Overton, [email protected] Abstract: Morphological studies and phylogenetic analyses of DNA sequences from the internal transcribed spacer (ITS) regions of the nuclear ribosomal gene repeat, a partial sequence of RNA polymerase II subunit (rpb2), and a partial sequence of the large exon of tef1 (LEtef1) were used to investigate the taxonomy and systematics of nine Hypocrea species with anamorphs assignable to Trichoderma sect. Hypocreanum. Hypocrea corticioides and H. sulphurea are reevaluated. Their Trichoderma anamorphs are described and the phylogenetic positions of these species are determined. Hypocrea sulphurea and H. subcitrina are distinct species based on studies of the type specimens. Hypocrea egmontensis is a facultative synonym of the older name H. subcitrina. Hypocrea with anamorphs assignable to Trichoderma sect. Hypocreanum formed a well-supported clade. Five species with anamorphs morphologically similar to sect. Hypocreanum, H. avellanea, H. parmastoi, H. megalocitrina, H. alcalifuscescens, and H. pezizoides, are not located in this clade. Protocrea farinosa belongs to Hypocrea s.s. This is also the first report of S. mycoparasitica as a biotrophic mycoparasite on Fusarium culmorum and F. equiseti in addition to its other reported hosts (F. avenaceum, F. graminearum, and F. oxysporum). In slide culture assays, S. mycoparasitica acted as a contact mycoparasite of F. culmorum, and F. equiseti producing hook-like attachment structures. Fluorescent and confocal laser scanning microscopy showed that S. mycoparasitica is an intracellular mycoparasite of F. equiseti but not of F. culmorum. All three mycoparasitic Sphaerodes species were observed to produce asexual (anamorphic) stages when challenged with Fusarium. Furthermore, a phylogenetic tree, based on (large subunit) LSU rDNA sequences, depicted closer relatedness to one another of these Fusarium-specific Sphaerodes taxa than to the non- mycoparasitic S. compressa, S. fimicola, and S. singaporensis. However, a critical examination of the original illustration reveals that the host is the golden carpenter ants carpenter ant, Camponotus sericeiventris (Camponotini, Formicinae). Because the holotype is no longer extant and epitype the original diagnosis lacks critical taxonomic information – specifically, on ascus and ascospore morphology – a new Ophiocordyceps type from Minas Gerais State of south-east Brazil is designated herein. A re-description of the fungus is provided and phylogeny a new phylogenetic tree of the O. unilateralis clade is presented. Stromata-producing endophytes were commonly encountered in populations of Agrostis capillaris, A. stolonifera, Dactylis glomerata, and Holcus lanatus. Asymp- tomatic endophytes were detected in Bromus ramosus, Festuca arundinacea, F ovina var. hispidula, F. pratensis, F. rubra, and Festulolium loliaceum. Here, we describe three newly discovered host-specific species, Ophiocordyceps camponoti-atricipis, O. camponoti- bispinosi and O. camponoti-indiani, on Camponotus ants from the central Amazonian region of Brazil, which can readily be separated using morphological traits, in particular the shape and behavior of the ascospores. DNA sequence data support inclusion of these species within the Ophiocordyceps unilateralis complex. Observations on Early Fungal Infections with Relevance for Replant Disease in Fine Roots of the Rose Rootstock Rosa Corymbifera

Popp1, A. Baumann2, S. Fricke1, M. Menssen3, T. Winkelmann 2* & E. Maiss1 Replant disease is a worldwide phenomenon afecting various woody plant genera and species, especially within the Rosaceae. Compared to decades of intensive studies regarding replant disease of apple (ARD), the replant disease of roses (RRD) has hardly been investigated. The etiology of RRD is also still unclear and a remedy desperately needed. In greenhouse pot trials with seedlings of the RRD‑sensitive rootstock Rosa corymbifera ‘Laxa’ cultured in replant disease afected soils from two diferent locations, early RRD symptom development was studied in fne roots. In microscopic analyses we found similarities to ARD symptoms with regards to structural damages, impairment in the root hair status, and necroses and blackening in the cortex tissue. Examinations of both whole mounts and thin sections of fne root segments revealed frequent conspicuous fungal infections in association with the cellular disorders. Particularly striking were fungal intracellular structures with pathogenic characteristics that are described for the frst time. Isolated fungi from these tissue areas were identifed by means of ITS primers, and many of them were members of the Nectriaceae. In a next step, 35 of these isolates were subjected to a multi‑locus sequence analysis and the results revealed that several genera and species were involved in the development of RRD within a single rose plant. Inoculations with selected single isolates (Rugonectria rugulosa and Ilyonectria robusta) in a Perlite assay confrmed their pathogenic relationship to early necrotic host plant reactions, and symptoms were similar to those exhibited in ARD. -

Endophytes have shown the potential for the biological control of diseases. The colonization ability of native endophyte strains of Beauveria bassiana and their antifungal effect against B. cinerea were evaluated in Solanaceae crops. Root drenching with B. bassiana was applied, and endophytic colonization capacity in roots, stems, and leaves was determined. The antagonistic activity was evaluated using in vitro dual culture and also plants by drenching the endophyte on the root and by pathogen inoculation in the leaves. Ten native strains were endophytes of tomato, and eight were endophytes of chili pepper. All strains showed significant in vitro antagonism against B. cinerea (30–36%). A high antifungal effect was observed, and strains RGM547 and RGM644 showed the lowest percentage of the surface affected by the pathogen. -
