ÖZET Yüksek Lisans Tezi HELLEBORUS VESICARIUS (RANUNCULACEAE) BİTKİSİNİN İÇERDİĞİ UÇUCU YAĞLARIN TAYİNİ VE FARKLI EKSTRELERİNİN ANTİMİKROBİYEL AKTİVİTELERİ Tarık KARABULUT Harran Üniversitesi Fen Bilimleri Enstitüsü Kimya Anabilim Dalı Danışman : Yrd. Doç. Dr. Zafer UYAR Yıl:2014, Sayfa:56 Bu çalışmada Helleborus vesicarius (ranunculaceae) bitkisi kimyasal ve mikrobiyolojik olmak üzere iki alanda incelendi. Türü belirlendikten sonra Helleborus vesicarius (ranunculaceae) bitkisi güneş ışığından uzak kurutuldu ve sonra öğütüldü. Daha sonra bitki bileşenleri metanol ile ekstrakte edilerek ham metanol ekstresi hazırlandı. Daha sonra bir dizi seçici katı-sıvı ve sıvı-sıvı ekstraksiyon kullanılarak ham metanol ekstresi polaritesine göre sırasıyla heksan, diklorometan, bütanol ve su fraksiyonlarına ayrıldı. Bu ekstrelerden uçucu ve esansiyel yağ asitlerinin zengin olduğu Heksan ekstresi metillenerek GC-MS analizi yapıldı ve elde edilen sonuçlar faydalı yağ asitleri açısından irdelendi. Ayrıca tüm ekstrelerin (metanol, heksan, diklorometan, bütanol ve su) Salmonella thyphimurium CCM 5445, Escherichia coli ATCC 25922, Escherichia coli 0157H7 gram-negatif bakterilerine ve Staphylococcus aureus ATCC 25923, Staphylococcus epidermidis ATCC 12228, Enterococcus faecalis ATCC 29212, Enterococcus faecium DSM 13590 gram-pozitif bakterilerine karşı antibakteriyel aktiviteleri ve Candida albicans ATCC 10239 mayasına karşı antifungal aktiviteleri agar-disk difüzyon ve minimum inhibisyon konsantrasyonu tayin yöntemleriyle referans ilaçlarla karşılaştırılarak ve incelendi. After identified taxanomically, Helleborus vesicarius (ranunculaceae) plant was dried away from sunlight and then finely ground. Then the crude methanol extract was prepared by extraction of the plant components with methanol. Then crude methanol extract was separated into hexane, dichloromethane, butanol, and water fractions respectively based on increasing polarity using a series of selective solid-liquid and liquid-liquid extractions. Out of all extracts, Hexane extract, which is rich in essential fatty acids and volatile fatty acids, was methylated and its GC-MS analysis was performed and the results obtained were evaluated in terms of beneficial fatty acids. Also antibacterial activity against Salmonella thyphimurium CCM 5445, Escherichia coli ATCC 25922, Escherichia coli 0157H7 gram-negative bacteria and Staphylococcus aureus ATCC 25923, Staphylococcus epidermidis ATCC 12228, Enterococcus faecalis ATCC 29212, Enterococcus faecium DSM 13590 gram-pozitive bacteria and antifungal activity against Candida albicans ATCC 10239 yeast of all extracts (methanol, hexane, dichloromethane, butanol and water) were examined by agar- disk diffusion assay and minimum inhibitory concentration assay compared with the reference drugs.     Hellebore FAQs By Post Office Farm Nursery Can I grow Hellebores where I live? Hellebores are temperate climate plants. They like a cool to cold Winter and not too much humidity. So Hellebores can be grown anywhere in Tasmania and Victoria. Temperate areas of South Australia, including Adelaide and surrounds, are fine for Hellebores. In NSW Hellebores can be grown in coastal areas up to about Newcastle. Coastal areas further North than this will be too humid for Hellebores. So Hellebores can be grown in Sydney and will be particularly at home in the Southern Highlands and Blue Mountains. In inland areas of NSW Hellebores enjoy the cold Winters and can be grown well in non-arid areas from Wagga, Canberra through Bathurst, to Armidale. Planting Hellebores in the correct position in the garden is the key to growing them successfully. In the wild Hellebores are found in and around deciduous woodland. They are adapted to having shade for most or all of the day in Summer (when the trees are in leaf) and good light if not full sun over Winter (when the woodland canopy is bare). So while it is true that Hellebores are ‘shade plants’, it is deciduous rather than year round evergreen shade that they want. Indeed planting Hellebores under large evergreen shrubs and trees is the most common mistake made when planting Hellebores. If too dark under evergreen shade in Winter, Hellebore will not grow and flower to their potential.

Sowing species seed provides potential hybridizers with variable prog- eny from which can be selected potential parental individuals displaying desirable characteristics. In the case of hellebores, these characteristics include both floral and foliar features combined with overall vigor. SPECIES Helleborus argutifolius. Native southern Europe/Corsica. Handsome caules- cent specimen. A superb performer in the Pacific Northwest. Magnificent, variable foliage. The bold foliage creates an attractive contrast with the heavy effect of conifer foliage, i.e., Thuja. Cultivars: ‘Janet Starnes’, speckled cream leaves; ‘Pacific Frost’, heavy cream foli- age speckling. Helleborus foetidus. Native to central Europe. Caulescent species with handsome variable foliage and terminal clusters of pendulous bell-shaped flowers. Short lived but self-seeds freely. Often flowers in second year from seed—blooms late winter, early spring. Cultivars: ‘Sienna’, dark green foliage and chartreuse flowers. Selected by Will McLewin—noted English species authority; Wester Flisk Group, red-stemmed form from east Scotland (Seeding progeny quickly looses this desirable trait if grown with non-red-stemmed forms. When people tell stories about their gardens, many reveal that their most treasured plants are those they received as gifts. An outdoor plant for the avid gardener makes a great gift any time hardy plant with vivid colors is particularly special. Plants with rich foliage, berries and flowers can brighten a winter landscape year after year, as well as attracting bees and other insects to help with early spring pollination of other flowers. Here in the Skagit Valley, you may be surprised at the number of interesting winter plants from which we may choose. To find the right gift for the gardener on your list, your best choices will probably come from a local nursery where plants are kept outside. Flowering plants grown in greenhouses and shipped to a retailer may look great now, but they are often spring or summer-blooming varieties that will not add much to the garden next winter. In the nursery, start by looking for eye-catching foliage. Many small blueberry and azalea varieties, for example, have striking red bark and leaves in winter, as well as fruit or flowers during the warmer months. Boxwoods, firs and pines offer a range of green foliage and often are available as small plants this time of year. I know that the general definition of a drought is a lack of rain or moisture, which we get sometimes in the winter, ask mortimer....pg. 4 but it can also mean a drought of a different kind too. For some it is the actual lack of water for those plants under your eaves that don’t get the moisture from the winter New Hellebores rains, but for others it is the drought of things to do out- side in the garden. Those cold and dreary days leave a lot of gardeners in limbo. There is not much to do if you have already done your winter pruning, cleaning and spraying. There is also the drought of ideas. We look for things to got to have it....pg. 6 think about when it comes to gardening. Jumping from magazine to magazine, seed catalog to plant catalog, looking for ideas for the upcoming season. Still, after you Rain Barrels have read all the articles and circled all the pictures, what do you do with the rest of your time? Just like the drought in the summer, there are things you can do to combat this ‘dry’ period. Indoor plants are a start. Your gardening is not limited to the outside and summer.

    Plant in Focus, October 2017 Helleborus Hellebores Friends of GeelongBotanic Gardens Helleborus lividus subsp. corsicus, GBG May 2015 Helleborus, GBG June 2015 Introduction Commonly known as Hellebores, the Eurasian genus Helleborus consists of about 20 species of herbaceous or evergreen perennial flowering plants in the family Ranunculaceae. This family includes Buttercup Ranunculus, Anemone, Clematis, Delphinium and Aquilegia. Many species are poisonous. Despite names such as “Winter Rose", "Christmas Rose" and "Lenten Rose", Hellebores are not closely related to the Rose family Rosaceae. Various species of this genus originated in Europe and Asia. The greatest concentration of species occurs in the Balkans. The flowers have five petal-like sepals surrounding a ring of small, cup-like nectaries which are actually "petals" modified to hold nectar. The sepals do not fall as petals would, but remain on the plant, sometimes for many months. Thus the flowers remain attractive for a long time. Research in Spain suggests that the persistence of the sepals contributes to the development of the seeds. Nectaries form a whorl surrounding the stamens of Helleborus. The nectaries are modified petals. Many Hellebore species are in fact toxic, causing their own set of symptoms such as vertigo, swelling of the tongue and throat, tinnitus, and thirst. In extreme cases the poison can lead to slowing of of the heart, heart attack, stupor, and even death. It GeelongBotanic Gardens is for these reason that the Hellebore has been cloaked in a number of dark myths and folklore.
