MADAYANTIKA (LAWSONIA INERMIS Linn.) –AN EXPERIMENTAL EVALUATION” 6. BRIEF RESUME OF THE INTENDED WORK:

6.1 Need for study-

Ayurvedic and Modern Science of Medicine have recognized the importance of Yakrt (liver). Ayurveda consider it as one among the Kosthangas. It is stated to be the seat of ranjaka pitta and root of raktavahasrotas. It takes part in various metabolic activities and Bhutagnipaka. The liver is the largest gland in human body, weighing approximately 3pounds and occupying a large region mostly on the right side of the body, below diaphragm and behind ribs five to ten.

Liver disease is categorized both by the cause and the effect it has on the liver. Causes may include infection, injury, exposure to drugs or toxic compounds, and autoimmune process. Liver disease may affect the capacity of liver to metabolise drugs. An impairment of this organ and infection caused to it leads to the liver diseases like Alcoholic liver diseases, Hepatitis, Primary biliary cirrhosis, liver abscess, hemochromatosis and liver cancer1. It is estimated that liver diseases are among the top ten killer diseases in India, causing lakhs of death every year. To treat liver diseases of unknown aetiology, the usage of herbal medications with activities like hepatoprotective, anti-hepatic viruses and stimulation of hepatocyte regeneration has to be developed. There are more than 87 medicinal plants used in different combinations in the preparation of patented herbal formulations. It is also used as a single drug therapy by most of folklore practitioners for hepatic disorders in India e.g. Bhumyamalaki, Kalamegha, Bhrungaraja, Guduchi etc.

It is reported that the folklore traditional practitioners are using the plant Lawsonia inermis (Madayantika ) for the treatment of Jaundice. In view of this while exploring the folk remedies it is found that the traditional tribal practitioners of Waynadu area of Kerala are using the leaves and roots of the drug Lawsonia inermis (Madayantika) against jaundice. It is a shrub which goes up to a height of 4meters baring small white flowers. According to Prof P V Sharma2 properties of Madayantika are:- Rasa- Tikta,Kashaya: Guna- Leghu,Ruksha, ;Virya- Sita; Vipaka- Katu; Karma- Kapha Pithahara .and indicated in Kamala, Mutrakricchra, Apasmara,Kandu & Kustha

The objective of this study is to determine the effect and efficacy of Lawsonia inermis (Madayantika) in the liver and to reveal the hepatoprotective activity of the drug. 6.2 Review of Literature:

1. Literary source: a) The data of drug Lawsonia inermis (Madayantika ) will be available in various Ayurvedic books. The modern writers of Ayurveda especially , P.V. Sharma2, Khare C P3 , J L N Sastry4

Nadkarni5 , P K Varrier6 , etc. have also identified and described this plant in detail.

b ) Literature data of hepatoprotectivity and studies will be collected from all the available modern medicine literature from library of Shri. J.G.C.H Society’s Ayurvedic Medical College,

Ghataprabha , Belgaum

2. Drug Source: The drug Lawsonia inermis Linn (Madayantika ) will be available in herb garden of Shri. J.G.C.H Society’s Ayurvedic Medical College, Ghataprabha , Belgaum

2) Experimental evaluation of hepatoprotective activity of Patola (Trichosanthus dioica Roxb) & Kethaki (Pandanus odovatissRoxb) by Dr R Ilanchezhiyan in 2004 RGUHS, Bangalore

3) Pharmacognostical & Phytochemical study of Parijata w s r to hepato protective activity by Dr Sarawad sashidhar S in 2005 RGUHS, Bangalore

4).An experimental study on the hepatoprotective action of Udumbara (Ficus recemosa Linn) by Pushpa S in 1994, Kerala University, Trivandrum

5). Experimental Evaluation of Hepato protective Effect of Kakamaci (Solanum nigrum L) by Dr Sanjeev L Athani in 2003, RGUHS, Bangalore 6.3 Objective of the study-

1. To experiment and observe the hepatoprotective effect of the leaves of Lawsonia inermis Linn.

2. To experiment and observe the hepatoprotective effect of the roots of Lawsonia inermis Linn.

3. To compare the hepatoprotective activity of leaves and roots of Lawsonia inermis Linn

4. To develop a cost effective hepatoprotective single herb therapy for liver diseases.

7. Materials and Methods-

7.1 Source of data- Single blind study will be conducted at govt approved animal house on albino rats.

7.2 Method of collection of data-

The experimental study will be carried on four groups. As per the inclusion criteria 24 albino rats of either sex will be selected for the study a) Inclusion Criteria-

The Sprague dawley type of Albino rats7 of either sex (weighing between 150 and 200gram) bred in animal house will be selected for the study. b)Assessment Criteria- The estimation of enzymes like serum glutamic oxaloacetic transaminase (S.G.O.T.), serum glutamic pyruvic transaminase (S.G.P.T.), bilirubin and proteins are considered as the criteria for the assessment of the result, along with the histopathological evaluations. c) Investigations-

Liver Function Test: e) Study design (Interventions)-

Study will be conducted at govt approved animal houses on Albino rats. The experimental study will be carried on four groups.

Group -I will be the control group which will be treated with the distilled water.

Group II - will be treated with hepato toxic reagent Carbon tetra chloride (CCl4)

Group III – will be treated with root kwatha of drug Lawsonia inermis Linn

Group IV- will be treated with leaf swarasa of drug Lawsonia inermis Linn

In various liver disorders the serum enzyme levels like serum glutamic oxaloacetic transaminase (S.G.O.T.), serum glutamic pyruvic transaminase (S.G.P.T.) and the serum levels of bilirubin etc., are elevated and the serum protein levels are decreased. To produce such a condition, experimentally, carbon tetrachloride will be given in Albino rats as per the approved methods. The estimation of above said enzymes, bilirubin and proteins will be considered as the criteria for the assessment of the result, along with the histopathological evaluations.

7.3 Does the study require any investigations or interventions to be done on patients or other humans or animals? If so please describe briefly.

Yes .The study will be conducted at govt approved animal houses on albino rats .The Sprague dawley7 type of Albino rats of either sex weighing between 150-200gram in animal house will be selected for the study. 24 animals will be separated in 4 groups each with six animals for the study.

7.4 Has ethical clearance been obtained from in case of 7.3?

Yes, clearance has been obtained through the ethical committee formulated in the institution.
