PARAQUAM Final Activity Report PARAQUAM Final Activity Report GENERAL PROGRESS OF THE PROJECT Due to continuing problems with the availability of Mediterranean amberjack, Seriola dumerilii for experimental work, the focus of the study on sanguinicolid parasites of the genus Paradeontacylix in amberjack was moved to molecular aspects. Various genes of two new species of Paradeontacylix from the Mediterranean were sequenced and analysed together with further samples of other Paradeontacylix species from S. dumerili obtained from Japan. This part of the project has now been published and attracted collaboration with the University of Adelaide, providing samples of Paradeontacylix spp. from Seriola lalandi and Seriola hippos from Australia and New Zealand, which will be used to complete the picture of the worldwide distribution, phylogeny and evolution of Paradeontacylix parasites and their Seriola spp. hosts. The study of the monogenean parasite Zeuxapta seriolae from S. dumerili was completed. For continuation of this line of research, experimental infections were conducted with other monogeneans occurring on gilthead seabream, Sparus aurata and sharpsnout seabream, Diplodus puntazzo. Egg development, larval behaviour and survival rates, as well as infection strategies were investigated in S. aurata, whereas in D. puntazzo the studies were focussed on the distribution of the parasite in the host gills and the related pathology. The planned immunological study of fish infected with monogeneans had to be postponed due to the unexpected early leave of a SR. However, this part of the study has been picked up again recently with the help of an immunologist, who joined the department in October 2007. A new research line focussing on myxozoan parasites of fish has been developed over the last year and a half and results are now being published. One of the studies involves a candidate species for Mediterranean aquaculture, the stripped seabream, Lithognathus mormyrus. Another new line of investigation targets the development of a modern taxonomic framework that will contribute to understanding parasite biodiversity patterns in mullets, which are cultured in the Delta of the Ebro. A similar study is being conducted on metazoan parasites of the annular sea bream, Diplodus annularis (L.), an aquaculture candidate. Qualitative indicators of progress and success Research results were presented orally and as posters at two international and one national conference. Several articles have been published in peer-reviewed journals and others are still being completed. Due to the interesting results from the sanguinicolid studies, the department was contacted by the University of Adelaide, Australia, for collaboration on this topic. Research in the current Marie Curie project has been amplified regarding parasite and fish species new to the Mediterranean aquaculture. The ER on the project has finished her work satisfactorily and is still publishing results. An SR was recruited but left after setup of a basic immunology lab (6months). Another SR was recruited and is focussing on taxonomical studies. PROJECT ACHIEVEMENTS Scientific highlights - Population studies on Paradeontacylix spp. showed that geographic isolation is mirrored in genetic distances leading to speciation of the two parasite species in the Mediterranean (published). One, morphologically similar species from Australia is currently under study to complete the picture of worldwide speciation according to tectonic events. - Monogenean research on aquaculture candidate species of sparids has expanded and infection in a controlled artificial system was established and is being maintained for the investigation of life cycle, biological and pathological aspects of these infections. - A new line of research, i.e. myxozoan parasites was successfully established and resulted in first publications. - In-depth taxonomical studies showed surprising differences between the results of morphological and molecular techniques, changing the known nomenclature of various digenean parasite groups. Teaching and training activities The researchers of the current project were involved in the setup of an inter-university “Master in Aquaculture” program involving the University of Valencia, the Polytechnic University of Valencia and the Institute of Aquaculture de Torre la Sal (CSIC) as participating scientific establishments and embracing the Erasmus-Socrates Thematic Network “Aquaculture, Fisheries and Aquatic Resource Management” (225991-CP-1- 2005-1-BE-ERASMUS-TN) of the European Union. Three PhD students were successfully trained in molecular biology and one PhD student is currently learning immunological techniques. Two master theses with topics embedded in the current lines of investigation and under supervision of the investigators of the present project were conducted and successfully completed in Sept. 2007. In the present study, molecular identification of this species from the Kievka River, Primorye and from Vietnam was performed by analysis of 28S rDNA sequences. Analysis of ITS1 rDNA sequences variation revealed two highly differentiated main groups, representing trematode specimens from the two regions. Genetic variation within each region was relatively low. Mitochondrial COI gene sequence data analysis revealed fixed nucle- otide and amino acid substitutions, and supported the existence of two genetically different groups associated with geographical origin. Analysis of the COI gene fragments showed extremely high variation within Russian and Vietnamese P. mugili samples. Our results for P. mugili most probably represent a case of initial step of allopotric speciation for this trematode, caused by living strategy of its definitive host at evolutionary scale. Монтичелли (Monticelli, 1892) и вошло в состав сем. Capsalidae Baird, 1853 с типовым и единственным родом Encotyllabe Diesing 1850, который позднее Прайс (Price, 1939) поместил в подсем. Benedeniinae Johnston, 1931. Спростон (Sproston, 1946) восстановила подсем. Enco- tyllabinae, что было поддержано Быховским (1957) и Ямагути (Yamaguti, 1963). Впоследствии для подсемейства были описаны еще два рода — Neoencotyllabe Gupta et Krishna, 1980 и Alio encotyllabe Khalil et Abdul-Salam, 1988. Род Neoencotyllabe с типовым и единственным видом N. muelleri был обоснован Гуптой и Кришной (Gupta, Krishna, 1980) на основании изучения только одного экземпляра моногенеи, собранного с жабр морской рыбы Drepane punctata из Индии. Основными признаками, отличающими его от типового рода Encotyllabe, авторы посчитали наличие одного семенника (что для капсалид нехарактерно) и замкнутого кишечника (признак важный на уровне подсемейства и не свойственный энкотилла- бинам). При этом схематичный рисунок сопровождается очень кратким описанием. Мы разделяем сомнения Халила и Абдул-Саляма (Khalil, Abdul-Salam, 1988) по поводу достоверности рода Neoencotyllabe. Попытки этих авторов получить типовой материал для повторного изучения и подтверждения валидности этого рода не увенчались успехом. В связи с этим мы поддерживаем предложение Халила и Абдул-Саляма считать род Neoencotyllabe genus iquirendum. Таким образом, мы предлагаем числить в подсем. Encotyllabinae два рода (Enco- tyllabe и Alloencotyllabe) и 19 видов. По данным Спростон (Sproston, 1946), Ямагути (Yamaguti, 1963) и Лалера (Lawler, 1981) число родов и видов в подсемействе было соответственно: 1/8, 1/12, 1/15. Большинство видов энкотиллабин было обнаружено в ротовой полости и глоточ- ном отделе их хозяев — рыб, в то время как основными местами обитания капсалид являются жабры, жаберная полость и поверхность тела.

ABSTRACT: Of the 107 species of trematodes found in West African (Mauritania to Gabon) marine fishes, 100 are allocated to 64 genera in 24 families while seven are immature didymozoids. Many of these genera are located in most of the world's seas with the exception of the polar seas; only five are en- demic to West Africa. The data for the 41 species known from West Africa and elsewhere, and those morphologically closest to the 55 endemic species, indicate that they are very widely distributed, particularly in the Western and North Atlantic, and Mediterranean. Historical and present- day events concerning physical and biological environmental factors and their effects on actual and po- tential hosts as well as on life cycle stages of the trematodes have resulted in the geographical distribution reported. The distribution of marine fishes has been emphasized to explain in part the trematode distribu- tion. The current knowledge of each of the digenean species is provided from the existing specimens and literature, including a morphological descrip- tion with figures, lists of its synonyms, hosts, localities and existing specimens, and a summary of its life cycle where known. The Aporocotylidae Odhner, 1912 (Sanguinicola ugui, Sanguinicola hasegawai sp. nov. and Sanguinicola sp.), Bivesiculidae Yamaguti, 1934 (Bivesicula sp.) and Hap- loporidae Nicoll, 1914 (Carassotrema koreanum) are reviewed. Sanguinicola hasegawai sp. nov. is described and figured on the basis of specimens found in the blood vessels and heart of Barbat- ula toni (Dybowski, 1869) (Nemacheilidae) from Hokkaido. This taxa includes 5 monogeneans ( Lamellodiscus fla - gellatus , L. ignoratus , L. mormyri , L. verberis and Pagellicotyle mormyri ), 13 digeneans ( Le - pidauchen stenostoma , Derogenes latus , Magnibursatus bartolii , Proctoeces maculatus , Holorchis pycnoporus , Lepocreadium album , L. pegorchis , Macvicaria maamouriae , M. mail - lardi , M. mormyri , Pycnadenoides senegalensis , Diphterostomum brusinae and Zoogonus rubellus ), 3 nematodes ( Hysterothylacium rhacodes , Dichelyne tripapillatus and Ascarophis sp.), 1 cestode ( Scolex pleuronectis ) and 2 Acanthocephala ( Acanthocephaloides propinquus and A. incrassatus ). All the species were recorded for the first time in the western Algerian coast in this host. Furthermore, Lepidauchen stenostoma and Hysterothylacium rhacodes are reported for the first time in the western Mediterranean. Magnibursatus bartolii , Zoogonus rubellus , Dichelyne tripapillatus , Ascarophis sp. and Scolex pleuronectis provide a new host record in Lithognathus mormyrus from the Mediterranean Sea. Epidemiological indexes (pre - valence, abundance and mean intensity) were calculated for each helminth species identified in this fish. The Algerian west coast shows the highest value in the helminth species richness among all the Mediterranean coasts. KEY WORDS Helminthfauna; Lithognathus mormyrus ; Mediterranean Sea; Sparidae.
