USP25 Antibody (Pab) 21.10.2014USP25 antibody (pAb) Rabbit Anti -Human/M ouse Ubiquitin specific peptidase 25 (USP21) Instruction Manual Catalog Number PK-AB718-7765 Synonyms Ubiquitin specific peptidase 25, deubiquitinating enzyme 25, ubiquitin carboxyl-terminal hydrolase 25, ubiquitin thiolesterase 25, USP21, USP on chromosome 21 Description USP25 (ubiquitin specific peptidase 25), also known as USP21, belongs to the peptidase C19 family and is a highly conserved 76-amino acid protein involved in regulation of intracellular protein breakdown, cell cycle regulation, and stress response (1). It contains one UBA-like domain and two UIM (ubiquitin-interacting motif) repeats. Due to alternative splicing events, USP25 is expressed as two short, ubiquitously expressed isoforms and one long, muscle-specific isoform (2). The long isoform of USP25 (USP25m) is upregulated in myogenesis and is implicated in regulation of muscular differentiation and function. USP25 is a deubiquitinating enzyme (DUB) that negatively regulates IL-17-triggered signaling (3,4). Purification Method Affinity chromatography purified via peptide column. Clone / IgG Subtype Polyclonal antibody Species Reactivity Human, Mouse Specificity USP25 antibody is human and mouse reactive. Multiple isoforms of USP25 are known to exist. Formulation Antibody is supplied in PBS containing 0.02% sodium azide. Reconstitution During shipment, small volumes of antibody will occasionally become entrapped in the seal of the product vial. For products with volumes of 200 μl or less, we recommend gently tapping the vial on a hard surface or briefly centrifuging the vial in a tabletop centrifuge to dislodge any liquid in the container’s cap. Storage & Stability Antibody can be stored at 4ºC for three months and at -20°C for up to one year. As with all antibodies care should be taken to avoid repeated freeze thaw cycles. Antibodies should not be exposed to prolonged high temperatures. Applications E, WB |Note: Antibody might be suitable for other applications not tested so far. Optimal concentrations for each application have to be determined indivudually. USP25 antibody can be used for detection of USP25 by Western blot at 1 - 2 µg/ml. Images Available upon request. Trisomy 21, an extra copy of human chromosome 21, causes the majority of DS cases. After middle age, individuals with DS inevitably develop Alzheimer’s disease, the most common form of neurodegenerative diseases characterized by extracellular amyloid plaque deposition, intracellular neuritic fibrillary tangles and neuronal loss. The extracellular amyloid plaques are made of amyloid β (Aβ) proteins derived from β- and γ- cleavage of amyloid precursor protein (APP). The abnormal accumulation of Aβ proteins plays an essential role in AD pathogenesis. Ubiquitin-specific protease 25 (USP25) is a deubiquitinating enzyme that locates in the DS critical region of human chromosome 21.

ARDS occurs most often in the settings of pneumonia, sepsis, aspiration of gastric contents, or severe trauma. The prevalence of ARDS is approximately 10% in patients of intensive care. There is no effective remedy with mortality high at 30–40%. Most functional proteins are dynamic and stringently governed by ubiquitin proteasomal degradation. Protein ubiquitination is reversible, the covalently attached monoubiquitin or polyubiquitin moieties within the targeted protein can be removed by a group of enzymes called deubiquitinating enzymes (DUBs). Deubiquitination plays an important role in the pathobiology of ALI/ARDS as it regulates proteins critical in engagement of the alveolo-capillary barrier and in the inflammatory response. In this review, we provide an overview of how DUBs emerge in pathogen-induced pulmonary inflammation and related aspects in ALI/ARDS. Better understanding of deubiquitination-relatedsignaling may lead to novel therapeutic approaches by targeting specific elements of the deubiquitination pathways.
