Sekian, wassalam. iv ABSTRAK Komponen bioaktif di dalam teh telah banyak memberi kesan positif kepada unsur biologi termasuklah memberi kesan antimutagenik, antimikrobial serta antiproliferatif. Namun kajian terhadap kesan antikanser dan antiproliferatif bagi teh hitam sisa teh hitam dan kompos belum dijalankan secara meluas. Dalam kajian ini, dua kaedah pengekstrakan digunakan iaitu kadeah air panas dan kaedah methanolik. Hasil kajian mendapati bahawa kaedah pengekstrakan air panas memberikan hasil ekstrak yang lebih tinggi berbanding kaedah methanolik dengan sampel kompos mencatatkan berat hasil ekstrak sebanyak 17.58g, diikuti sisa teh sebanyak 14.31g dan sampel teh hitam sebanyak 12.94g. Bagi kaedah methanolik, sampel kompos mencatatkan berat hasil ekstrak sebanyak 6.33g, teh hitam dengan hasil seberat 6.02g dan sisa teh dengan hasilan ekstrak sebanyak 4.43g. Nilai kepekatan rencatan, ICso bagi kedua-dua jenis sel kanser dikira dan didapati bahawa sampel teh hitam dengan kaedah pengekstrakan air panas memberikan nilai ICso terendah (ICso MDA­ MB231=27.89±4.97, dan ICso Hep G2=147.05±37.21mg/mL). v ABSTRACT ANTIPROLIFERA TIVE PROPERTIES OF BLACK TEA, TEA WASTE AND COMPOST EXTRACTS OF SABAH TEA ON TWO TYPES OF CANCER CELL LINES The bioactive components in tea have been proved to give some beneficial effects on the biological entities including the antimutagenic, antimicrobial, and antiproliferative properties. But there are limited studies involving the usages of tea waste and compost as an antiproliferative and anticancer. In this study, two type of extraction methods are used, namely hot water extraction method and methanolic extraction method. The results give that the hot water extract give the highest extraction yield as the compost gives yield of 17.58g of extract, followed by the tea waste that gives 14.31g and lastly the black tea that gives the yield of 12.94g of the extract. For the methanolic extraction, compost gives the yield weighed 6.33g, followed by black tea that gives yield of 6.02g and tea waste gives the extraction yield of 4.43g. Applications of Biotechnology in Food By: Prof. Jagadish Pai Introduction Since ancient times, microorganisms have been used for a variety of applications in food. Fermented beverages, cheese, bread etc. were produced using microbes although people were not aware of the role of microbes until Louis Pasteur showed about 200 years ago the cause of fermentation and spoilage and laid the foundation of microbiology. The word biotechnology was coined towards the end of World War I in reference to intensive agricultural methods. UN Convention defined it as any technological application that uses biological systems, living organisms or derivatives thereof to make or modify products or processes for specific use. Although biotechnology has been used in pharmaceutical, medicine, non-food agriculture, cosmetics, textile, detergent, chemicals, fuel and environmental uses, it is commonly associated with agricultural food production and processing. Modern biotechnology is associated only to products or processes involving genetic engineering. Global market for biotechnology products is already well over US $200 billion and is expected to reach well over US $300 billion by 2015. Currently most applications are in health care industry while agriculture and food applications are growing rapidly. Biotechnology seeds market reached over US $13 billion. Farmers throughout the world are cultivating improved seeds resistant to herbicides and insects.

RGS proteins stabilize the transition state for GTP hydrolysis on Gα and thus induce a conformational change in the Gα subunit that accelerates GTP hydrolysis, thereby effectively turning off signaling cascades mediated by GPCRs. This GTPase accelerating protein (GAP) activity is the canonical mechanism of action for RGS proteins, although many also possess additional functions and domains.

Sarhan Department of Pharmacology and Physiology, College of Veterinary Medicine, University of Wasit, Wasit, Iraq Email: [email protected] (Received June 10, 2018; Accepted November 27, 2018) Abstract This current work aimed to study the pharmacokinetics of theaflavin in healthy and hepatotoxic rabbits for comparison. Aspartate aminotransferase (AST), alkaline phosphatase (ALP) and alanine aminotransferase (ALT) were significantly raised (P<0.05) after administration of 0.2 mg/kg body weight (BW) Carbone tetrachloride (CCL4) subcutaneously. Pharmacokinetic parameters calculated following administration of theaflavin intravenously and orally at 30 mg/kg and 500 mg/kg respectively to both healthy animals and those with damaged liver. Theaflavin concentration in blood measured by HLPC at various time intervals. Pharmacokinetic results showed that theaflavin concentration when given orally reached its maximum concentration after 5 hours in healthy rabbits. While in hepatotoxic group, theaflavin concentration achieved the highest level in blood after three hours. Theaflavin bioavailability in hepatotoxic animals was significantly high and almost double its bioavailability in healthy animals. Results revealed that the area under curve (AUC) value in rabbits with damaged liver was significantly greater than in healthy group (P<0.05). t ½ of theaflavin after intravenous administration was 6.3±0.82 hour in damaged liver group which is significantly higher than that in healthy group (P<0.05). Theaflavin mean concentration in hepatotoxic group required more than 3 hours to decline to 352±19.4 ng/ml when compared to its concentration in healthy group which is required only 45 minutes to decrease to 310± 9.5 ng/ml.

O-methylation is a major Phase II metabolic pathway of green tea catechins (GTCs) via the enzyme catechol-O- methyltransferase (COMT). A single nucleotide polymorphism in the gene coding for COMT leads to individuals with a high-, intermediate-, or low-activity COMT enzyme. An epidemiological case-control study suggests that green tea consumption is associated with reduced risk of breast cancer in women with an intermediate- or low-activity COMT genotype. A cross-sectional analysis discovered that men homozygous for the low- activity COMT genotype showed a reduction in total tea polyphenols in spot urine samples compared to the intermediate- and high-activity genotypes. Several human intervention trials have assessed green tea intake, metabolism, and COMT genotype with conflicting results. The aim of the present study was to determine if the COMT polymorphism would modify the excretion and plasma levels of GTCs in 180 postmenopausal women at high risk for breast cancer consuming a green tea extract supplement containing 1222 mg total catechins daily for 12 months.

An addition of (-) epigallocatechin gallate (EGCG) significantly inactivated SARS-CoV-2, while theasinensin A (TSA) and galloylated theaflavins including theaflavin 3, 3’-di-gallate (TFDG) had more remarkable anti-viral activities.

Theaflavin-3,3'-digallate (TF3), a theaflavin derivative in black tea, has 2 galloyl groups. Matrix metalloproteinases (MMPs) are associated with extracellular matrix degradation, cellular migration, and angiogenesis, and (-) -epigallocatechin-3-gallate (EGCG) is an inhibitor of MMP activity and secre- tion; thus one of its major actions is the inhibition of angiogenesis. However, there are few studies of angiogenesis in theaflavin derivatives. We investigated the effects of TF3 on angiogenesis in vitro. Angiogenesis was assayed using cocultured human umbilical vein endothelial cells with fibroblasts. Cells were cultivated in various concentrations of TF3 and EGCG in the presence or absence of vascular endothelial growth factor-A. After 11 days, MMP-2 and MMP-9 activities and the pro-MMP-2 protein in the medium were measured by gelatin zymography and immunoassay, respectively. Tube formation was markedly inhibited by 100 umol/L TF3 or EGCG. Even at 10 iumolIL, TF3 or EGCG inhibited tube formation. The MMP-2 and MMP-9 activities were inhibited and pro-MMP-2 protein concentrations were reduced by TF3 or EGCG in a concentration-dependent manner, regardless of the presence of vascular endothelial growth factor. The effect of TF3 was similar to that of EGCG, indicating that the tube formation of endothelial cells was suppressed via decreased both MMP-2 and MMP-9 activities in vitro.
