Voglmayr1,2 Key words Abstract Fresh collections and their ascospore and conidial isolates backed up by type studies and molecular phylogenetic analyses of a multigene matrix of partial nuSSU-, complete ITS, partial LSU rDNA, rpb2, tef1 and tub2 Cucurbitaria sequences were used to evaluate the boundaries and species composition of Fenestella and related genera of the Dothideomycetes Cucurbitariaceae. Eight species, of which five are new, are recognised in Fenestella s.str., 13 in Parafenestella with multigene phylogenetic analysis eight new species and two in the new genus Synfenestella with one new species. Cucurbitaria crataegi is combined new taxa in Fenestella, C. sorbi in Synfenestella, Fenestella faberi and Thyridium salicis in Parafenestella. Cucurbitaria Phoma subcaespitosa is distinct from C. sorbi and combined in Neocucurbitaria. Fenestella minor is a synonym of Valsa Pleosporales tetratrupha, which is combined in Parafenestella. Cucurbitaria marchica is synonymous with Parafenestella salicis, Pyrenochaeta Fenestella bavarica with S. sorbi, F. macrospora with F. media, and P. mackenziei is synonymous with P. faberi, and the latter is lectotypified. Cucurbitaria sorbi, C. subcaespitosa and Fenestella macrospora are lecto- and epitypified, Cucurbitaria crataegi, Fenestella media, F. minor and Valsa tetratrupha are epitypified in order to stabilise the names in their phylogenetic positions. A neotype is proposed for Thyridium salicis. A determinative key to species is given. Asexual morphs of fenestelloid fungi are phoma-like and do not differ from those of other representatives of the Cucurbitariaceae. -

Jones, MD ABSTRACT Purpose: To determine the risk factors and clinical signs of Curvularia keratitis and to evaluate the management and out- come of this corneal phæohyphomycosis. Methods: We reviewed clinical and laboratory records from 1970 to 1999 to identify patients treated at our institution for culture-proven Curvularia keratitis. Descriptive statistics and regression models were used to identify variables associ- ated with the length of antifungal therapy and with visual outcome. In vitro susceptibilities were compared to the clini- cal results obtained with topical natamycin. Results: During the 30-year period, our laboratory isolated and identified Curvularia from 43 patients with keratitis, of whom 32 individuals were treated and followed up at our institute and whose data were analyzed. Trauma, usually with plants or dirt, was the risk factor in one half; and 69% occurred during the hot, humid summer months along the US Gulf Coast. Presenting signs varied from superficial, feathery infiltrates of the central cornea to suppurative ulceration of the peripheral cornea. A hypopyon was unusual, occurring in only 4 (12%) of the eyes but indicated a significantly (P = .01) increased risk of subsequent complications. The sensitivity of stained smears of corneal scrapings was 78%. Curvularia could be detected by a panfungal polymerase chain reaction. Fungi were detected on blood or chocolate agar at or before the time that growth occurred on Sabouraud agar or in brain-heart infusion in 83% of cases, although colonies appeared only on the fungal media from the remaining 4 sets of specimens. -

Crous1,4,5,6 Key words Abstract The family Stachybotriaceae was recently introduced to include the genera Myrothecium, Peethambara and Stachybotrys. Members of this family include important plant and human pathogens, as well as several spe- biodegraders cies used in industrial and commercial applications as biodegraders and biocontrol agents. However, the generic generic concept boundaries in Stachybotriaceae are still poorly defined, as type material and sequence data are not readily avail- human and plant pathogens able for taxonomic studies. To address this issue, we performed multi-locus phylogenetic analyses using partial indoor mycobiota gene sequences of the 28S large subunit (LSU), the internal transcribed spacer regions and intervening 5.8S multi-gene phylogeny nrRNA (ITS), the RNA polymerase II second largest subunit (rpb2), calmodulin (cmdA), translation elongation species concept factor 1-alpha (tef1) and β-tubulin (tub2) for all available type and authentic strains. Supported by morphological taxonomy characters these data resolved 33 genera in the Stachybotriaceae. These included the nine already established genera Albosynnema, Alfaria, Didymostilbe, Myrothecium, Parasarcopodium, Peethambara, Septomyrothecium, Stachybotrys and Xepicula. At the same time the generic names Melanopsamma, Memnoniella and Virgatospora were resurrected. Phylogenetic inference further showed that both the genera Myrothecium and Stachybotrys are polyphyletic resulting in the introduction of 13 new genera with myrothecium-like morphology and eight new genera with stachybotrys-like morphology. -

The first patient presented for investigation of a lesion at the right hilum on chest radiograph and a raised blood eosinophil count. A bronchogram showed complete block of the apical segmental bronchus which at operation was shown to be caused by inspissated material. The second patient was investigated because of a cough productive of plugs of sputum and irregular opacities in both upper zones on chest radiograph and a raised blood eosinophil count. This only cleared after one month on high dose oral prednisone therapy. The third patient with a previous history ofleft lingular pneumonia and bronchiectasis ofthe lingular segment ofthe left upper lobe was investigated three years later for right basal shadowing and a raised blood eosinophil count. The radio- graph cleared after one month on high dose oral prednisone treatment. The aetiological agents in these cases were dematiaceous hyphomycetes, fungi ubiquitous in nature, and also agents of plant disease. The causal fungi, Curvularia hlnata and Drechslera hawaiiensis, have on a few occasions been reported as causing human disease but in cases quite dissimilar to the three reported here. Septate branching dematiaceous mycelium was consistently seen in the clinical material and isolated from http://thorax.bmj.com/ successive sputum specimens from each patient. -

Abstract The quality of raising seedlings in a perennial crop like coffee may be affected by several abiotic and biotic factors. In India, coffee seedlings are affected by three different diseases in the nursery viz., collar rot, brown eye spot, stem necrosis and leaf spot. The stem necrosis and leaf spot disease caused by the fungus Myrothecium roridum Tode ex Fr. is posing a serious problem in coffee nurseries particularly during rainy period of July and August months. The present study was under taken with a fixed plot survey to assess the distribution, incidence and severity of stem necrosis and leaf spot disease in major coffee growing taluks of Chikmagalur district in the year 2016 and 2017. Out of 22 coffee nurseries surveyed in four major coffee growing taluks of Chikmagalur district, the survey results (pooled data analysis of two years 2016 & 2017) indicated that maximum leaf spot incidence (23.98%) was recorded on Chandragiri cultivar of arabica coffee in Koppa taluk and minimum incidence (16.40%) in Mudigere taluk on C×R cultivar of robusta coffee. Maximum leaf spot severity (30.34%) was recorded on Chandragiri in Chikmagalur taluk and minimum severity (14.87%) in Koppa taluk on C×R. -

Lluís Companys 23, 08010 Barcelona, Spain ABSTRACT Fungi are a highly diverse group of heterotrophic eukaryotes characterized by the absence of phagotrophy and the presence of a chitinous cell wall. While unicellular fungi are far from rare, part of the evolutionary success of the group resides in their ability to grow indefinitely as a cylindrical multinucleated cell (hypha). Armed with these morphological traits and with an extremely high metabolical diversity, fungi have conquered numerous ecological niches and have shaped a whole world of interactions with other living organisms. Herein we survey the main evolutionary and ecological processes that have guided fungal diversity. We will first review the ecology and evolution of the zoosporic lineages and the process of terrestrialization, as one of the major evolutionary transitions in this kingdom. Several plausible scenarios have been proposed for fungal terrestralization and we here propose a new scenario, which considers icy environments as a transitory niche between water and emerged land. We then focus on exploring the main ecological relationships of Fungi with other organisms (other fungi, protozoans, animals and plants), as well as the origin of adaptations to certain specialized ecological niches within the group (lichens, black fungi and yeasts). -

He described Myrothecium as a cup shaped fungus with spores becoming slowly viscous and included five species in the genus: M. roridum, M. inundatum, M. stercoreum, M. hispidum and M. dubium. None of his original material remains. In 1803, according to Fries (1829), Schumacher published a sixth species, M. scybalorum. Albertini & Schweinitz (1805) described a species Peziza verrucaria with green viscous spores and a white margin to the fructification, noting its resemblance *Nie Fitton to Myrothecium. A single ascospore isolate was obtained and formed the asexual morph in culture. ITS, LSU, RPB2 and TEF1α gene regions were sequenced and analyzed with molecular data from related taxa. In a phylogenetic analysis the new isolate clustered with Leptosphaerulina americana, L. arachidicola, L. australis and L. trifolii (Didymellaceae) and the morphology was also comparable with Leptosphaerulina species. Leptosphaerulina saccharicola is introduced to accommodate this new collection which is morphologically and phylogenetically distinct from other species of Leptosphaerulina. A detailed description and illustration is provided for the new species, which is compared with similar taxa. The type specimen of Pleosphaerulina oryzae, is transferred to Leptosphaerulina. It is redescribed and is a distinct species from L. australis, with which it was formerly synonymized. Leptosphaerulina species have been linked to Pithomyces but the lack of phylogenetic support for this link is discussed. -

Stoneville MS 38776, USA; [email protected] Received: 15 February 2008 / Accepted: 16 December 2008 © 2009 Wageningen Academic Publishers Abstract Myrothecium verrucaria is a pathogen of several invasive weed species, including kudzu, and is currently being evaluated for use as a bioherbicide. However, the fungus also produces macrocyclic trichothecene mycotoxins. The safety of this biological control agent during production and handling would be improved if an inoculum could be produced without concomitant accumulation of macrocyclic trichothecenes. Sporulation and trichothecene production by M. verrucaria was evaluated on standard potato dextrose agar (PDA) and a series of complex and defined media. Sporulation on PDA and on agar media with nitrogen as ammonium nitrate or potassium nitrate was more than ten-fold greater then sporulation on the medium with ammonium sulphate as the nitrogen source. Accumulation of macrocyclic trichothecenes was strongly affected by the media composition, with higher levels often associated with higher carbon content in the media. Overall, incubation in continuous darkness resulted in higher macrocyclic trichothecene concentrations. Results support the hypothesis that accumulation of macrocyclic trichothecenes by this fungus can be altered by manipulating carbon and nitrogen sources. Furthermore, the biosynthesis of these mycotoxins may be independent of sporulation, demonstrating that the bioherbicide can be readily produced on solid substrates while simultaneously yielding conidia that are less threatening to worker safety. -

Abstract Aquatic hyphomycetes are a diverse, polyphyletic group of asexually reproducing fungi involved in the decomposition of litter in freshwater ecosystems. Curvularia eragrostidis, C. verruculosa and Phragmocephala atra were identified from sub- merged wood collected from freshwater streams in Yunnan Province, Southwestern China. They were characterised based on morphology and LSU, ITS and SSU sequence data. Phylogenetic analysis of LSU sequences placed the isolates within the order Pleosporales. Curvularia eragrostidis and C. verruculosa are reported from freshwater habitats for the first time. An epitype is designated for Curvularia verruculosa. This is the first phylogenetic placement of the genus Phragmocephala in the family Melanommataceae in Dothideomycetes, providing new DNA sequence data. -

Biology and recent developments in the systematics of Phoma, a complex genus of major quarantine significance. Fungal Diversity 31: 1-18. Species of the coelomycetous genus Phoma are ubiquitously present in the environment, and occupy numerous ecological niches. More than 220 species are currently recognised, but the actual number of taxa within this genus is probably much higher, as only a fraction of the thousands of species described in literature have been verified in vitro. For as long as the genus exists, identification has posed problems to taxonomists due to the asexual nature of most species, the high morphological variability in vivo, and the vague generic circumscription according to the Saccardoan system. In recent years the genus was revised in a series of papers by Gerhard Boerema and co-workers, using culturing techniques and morphological data. This resulted in an extensive handbook, the “Phoma Identification Manual” which was published in 2004. The present review discusses the taxonomic revision of Phoma and its teleomorphs, with a special focus on its molecular biology and papers published in the post-Boerema era.
