Rederivation from Frozen Embryos

Quick Guide (see below for more detailed flow chart):

1. PI submits a project request form in PPMS

2. PI arranges acquisition of frozen embryos (week1)

3. Frozen embryos are thawed and implanted into recipient females (week 2)

4. PI genotypes toe biopsies from pups born (week 6-7)

5. Mice are weaned onto PI’s smart key and protocol number and recipient moms are sent for pathogen testing (week 9)

6. Clean pups are transferred to PI (week 10)

Detailed Guide:

Before Project Begins:

1. PI is responsible to order the project from PPMS 2. PI needs contact TMF to discuss project 3. PI is responsible for arranging acquisition of the frozen embryos through Kristy Weed in DAR if they are not already being stored by the Core.

Project Flow:

1. PI lab is responsible for arranging transfer of frozen embryos to TMF via DAR. Kristy Weed is the contact person for arranging acquisition.

PI lab is responsible for submitting thawing protocols from the original source of the frozen embryos to TMF

2. TMF will thaw embryos and transfer them into recipient females.

3. Toe biopsies will be taken from the pups for genotyping when the pups are 7 days old

4. PI lab is responsible for screening rederived pups from toe biopsies provided by TMF

5. PI lab is responsible to send TMF screening result and electronic image

6. When rederived pups are 4 weeks old, TMF will use PI’s IACUC protocol and smart key to wean the pups 7. TMF will send recipient moms for Pathogen testing through DAR, PI is responsible to pay testing fee.

8. When negative testing results are received, TMF will arrange to transfer the clean, rederived mice to PI lab

9. PI lab is responsible for checking that the mice have been received.

Service Rate Structure:

1. TMF will bill PI on the month of the project initiated:

 Rederivation from frozen embryo fee: $934

2. Extra fee will be billed to PI lab in the following cases:

 $173.67 for every two recipient moms for pathogen test.  Per diem fee after pups wean  $38 per hour plus material cost when TMF extract DNA from tail samples  Mouse transport costs if rederived mice are being shipped outside of Emory

Project Guarantee and Quality Control

1 TMF will guarantee the existing Core protocol and Core SOP are followed, and will follow any protocols provided from PI
