Captions for Supplementary Movies
Two movies show typical movements of wild-type plVirE2 (movie 1) and “animalized” mutant anVirE2 (movie 2) complexes with ssDNA in interphase Xenopus egg extracts containing networks of microtubules and F-actin. The movements are qualitatively different, with the plVirE2 complexes showing apparently Brownian motion (in fact a thermally-driven subdiffusion) and the anVirE2 complexes showing more extended directional excursions. Note that in the plVirE2 case the complexes drift vertically, out of focus, while in the anVirE2 movie they remain better confined to the plane of focus. The frame sizes of the movies are 53 x 45 and 47 x 41 microns for the anVirE2 and plVirE2 sequences, respectively. The frame rates have been sped up by a factor of ~5, from 6.25 to 30 frames/sec.
