Tau- tubulin kinase 1 and 2 (TTBK1 and TTBK2) belong to the casein kinase 1 superfamily of kinases. TTBK2 is ubiquitously expressed, while TTBK1 is neuron-specific and can directly phosphorylate tau at multiple Ser/Thr residues that are found on paired helical filament-tau or PHF-tau in Alzheimers disease (AD) brains. Consistently, transgenic mice over-expressiong human TTBK1 develop age-dependent pathology in brain, including intraneuronal accumulation of phosphorylated neurofilament (p-NF), tau phosphorylation, cortical and hippocampal astro/microgliosis, calpain I up-regulation, enhanced CDK5/p35 complex formation, down regulation of NMDA receptor, and memory impairment.
