters, clams, rotifers, brine shrimps, etc Some of the types commonly used as foods are listed in Table 1 They comprise two groups the flagellates, which can swim by the action of one or more flagellae, and the diatoms, which have an outer shell composed of silica, but can nevertheless remain buoyant Individual species of algae can be used as food, but feeding mixed diets, consisting of at least one type of diatom and one type of flagellate, nearly always gives much better growth Examples of some of these algae are shown in Figure 1 Chaetoceros calcitrans is one of the more useful diatoms which, because of its small size, is especially suitable for feeding to larval stages of marine animals However, it is not very easy to grow in large quantities Skeletonema costatum and of these algae are kept at the Fisheries Laboratory, Conwy' Samples can usually be supplied for a small handling charge' Laboratory Leaflet (67) 7 Similarly, with the flagellates, the small Isochrysis galbana and Pseudoisochrysis paradoxa are very good foods but difficult to culture Tetraselmis suecica is much easier to grow and is often used to feed larger animals Marine algae can be observed using a microscope with a magnification of x100 to x400 When feeding algae to animals, it is important to know the concentration of cells in the culture, and this can be determined using an haemocytometer (Appendix 1) Laboratory Leaflet (67) 8 2 CULTURE CONDITIONS NECESSARY FOR GROWTH OF MARINE ALGAE 21 Light This is normally provided by fluorescent lamps The most commonly used types are cool-white or daylight Increasing the light intensity usually means better growth and faster division of algal cells and, therefore, the production of more food It is important to make the most efficient use of the artificial light as lamps also generate heat and may make the culture too hot The indoor culture systems described in Section 5 of this leaflet are all designed to use light efficiently Cultures of algae can also be grown outdoors, using natural daylight, and examples of this method are also given in Section 5 22 Heat Most types of algae grow well at temperatures from 17oC to 22oC Lower temperatures will not usually kill the algae, but will reduce their growth rate Above 27oC, most types of algae will die If necessary, cultures can be cooled by a flow of cold water over the surface of the culture vessel or by controlling the air temperature with refrigerated air conditioning units 23 Nutrients Nutrients are the inorganic salts required for plant growth For algae culture, it is convenient to make up strong standard solutions which, in appropriate dilutions in sea water, provide the culture medium Recipes for the nutrient salt solutions, used for culture of algae at MAFFs Fisheries Laboratory at Conwy are given in Table 2 24 Mixing By mixing, the necessary light and nutrients become available to all of the cells Algae cultures are usually mixed by bubbling air through them This can be supplied from a compressor or via an air blower, and will also act as a carrier gas for carbon dioxide Cultures may also be mixed by mechanical means using, for example, stirrers 25Carbon dioxide Providing the algae with extra carbon, in the form of the gas carbon dioxide (CO ), will give much faster growth CO is supplied from compressed gas cylinders, and only a very little is2 needed (about half of one percent)2 in the air supplied to the culture The CO should be passed through a flow meter to ensure that the amount used will keep the pH of the culture2 between 7 8 and 8 0 The pH can be checked with indicator papers, which change colour with a change in pH, or with a pH meter Both the air and the CO should be filtered through an in-line filter unit of 0 3 microns to 0 5 microns before entering the culture,2 as this helps to prevent other, possibly contaminating, organisms from getting into the cultures 26 Salinity Salinities of between 25 and 30 psu (practical salinity units) (UNESCO, 1981)* are generally best for the culture of flagellates, and between 20 and 25 psu for the culture of diatoms These salinities can be obtained by diluting sea water with tap water Salinity can be measured with an hydrometer or a refractometer *UNESCO, 1981' The practical salinity scale 1978 and the international equation of state of     Proper Use of a Bunsen Burner If using a Bunsen burner in a biological safety cabinet, see: http://research.wayne.edu/oehs/pdf/gas-in-bsc.pdf Prior to Lighting Bunsen Burner: • Know the location of the laboratory’s main gas shut off valve and fire extinguisher. • Place the Bunsen burner away from any overhead shelving, equipment, or light fixtures by at least 12 inches. • Remove all papers, notebooks, combustible materials, and excess chemicals from the area. • Tie-back any long hair, dangling jewelry, or loose clothing. • Use correct tubing (butyl rubber hose). Inspect hose for cracks, holes, pinch points, or any defect and replace if necessary. Ensure that the hose fits securely on the gas valve and the burner. • Notify others in the lab that the burner will be in use. Lighting Bunsen Burner: • Before turning on gas, have a sparker/lighter with extended nozzle to ignite burner. Never use matches. • Check connections to burner and desk outlet valve. • Close needle valve and collar. • Open gas valve fully. • Open needle valve 1/2 turn. • Use the spark/lighter to light the flame. • Adjust flame by turning the collar to regulate air flow and produce an appropriate flame for the experiment (typically a medium blue flame). During Use: • Never leave a lighted burner unattended. Drafts (including those created by a hood) may extinguish the flame, but the gas will still be on. This could result in an explosion. • Never face the vessel (test tube, beaker, etc.) being used to heat a material toward yourself or others near the burner. In some experiments, a hot plate may be used in place of the Bunsen burner.

Learning basic microbiology lab techniques and procedures is interesting and relevant to most students—many of them, after all, have had at least one throat culture or other microbiology lab test in their lives. The requirements for safe microbiological work in the classroom include the knowledge and practice of aseptic technique and sterilization procedures. Standard Microbiology Practices The federal government has outlined general safety standards for microbiological work in their publication Biosafety in Microbiological and Biomedical Laboratories. Biosafety Level I (BSL 1) practices described in this book are appropriate for secondary educational teaching laboratories. BSL 1 work involves using strains of viable, nonpathogenic microorganisms that are known not to cause disease in healthy adult humans and are thus of minimal potential hazard to students, teachers or the environ- ment. The level of containment in BSL 1 relies on standard microbiological practices in laboratory settings. Students and lab per- sonnel should receive specific training in the procedures conducted in the laboratory. Standard microbiological practices include: • Wash hands thoroughly with soap and water before and after handling viable materials, after performing microbiological transfers and observations or working with potentially hazardous materials, and before leaving the laboratory. • Eating, drinking, smoking, handling of contact lenses, applying cosmetics, and storing food for human use are not permit- ted in laboratory work areas. • Pipetting by mouth is prohibited—use only mechanical pipetting devices. • There should be policies and procedures for safe disposal of sharps, including broken glassware.

    KNOW YOUR LAB EQUIPMENTS Test tube A test tube, also known as a sample tube, is a common piece of laboratory glassware consisting of a finger-like length of glass or clear plastic tubing, open at the top and closed at the bottom. Beakers Beakers are used as containers. They are available in a variety of sizes. Although they often possess volume markings, these are only rough estimates of the liquid volume. The markings are not necessarily accurate. Erlenmeyer flask Erlenmeyer flasks are often used as reaction vessels, particularly in titrations. As with beakers, the volume markings should not be considered accurate. Volumetric flask Volumetric flasks are used to measure and store solutions with a high degree of accuracy. These flasks generally possess a marking near the top that indicates the level at which the volume of the liquid is equal to the volume written on the outside of the flask. These devices are often used when solutions containing dissolved solids of known concentration are needed. Graduated cylinder Graduated cylinders are used to transfer liquids with a moderate degree of accuracy. Pipette Pipettes are used for transferring liquids with a fixed volume and quantity of liquid must be known to a high degree of accuracy. Graduated pipette These Pipettes are calibrated in the factory to release the desired quantity of liquid. Disposable pipette Disposable transfer. These Pipettes are made of plastic and are useful for transferring liquids dropwise. Burette Burettes are devices used typically in analytical, quantitative chemistry applications for measuring liquid solution. Differing from a pipette since the sample quantity delivered is changeable, graduated Burettes are used heavily in titration experiments.

However, software narrowly designed to automate traditional hand-pipetting protocols often struggles to harness the full capaBilities of roBotic manipulation. Here we present Pyhamilton, an open-source Python package that eliminates these constraints, enabling experiments that could never be done by hand. We used Pyhamilton to double the speed of automated bacterial assays over current software and execute complex pipetting patterns to simulate population dynamics. Next, we incorporated feedBack-control to maintain hundreds of remotely monitored Bacterial cultures in log-phase growth without user intervention. Finally, we applied these capaBilities to comprehensively optimize Bioreactor protein production By maintaining and monitoring fluorescent protein expression of nearly 500 different continuous cultures to explore the carBon, nitrogen, and phosphorus fitness landscape.

Put drop of water onto glass slide, put piece of onion skin onto drop of water on slide 3. Lower cover slip over the onion skin (protects microscope lens) 4.
