While respiratory carbon oxidation pathways, electron transport, and ATP turnover are tightly coupled processes, AOX provides a means to relax this coupling, thus providing a degree of metabolic homeostasis to carbon and energy metabolism. Beside their role in primary metabolism, plant mitochondria also act as “signaling organelles”, able to influence processes such as nuclear gene expression. AOX activity can control the level of potential mitochondrial signaling molecules such as superoxide, nitric oxide and important redox couples. In this way, AOX also provides a degree of signaling homeostasis to the organelle. Evidence suggests that AOX function in metabolic and signaling homeostasis is particularly important during stress. These include abiotic stresses such as low temperature, drought, and nutrient deficiency, as well as biotic stresses such as bacterial infection. This review provides an introduction to the genetic and biochemical control of AOX respiration, as well as providing generalized examples of how AOX activity can provide metabolic and signaling homeostasis. -
Catalase and Oxidase Test
CATALASE TEST Catalase is the enzyme that breaks hydrogen peroxide (H 2O2) into H 2O and O 2. Hydrogen peroxide is often used as a topical disinfectant in wounds, and the bubbling that is seen is due to the evolution of O 2 gas. H 2O2 is a potent oxidizing agent that can wreak havoc in a cell; because of this, any cell that uses O 2 or can live in the presence of O 2 must have a way to get rid of the peroxide. One of those ways is to make catalase. PROCEDURE a. Place a small amount of growth from your culture onto a clean microscope slide. If using colonies from a blood agar plate, be very careful not to scrape up any of the blood agar— blood cells are catalase positive and any contaminating agar could give a false positive. b. Add a few drops of H 2O2 onto the smear. If needed, mix with a toothpick. DO NOT use a metal loop or needle with H 2O2; it will give a false positive and degrade the metal. c. A positive result is the rapid evolution of O 2 as evidenced by bubbling. d. A negative result is no bubbles or only a few scattered bubbles. e. Dispose of your slide in the biohazard glass disposal container. Dispose of any toothpicks in the Pipet Keeper. OXIDASE TEST Basically, this is a test to see if an organism is an aerobe. It is a check for the presence of the electron transport chain that is the final phase of aerobic respiration. -

These enzymes utilize a wide range of substrates from methylamine to polypeptides. Changes in CAO activity are correlated with a variety of human diseases, including diabetes mellitus, Alzheimer’s disease, and inflammatory disorders. CAOs contain a cofactor, 2,4,5-trihydroxyphenylalanine quinone (TPQ), that is required for catalytic activity and synthesized through the post-translational modification of a tyrosine residue within the CAO polypeptide. TPQ generation is a self-processing event only requiring the addition of oxygen and Cu(II) to the apoCAO. Thus, the CAO active site supports two very different reactions: TPQ synthesis, and the two electron oxidation of primary amines. Crystal structures are available from bacterial through to human sources, and have given insight into substrate preference, stereospecificity, and structural changes during biogenesis and catalysis. -

ABSTRACT OF THESIS CHARACTERIZATION OF POLYPHENOL OXIDASE AND ANTIOXIDANTS FROM PAWPAW (ASIMINA TRIBOLA) FRUIT Crude polyphenol oxidase (PPO) was extracted from pawpaw (Asimina triloba) fruit. The enzyme exhibited a maximum activity at pH 6.5–7.0 and 5–20 °C, and had -1 a maximum catalysis rate (Vmax) of 0.1363 s and a reaction constant (Km) of 0.3266 M. It was almost completely inactivated when incubated at 80 °C for 10 min. Two isoforms of PPO (MW 28.2 and 38.3 kDa) were identified by Sephadex gel filtration chromatography and polyacrylamide gel electrophoresis. Both the concentration and the total activity of the two isoforms differed (P < 0.05) between seven genotypes of pawpaw tested. Thermal stability (92 °C, 1–5 min) and colorimetry (L* a* b*) analyses showed significant variations between genotypes. -
3. Methodology
3. METHODOLOGY The present study focused on the responses evoked by B. monnieri under conditions of oxidative stress. The plan of work and the methodology adopted are presented in this chapter. The work was carried out in four phases. In Phase I, different parts of B. monnieri , namely leaves, stolon and roots, were assessed for their antioxidant potential and free radical scavenging activity. The biomolecular protective effects and the apoptosis- modulating effects of the leaf extract in cell free and in vitro systems were studied in Phase II. Phase III involved assessing the antioxidant status in precision-cut liver slices exposed to oxidative stress and confirming the results in experimental animals. In the final phase of the study, phytochemical analysis of the active component in B. monnieri leaves was carried out. The layout of the study is presented below. PHASE I In this phase, the antioxidant status in different parts of B. monnieri was assessed. The leaves, stolon and roots of the plantlets were collected from the plants grown in pots in the University campus. They were washed thoroughly in running tap water in order to remove any dirt or soil particles adhered and blotted gently between folds of tissue paper to remove any water droplets. Both enzymic and non-enzymic antioxidants were analysed in all the three parts of the plant. ENZYMIC ANTIOXIDANTS The enzymic antioxidants analyzed in the parts of B. monnieri w ere superoxide dismutase, catalase, peroxidase, glutathione S-transferase and polyphenol oxidase. The main aspects considered are the structure, dis- tribution, location and properties of polyphenol oxidase (PPO) as well as newly discovered inhibitors of the enzyme. Particular stress is given to the possible function of the enzyme. The cloning and characterization of a large number of PPOs is surveyed. Although the active site of the enzyme is conserved, the amino acid sequence shows very considerable variability among species. Most plants and fungi PPO have multiple forms of PPO. Expression of the genes coding for the enzyme is tissue specific and also developmentally controlled. Many inhibitors of PPO have been described, which belong to very diverse chemical structures; however, their usefulness for controlling PPO activity remains in doubt. The function of PPO still remains enigmatic. In plants the positive correlation between levels of PPO and the resistance to pathogens and herbivores is frequently observed, but convincing proof of a causal relationship, in most cases, still has not been published. Evidence for the induction of PPO in plants, particularly under conditions of stress and pathogen attack is consid- ered, including the role of jasmonate in the induction process. A clear role of PPO in a least two biosynthetic processes has been clearly demonstrated. -

Chemical and Technical Assessment 63rdJECFA Hexose Oxidase from Chondrus Crispus expressed in Hansenula Polymorpha CHEMICAL AND TECHNICAL ASSESSMENT (CTA) Prepared by Jim Smith and Zofia Olempska-Beer © FAO 2004 1 Summary Hexose oxidase (HOX) is an enzyme that catalyses the oxidation of C6-sugars to their corresponding lactones. A hexose oxidase enzyme produced from a nonpathogenic and nontoxigenic genetically engineered strain of the yeast Hansenula polymorpha has been developed for use in several food applications. It is encoded by the hexose oxidase gene from the red alga Chondrus crispus that is not known to be pathogenic or toxigenic. C. crispus has a long history of use in food in Asia and is a source of carrageenan used in food as a stabilizer. As this alga is not feasible as a production organism for HOX, Danisco has inserted the HOX gene into the yeast Hansenula polymorpha. The information about this enzyme is provided in a dossier submitted to JECFA by Danisco, Inc. (Danisco, 2003). Based on the hexose oxidase cDNA from C. crispus, a synthetic gene was constructed that is more suitable for expression in yeast. The synthetic gene encodes hexose oxidase with the same amino acid sequence as that of the native C. crispus enzyme. The synthetic gene was combined with regulatory sequences, promoter and terminator derived from H. polymorpha, and inserted into the well-known plasmid pBR322. The URA3 gene from Saccharomyces cerevisiae (Baker’s yeast) and the HARS1 sequence from H. polymorpha were also inserted into the plasmid. The URA3 gene serves as a selectable marker to identify cells containing the transformation vector. -

One of the main causes of aging is oxidative stress deriving from the imbalance between the production of reactive oxygen (ROS) and nitrogen (RNS) species and their scavenging through antioxidants. Xanthine oxidoreductase (XOR) activities produce uric acid, as well as reactive oxygen and nitrogen species, which all may be relevant to such equilibrium. This review analyzes XOR activity through in vitro experiments, animal studies and clinical reports, which highlight the pro-aging effects of XOR products. However, XOR activity contributes to a regular level of ROS and RNS, which appears essential for the proper functioning of many physiological pathways. This discourages the use of therapies with XOR inhibitors, unless symptomatic hyperuricemia is present. -

The kidney not only regulates fluid and electrolyte balance but also functions as an endocrine organ. For instance, it is the major source of circulating erythropoietin and renin. Despite currently available therapies, there is a marked increase in cardiovascular morbidity and mortality among patients suffering from end-stage renal disease. We hypothesized that the current understanding of the endocrine function of the kidney was incomplete and that the organ might secrete additional proteins with important biological roles. Here we report the identification of a novel flavin adenine dinucleotide–dependent amine oxidase (renalase) that is secreted into the blood by the kidney and metabolizes catecholamines in vitro (renalase metabolizes dopamine most efficiently, followed by epinephrine, and then norepinephrine). In humans, renalase gene expression is highest in the kidney but is also detectable in the heart, skeletal muscle, and the small intestine. The plasma concentration of renalase is markedly reduced in patients with end-stage renal disease, as compared with healthy subjects. -
