A. BRIEF RESUME OF INTENDED WORK

A.1. Need for study:

The man has always turned to the nature in quest for his well being.

Although the man, himself part of the nature, has learnt and applied various intricacies of nature to solve problems confronting him, in various fields. The field of medicine is no exception. Even before the dawn of civilization man used resources of nature for healing purpose. With the advancement of scientific discoveries, which is of only a recent past in the scale of human civilization, man began to use pure chemicals, either synthetic or isolated from nature, for the treatment purpose. The experience with these chemicals as drug is not too a pleasant one. These drugs, although well known for quick relief, suffers from the major side effects, adverse effects, sometimes more harmful than the diseases itself. As the world understands the limitations of pure chemicals as drugs, it has turned to the nature once again for solving health related problems without any adverse or side effects. In this context ancient Indian system of medicine like Ayurveda, holds the key in solving the problems faced by modern medicine.

The Ayurvedic system of medicine mainly uses herbs, herbal products, in the form of dry powders, extracts, kashayas, asavas, aristas etc. These formulations usually contain more than one type of herb or herbal products. Very often raw materials are being collected from different geographical sources and at different seasons. Hence the amount of active constituents present in the sample material varies and its effects are likely to reflect in the final product. It is an immediate and important task for Ayurvedic medicine to be standardised on lines with modern medicine system. In modern context the word standardizations refers to those an

1 activity which ensures every batch of drugs prepared contains the original materials and in the proportion intended.

Talisadi churna is an herbal ayurvedic medicine contains Talisha (Abies webbiana), Maricha (Piper nigrum), Shunthi (Zingiber officinale), Pippapi (Piper longum), Vanshalochana, Dalchini (Cinnamomum zeylanicum), Ela (Elettaria cardamomum), and sugar candy. Talisadi churna is one of the best medicaments in the treatment of loss of appetite, indigestion, respiratory discomfort, cough, and is best remedy in acute, chronic and allergic bronchitis. It is very useful in acute exacerbation of asthma, as it helps in expectoration by liquefying phlegm. In chronic asthma,

Talisadi churna reduces the frequency and severity of attacks.1

Number of studies has been reported on standardization of many churna like lavan bhaskar churna,2 amukkaram curanam,3 nyogrodhadi churna,4 avipattikar churna,5 hingwastalk churna.6 But there are no reports on standardization of Talisadi churna. Hence, the proposed work is intended to standardise Talisadi churna.

A.2: Review of literature:

 Standardization of lavan bhaskar churna was carried out as per the guidelines

of World Health Organization and parameters such as, extractive value, ash

value, phyto-constituents, were determined and subjected for spectroscopic

studies. This study on lavan bhaskar churna was reproducible precise and may

be considered as a method for its quality control.2

 Standardization of a siddha formulation, amukkara churnam by HPTLC was

investigated for the estimation of marker compounds withferine A and

piperine contents in a prepared standard and commercial formulation by using

2 HPTLC method of analysis. The two formulations were subjected to methanol,

ethyl acetate and chloroform extractions by using soxhlet apparatus. The

chromatogram was developed using chloroform: methanol (8.5:1.5) and

toluene: ethyl acetate (7:3) as mobile phases for the estimation of withferine A

and piperine respectively and absorbance was measured at 220nm. The

proposed HPTLC method was found to be rapid, simple and accurate for

quantitative estimation of withferine A and piperine in different formulation

extracts.3

 An Ayurvedic polyherbal formulation, nyagrodhadi churna was standardized

by specific parameters, both for the raw materials and finished products. The

obtained values of physical and chemical parameters can be adopted to lay

down new pharmacopoeial standards for the traditional preparation of

nyagrodhadi churna. It helps to maintain derived level at active constituent in

every batch of formulation manufactured. In this way it possibly facilitates the

desirable therapeutic efficacy. 4

 Standardization of avipattikar churna a polyherbal formulation has been

reported. In-house preparation and two marketed products have been

standardized on the basis of organoleptic, physical and physico- chemical

properties. The set parameters were found to be sufficient to evaluate the

churna and can be used as reference standards for the quality control or quality

assurance laboratory of a pharmaceutical house.5

 Hingwastak churna, a polyherbal formulation, standardised according to WHO

guidelines. In-house preparation and three marketed samples have been

standardized on the basis of organoleptic, physical and physico-chemical

properties. The set parameters were found to be sufficient to evaluate the

3 churna and can be used as reference standard for the quality control or quality

assurance laboratory of a pharmaceutical house.6

B. MATERIALS AND METHODS:

B.1: Source of the data:

The required data will be obtained from:

B.2: Methodology:

. Preparation of Talisadi churna: Churna will be prepared as per the procedure

given in Sharanghadhara samhita.

4 . Powder microscopy: It will be done for the examination of fibers, cells starch,

calcium oxalate, lignin etc.

. Physico-chemical evaluation: The parameters such as extractive values, ash

values, and pH will be studied.

. Physical characteristics of powder formulation: The parameters such as tap

density, bulk density, angle of repose, Hausner ratio, and Carr’s index will be

studied.

. Phyto-chemicals investigations: To confirm the various secondary metabolites

preliminary phyto-chemical investigation will be performed.

. Standardization based on TLC and HPTLC profiles: All the preparations will

be subjected for chromatographic fingerprint analysis and HPTLC.

. Compilation of data collected by in-house preparation and marketed products

will be carried out in order to establish the standards for Talisadi churna.
