Üreme dönemleri değerlendirildiğinde, A. mossulensis, C. macrostomus ve C. kais‟in Mayıs ayında, A. marmid‟in ise Mayıs-Haziran aylarında yumurta dökmeye baĢladığı tespit edilmiĢtir. Prof. Dr. Didem Gökçe Karakaya Dam Lake is an important aquatic ecosystem, used for irrigation, fishing and recreational activity by Malatya province and the surrounding residential areas. Four sampling points were selected to determine the impact of cage aquaculture in Karakaya Dam Lake on water quality and growth and reproductive characteristics of Alburnus mossulensis, Acanthobrama marmid, Cyprinion macrostomus ve Cyprinion kais. Vertical water and fish samples were collected from the selected locations at the same time between December 2008-2009 and physico-chemical parameters of water were analyzed on a monthly basis. According to water quality values, the trophic level of Karakaya Dam Lake was determined as oligotrophic and mesotrophic. Length-weight-sex distribution, age-sex composition, the absolute-proportional growth in lenght and weight, length-weight, age-lenght, age-weight relationship, condition factor, gonadosomatic index, egg diameter and fecundity of collected fish were determined. Differences in male and female individuals according to months and sampling points with the degree of importance were determined by ANOVA and Tukey test. In addition, by the Canonical Correspoinding Analysis (CCA) used, multiple relations between environmental variables that effect ecosystem structure of dam lake and growth-reproduction parameters have been analysed and depending on the sampling points, the effect and degree of environmental factors on fish composition have been shown. Evaluation of reproductive periods show, spawning periods of A. mossulensis, C. macrostomus and C. kais begin in May and A. marmid begins in May-June. In female subjects, distributions of species were not affected significantly by the environmental variables and ist. 1 and ist. 2 were similar. As a result, amounts of nitrogen and phosphorus in the sampling points 1 and 2 were higher than the sampling points 3 and 4 which were built after the first two and biological parameters of fish were affected accordingly. It five widely separated regions; the Mediterranean breeds in late winter and early spring, a window basin, parts of western North America, parts of of opportunity between flash floods and habitat western and southern Australia, southwestern desiccation. Being a multiple spawner allows the South Africa and parts of central Chile. Streams fish to compensate for the potential loss of part of in these regions feature seasonal disturbances of its reproductive output due to scouring flows of contrasting hydrology with high predictability of late floods. The ability of the Yarqon bleak to the timing of flooding and drying but low con- spawn on different substrate-types enables it to stancy. We would expect fish living in these take advantage of different stream conditions that streams to avoid scouring flow and breed after pertain in different years. The fish attains pre- cessation of the flood period. Received: May 2012 Accepted: December 2014 Abstract In this study, for the first time in Iran, the karyotype of bigmouth Lotak, Cyprinion macrostomum Heckel, 1843, was investigated through examining metaphase chromosomes of seven fish with mean weight 30±5g caught by electrofishing from Godarkhosh River in Ilam Province. To stimulate cell divisions, fish were injected intraperitoneally two times by phytohemagglutinin (PHA). The cell divisions were arrested in metaphase stage by intraperitoneal injection of colchicine. Well-separated cells were obtained from kidney and gill filament and chromosome spreads were prepared and stained with giemsa. Karyotype was obtained as 2n=50. The karyotype consisted of 5 metacentric, 12 submetacentric and 8 telocentric chromosome pairs. Centromeric index, arm ratio and Fundamental Number (FN) were determined as 0-50, 1-∞, and 84, respectively. Keywords: Bigmouth lotak, Cyprinion macrostomum, Godarkhosh River; Iran, Karyotype. • Karyological and Molecular Analysis of Three Endemic Loaches (Actinopterygii: Cobitoidea) from Kor River Basin, Iran
Molecular Biology Research Communications 2015;4(1):1-13 MBRC Original Article Open Access Karyological and molecular analysis of three endemic loaches (Actinopterygii: Cobitoidea) from Kor River basin, Iran Hamid Reza Esmaeili1,*, Zeinab Pirvar1, Mehragan Ebrahimi1, Matthias F. Geiger2 1) Department of Biology, College of Sciences, Shiraz University, Shiraz, Iran 2) Zoological Research Museum Alexander Koenig, Leibniz Institute for Animal Biodiversity, Adenauerallee, Germany ABSTRACT This study provides new data on chromosomal characteristics and DNA barcoding of three endemic loaches of Iran: spiny southern loach Cobitis linea (Heckel, 1847), Persian stream loach Oxynoemacheilus persa (Heckel, 1848) and Tongiorgi stream loach Oxynoemacheilus tongiorgii (Nalbant & Bianco, 1998). The chromosomes of these fishes were investigated by examining metaphase chromosome spreads obtained from epithelial gill and kidney cells. The diploid chromosome numbers of all three species were 2n=50. The karyotypes of C. linea consisted of 4M + 40SM + 6ST, NF=94; of O. persa by 20M + 22SM + 8ST, NF=90 and of O. tongiorgii by 18M + 24SM + 8ST, NF= 92. Sex chromosomes were cytologically indistinguishable in these loaches. Maximum likelihood-based estimation of the phylogenetic relationships based on the COI barcode region clearly separates the three Iranian loach species of the Kor River basin. All species distinguished by morphological characters were recovered as monophyletic clades by the COI barcodes. The obtained results could be used for population studies,Archive management and conservatio n programs.of SID Key words: Loaches; Phylogenetic relationships; COI barcode region; Idiogram; Iran INTRODUCTION The confirmed freshwater ichthyofauna of Iran are represented by 202 species in 104 genera, 28 families, 17 orders and 3 classes found in 19 different basins [1].
